Vi beskriver prosedyrer for merking og genotyping nyfødte mus og generere primære nevronkulturer fra dem. Den genotyping er rask, effektiv og pålitelig, og gir mulighet for automatisert nukleinsyre-syre utvinning. Dette er spesielt nyttig for neonatally dødelige mus og deres kulturer som krever forutgående gjennomføring av genotyping.
Høy oppløsning analyse av morfologi og funksjon av nerveceller fra pattedyr krever ofte genotyping av individuelle dyr, fulgt av analyse av primære kulturer av neuroner. Vi beskriver et sett av prosedyrer for: merking nyfødte mus for å bli genotypet, rask genotyping, og etablering av lav tetthet kulturer av hjernens nerveceller fra disse musene. Individuelle mus er merket med tatovering, som gir mulighet for langsiktig identifikasjon varig inn i voksenlivet. Genotyping av den beskrevne protokoll er rask og effektiv og gir mulighet for automatisert uttrekking av nukleinsyre med god pålitelighet. Dette er nyttig under omstendigheter der tilstrekkelig tid for konvensjonell genotyping ikke er tilgjengelig, for eksempel, i mus som lider av neonatal dødelighet. Primære nevronale kulturer blir generert ved lav tetthet, noe som gjør det mulig for bildebehandling eksperimenter med høy romlig oppløsning. Derfor er denne protokollen vil ha brede programmer, ikke bare i nevrovitenskap, men også i andre felt av biologiske og medisinske fag.
Gnagermodeller av genetiske sykdommer har vist seg nyttig i å etablere de fysiologiske funksjonene til normale proteiner og nukleinsyrer, så vel som de patofysiologiske konsekvenser av defekter i disse. Eksempler inkluderer mus som mangler for proteiner som er involvert i viktige cellefunksjoner i tillegg til musemodeller av lidelser så som Alzheimers sykdom. Imidlertid kan visse genetiske manipulasjoner føre til neonatal dødelighet kort tid eller et par dager etter fødselen. I disse tilfellene, primære cellekulturer er et viktig verktøy fordi levende celler kan fås fra de embryonale eller neonatale unger før døden, de kan opprettholdes i minst et par uker in vitro, og i løpet av denne tiden tidlig neuronal utvikling kan følges av biokjemiske, funksjonelle og morfologiske eksperimenter. For de primære kulturer, kan det være gunstig å plate nervecellene ved lav tetthet; Dette gjør det mulig å visualisere den enkelte somata, dendritter, aksonal aksler og nerveklemlene på høy romlig oppløsning. Imidlertid overlevelse og differensiering av nerveceller med lav tetthet krever typisk at de er belagt på en glial mateslag, ko-dyrket med gliaceller i fravær av fysisk kontakt med dem, og dyrket i medium kondisjonert ved en gliaceller.
Etableringen av lav tetthet nevronale kulturer på glial næringslag kan være avhengig av en rask og pålitelig genotyping forhånd – i løpet av få timer, i motsetning til noen få dager. Hastighet er spesielt viktig når det nevronale genotype må matches til det av en glial mater lag forberedt på forhånd. Som en mer praktisk eksempel, kan det være nødvendig å bestemme hvilke unger som genotype til bruk ved generering av kulturer, for å optimalisere effektiviteten av et eksperiment.
Her demonstrerer vi arbeidsprotokollen som har vært brukt for rask, forenklet og pålitelig mus genotyping i tidligere publikasjoner 2-6. Musehaler oget kommersielt tilgjengelig kit benyttes. Denne protokollen innbefatter ett-trinns ekstrahering av nukleinsyrer fra vevet, og krever heller ikke en nukleinsyre-syre rensetrinn eller anvendelse av en termineringsbuffer ("stopp-løsning"). Påliteligheten av denne genotyping fremgangsmåte er illustrert ved å presentere resultatene av en serie tester ved forskjeller blir innført med hensyn til utgangsmengden av prøvene, i en alder av dyrene og lengden av PCR-amplikoner. Dette settet tilbyr fordelene ved automatisert utvinning og pålitelighet.
For å få til å være omfattende, er bruken av tatovering for langsiktig identifikasjon av genotypede mus også demonstrert. Vi bruker en genetisk musemodell for den arvet nevrologisk lidelse DYT1 dystoni – en autosomal-dominant bevegelse lidelse forårsaket av en mutasjon i genet TOR1A (c.904_906delGAG / c.907_909delGAG; p.Glu302del / p.Glu303del) 9. Den kodede protein, Torsina, tilhører de "ATPaser forbundet med ulike cellulære aktiviteter" (AAA +) familie av proteiner, hvis medlemmer generelt utføre anstand-lignende funksjoner, bistå i: protein utfolder seg, protein-kompleks demontering, membran menneskehandel, og vesikkelfusjon 10-13. Mutasjonen resulterer i en i-ramme sletting av et kodon for glutaminsyre, og kan føre til manifestasjon av "early-onset generalisert isolert dystoni '14,15. Imidlertid banenophysiological mekanismene som er ansvarlig for denne lidelsen er fortsatt dårlig forstått. I en knock-in mus modell, er mutant allel Tor1a tm2Wtd, nevnt heretter som Tor1a AE. Heterozygot AE-Torsina knock-in mus er levedyktige og genetisk ligne menneskelige pasienter med DYT1 dystoni, mens homozygot knock-in mus dør etter fødselen 16,17, med ventetid til postnatal død påvirket av genetisk bakgrunn 18. Tidlige død homozygot knock-in mus krever at både genotyping av dyr og etablering av nevrale kulturer er gjennomført raskt. Som et annet eksempel på genotyping, Tfap2a (transkripsjonsfaktor AP-2α, aktive enhancer bindende protein 2α) vil bli brukt. Proteinet kodet for av dette genet er viktige i regulering av flere cellulære prosesser, for eksempel proliferasjon, differensiering, overlevelse og apoptose 19.
Protokollen som presenteres her inkluderer prosedyrer for tatovering å merke / identifisere mus, for genotyping mus fra halen tips, og for dyrking mus hjernens nerveceller ved lav tetthet. I en runde av forsøk under anvendelse av 6-8 unger disse fremgangsmåter krever typisk ~ 0,5 timer, 4 timer og ~ ~ 2 timer, henholdsvis, ved en total på 6-7 timer.
