Introduction
De opname en verdeling van nutriënten en toxische stoffen sterk beïnvloeden de groei van planten. Dienovereenkomstig, het onderzoek van de onderliggende transport processen vormt een belangrijk onderzoeksgebied in plantaardige biologie en landbouwwetenschappen 1,2, in het bijzonder in de context van de voedings-optimalisatie en milieu-invloeden (bijvoorbeeld zout stress, toxiciteit ammonium). De belangrijkste van werkwijzen voor het meten van stromen in planten is het gebruik van radio-isotopische tracers, ontwikkeld significant in 1950 (zie voorbeeld 3) en blijft nu algemeen gebruikt. Andere methoden, zoals meten tekort aan nutriënten van de wortel medium en / of accumulatie in weefsels, gebruik van ion-selectieve trillende micro-elektroden zoals MIFE (micro-elektrode ion flux schatting) en siet (scannen ion-selectieve electrode techniek), en het gebruik van ion-selectieve fluorescente kleurstoffen, ook algemeen worden toegepast, maar zijn beperkt in hun vermogen om netto griep detecterenxes (dwz het verschil tussen instroom en uitstroom). Het gebruik van radio-isotopen, anderzijds, kan de onderzoeker het unieke vermogen om te isoleren en te kwantificeren unidirectionele stromen, die kan worden gebruikt om de kinetische parameters te lossen (bijvoorbeeld K M en V max) en inzicht in de capaciteit energetica mechanismen en regelgeving, van transportsystemen. Hier bieden we gedetailleerde stappen voor het radio-isotoop meting van unidirectionele en net stromen van minerale nutriënten en toxische stoffen in intacte planten. Nadruk worden aangegaan flux meting van kalium (K +), een plant macronutriënt 5 en ammonia / ammonium (NH3 / NH4 +), andere macronutriënten die echter toxisch wanneer aanwezig bij hoge concentraties (bijvoorbeeld 1- 10 mM) 2. We zullen radioisotopen 42 K + (t 1/2 = 12,36 uur) en 13 NH 3/13 NH 4 + (t 1/2 = 9,98 min) respectievelijk in intacte zaailingen van het modelsysteem gerst (Hordeum vulgare L gebruiken .), in de beschrijving van twee belangrijke protocollen: directe instroom (DI) en compartimentale analyse door tracer efflux (CATE). We moeten er rekening mee vanaf het begin dat dit artikel gewoon de stappen die nodig zijn om elk protocol voeren beschrijft.
