Interstitiële vloeistof stroom wordt verhoogd in solide tumoren en kan moduleren tumorcel invasie. Hier beschrijven een techniek interstitiële fluïdumstroming toepassing cellen ingebed in een matrix en meet de gevolgen celinvasie. Deze techniek kan gemakkelijk worden aangepast aan andere te bestuderen.
De groei en progressie van solide tumoren meest afhangen van de eerste transformatie van de kankercellen en hun reactie op stroma-geassocieerde signalering in de tumor micro 1. Eerder heeft onderzoek gedaan naar de tumor micro-omgeving zich vooral gericht op de tumor-stroma interacties 1-2. Echter, de tumor micro-omgeving omvat ook een aantal biofysische krachten, waarvan de gevolgen nog steeds slecht begrepen. Deze krachten zijn biomechanische gevolgen van de groei van tumoren die leiden tot veranderingen in genexpressie, celdeling, differentiatie en invasie 3. Matrix dichtheid 4, stijfheid 5-6, en de structuur 6-7, interstitiële vloeistofdruk 8, en interstitiële vloeistofstroming 8 zijn allemaal veranderen tijdens de voortgang van kanker.
Interstitiële fluïdumstroom in het bijzonder hoger in tumoren ten opzichte van normale weefsels 8-10. De geschatte interstitiële vloeistof flow snelheden werden gemeten en in het gebied van 0,1-3 um en -1, afhankelijk tumorgrootte en differentiatie 9, 11. Dit is te wijten aan hoge interstitiële druk door tumor geïnduceerde angiogenèse-en verhoogde vasculaire permeabiliteit 12. Interstitiale fluïdum debiet is aangetoond invasie van kankercellen 13-14 vasculaire fibroblasten en gladde spiercellen 15 verhogen. Dit invasie kan het gevolg autologe chemotactische gradiënten die rond cellen 3-D 16 of meer matrix metalloproteinase (MMP) expressie 15 chemokine secretie en cell adhesion molecule expressie 17. Echter, het mechanisme waardoor cellen voelen stroming niet goed begrepen. Naast het veranderen van het gedrag van tumorcellen, interstitiële vloeistofstroming moduleert de werking van andere cellen in de tumor micro-omgeving. Het is gekoppeld met (een) rijden differentiatie van fibroblasten in tumor-bevorderende myofibroblasts 18 (b) transporteren van antigenen en andere oplosbare factoren lymfeknopen 19 en (c) moduleren lymfatische endotheelcellen morfogenese 20.
De techniek die hier stelt interstitiële vloeistofstroom op cellen in vitro en kwantificeert het effect ervan op invasie (figuur 1). Deze methode is gepubliceerd in meerdere studies het effect van de vloeistofstroom meten stromale en kankercel invasie 13-15, 17.
