This 3D microfluidic printing technology prints arrays of cells onto submerged surfaces. We describe how arrays of cells are delivered microfluidically in 3D flow cells onto submerged surfaces. By printing onto submerged surfaces, cell microarrays were produced that allow for drug screening and cytotoxicity assessment in a multitude of areas.
The printing of cells for microarray applications possesses significant challenges including the problem of maintaining physiologically relevant cell phenotype after printing, poor organization and distribution of desired cells, and the inability to deliver drugs and/or nutrients to targeted areas in the array. Our 3D microfluidic printing technology is uniquely capable of sealing and printing arrays of cells onto submerged surfaces in an automated and multiplexed manner. The design of the microfluidic cell array (MFCA) 3D fluidics enables the printhead tip to be lowered into a liquid-filled well or dish and compressed against a surface to form a seal. The soft silicone tip of the printhead behaves like a gasket and is able to form a reversible seal by applying pressure or backing away. Other cells printing technologies such as pin or ink-jet printers are unable to print in submerged applications. Submerged surface printing is essential to maintain phenotypes of cells and to monitor these cells on a surface without disturbing the material surface characteristics. By printing onto submerged surfaces, cell microarrays are produced that allow for drug screening and cytotoxicity assessment in a multitude of areas including cancer, diabetes, inflammation, infections, and cardiovascular disease.
Recente ontwikkelingen in de farmaceutische industrie hebben geleid tot een toegenomen belangstelling voor het gebruik van cellulaire microarrays in de drug discovery proces voor drug discovery en cytotoxicological analyse 1,2,3. De ontwikkeling van in vitro high-throughput testen en screening methoden met behulp van microarrays cel zou de snelle en kosteneffectieve ontwikkeling van kandidaat-geneesmiddelen, alsook van tevoren het fundamenteel begrip van de cel 1,4 vergemakkelijken. De traditionele benadering van screening met cellen maakt gebruik van conventionele goed plaat platforms; maar deze benadering is beperkt vanwege de hoge kosten, beperkte verwerkingscapaciteit en beperkt vermogen om kwantitatieve informatie over celfunctie 1,5. Vanwege deze beperkingen, is het onderzoek in cellulaire microarray technologie ontluikende voor moleculair biologische karakterisering, tissue engineering, en drug screening 1,6. De voordelen van cellulaire microarrays omvatten kleinere steekproef gebruik, minimale effecten vancellulaire fenotype heterogeniteit maskeren informatie, en vooral de mogelijkheid om te testen automatiseren voor meer high-throughput toepassingen 1,7,8.
De farmaceutische industrie maakt gebruik momenteel high-throughput-cel-gebaseerde Screeningtesten met 2D cel monolaagculturen voor drug discovery in microtiterputje platen 9. Multiplexing cellen in putjes van microtiterplaten biedt het potentieel voor hogere doorvoer met unieke experimenten opties. Verder zijn de huidige technologieën voor cellulaire microarrays laat de cellen drogen die drastisch kunnen wijzigen fenotype van de cellen van in vivo 10,11. Om deze problemen te overwinnen, werd de MFCA ontworpen en wordt getoond in Figuur 1. Het ontwerp van de MFCA 3D fluïdica kan de printkop punt in figuur 1 worden neergelaten in een bad en samengeperst tegen een oppervlak om een afdichting te vormen. De zachte siliconen tip van de printkop zich gedraagt als eenpakking en vormt een omkeerbare afdichting. De MFCA technologie is uniek geschikt om met ondergedompelde oppervlakken, die nodig is voor zowel celculturen en weefselplak systemen en moeilijk of onmogelijk met de meeste andere benaderingen. Pinnen of inkt-jet printen zal niet werken, en 2D microfluïdische apparaten zijn niet geschikt voor afzetting of interfacing met grote arrays van discrete plekken. Verder, door miniaturisering en lokaliseren van het experiment – de cellulaire microarray – de MFCA overwint de belangrijkste problemen geassocieerd met high-throughput cel-gebaseerde screeningtesten.
De CFM maakt gebruik van 3D-kanaal netwerken te fietsen klein volume vloeistof monsters na microscopisch spot locaties op een oppervlak 12,13. Door druk met stroom worden biomoleculen, cellen en andere reagentia gehandhaafd in een vloeibare omgeving gedurende het printproces, waardoor de drukken van gevoelige biomoleculen en cellen zonder blootstelling aan lucht, waarbij de huidige cel druktechnieken belemmert. Het is ook mogelijk om afdrukken uit ruw materiaal zoals hybridoma supernatanten of mits er een opname mechanisme op de array oppervlak. Het doel van dit manuscript is in detail de ondergedompelde drukken van twee celtypen op een oppervlak.
De 3D microfluïdische printtechnologie hier beschreven is uniek in staat microfluidically drukken arrays van cellen in een vloeistof goed gevuld, dat wil zeggen een verzonken oppervlak.
