Hier presenteren we een protocol om elektrogesponnen nanovezel steigers met gradated organisatie van vezels fabriceren en hun toepassingen verkennen in het reguleren van de cel morfologie / oriëntatie. Verlopen met betrekking tot fysische en chemische eigenschappen van de nanovezel scaffolds biedt een grote verscheidenheid aan toepassingen op biomedisch gebied.
The goal of this protocol is to report a simple method for generating nanofiber scaffolds with gradations in fiber organization and test their possible applications in controlling cell morphology/orientation. Nanofiber organization is controlled with a new fabrication apparatus that enables the gradual decrease of fiber organization in a scaffold. Changing the alignment of fibers is achieved through decreasing deposition time of random electrospun fibers on a uniaxially aligned fiber mat. By covering the collector with a moving barrier/mask, along the same axis as fiber deposition, the organizational structure is easily controlled. For tissue engineering purposes, adipose-derived stem cells can be seeded to these scaffolds. Stem cells undergo morphological changes as a result of their position on the varied organizational structure, and can potentially differentiate into different cell types depending on their locations. Additionally, the graded organization of fibers enhances the biomimicry of nanofiber scaffolds so they more closely resemble the natural orientations of collagen nanofibers at tendon-to-bone insertion site compared to traditional scaffolds. Through nanoencapsulation, the gradated fibers also afford the possibility to construct chemical gradients in fiber scaffolds, and thereby further strengthen their potential applications in fast screening of cell-materials interaction and interfacial tissue regeneration. This technique enables the production of continuous gradient scaffolds, but it also can potentially produce fibers in discrete steps by controlling the movement of the moving barrier/mask in a discrete fashion.
Nanovezels zijn een populaire programma voor weefselregeneratie vanwege hun vermogen om de extracellulaire matrix in de structuur en relatieve grootte 1 nabootsen. Sommige natief weefsel interfaces, zoals de pees te bone insertieplaats bevatten collageenvezels, die een variabele organisatiestructuur die toeneemt in aanpassing aan de pees en vermindert de botplaats 2-5 vertonen. Dus, effectieve weefselregeneratie er behoefte aan een scaffold die effectief deze structurele verloop kan nabootsen fabriceren.
Eerder is er onderzoek gedaan naar geleidelijke veranderingen in vezelsamenstelling, specifiek mineraalgehalte 6. Echter, herscheppen van de structurele component van bindweefsel blijft grotendeels onontgonnen. Een eerdere studie onderzocht morfologische verlopen door het bestuderen van het effect van oppervlakte silica deeltjesdichtheid op de proliferatie van rat calvarial osteoblasten en vond een inverse verhouding tussen siliciumoxide deeltjesdichtheid en celproliferatie 7. Maar de morfologische veranderingen die celproliferatie gemedieerde in eerdere werk waren meestal gerelateerd aan oppervlakte-ruwheid ontbreekt het vermogen in het nabootsen van vezels organisatorische 7,8 veranderingen. Een recente studie geprobeerd om een steiger die de unieke collageen vezeloriëntaties nagebootst met behulp van een nieuwe collector voor elektrospinnen 9 fabriceren. Hoewel deze studie geslaagd in het produceren van een steiger met zowel uitgelijnd en willekeurige vezels, is het niet aan de geleidelijke veranderingen tentoongesteld in de inheemse weefsels na te bootsen. Ook in het produceren van afzonderlijke componenten, met een onmiddellijke verandering van afgestemd op willekeurige oriëntatie, de biomechanische eigenschappen van deze steiger aanzienlijk afgenomen. Geen eerdere werk heeft kunnen toepasselijke nanovezel steigers te produceren met continue gradaties in vezeloriëntaties uit uitgelijnd en willekeurige geweest. Onze recente studie heeft aangetoond succesvol recreatie van nanovezel steigersmet gradaties aan vezels organisatie die mogelijk de inheemse collageen organisatie op pees-to-bone inbrengen 10 kan nabootsen. Dit werk beoogt de protocollen voor de productie van nanovezels scaffolds met een structuur die lijkt op die van vezels organisatie in de natieve pees naar botweefsel-interface aanwezig.
Gradiënt nanovezel structuren zijn mogelijk verregaande toepassingen in een verscheidenheid van terreinen. We concentreerden ons op de applicaties om tissue engineering van de pees-to-bone inbrengen site door het combineren van onze steigers met-vet-afgeleide stamcellen (ADSCs) die al worden gebruikt voor weefselregeneratie op diverse ondergronden 11-14. Daarnaast ADSCs zijn zeer vergelijkbaar in de natuur te beenmerg stamcellen in termen van multipotentie en hun bron is er in overvloed die kan worden geoogst met behulp van een eenvoudige liposuctie procedure 15,16. Zaaien deze cellen te gradated nanovezel steigers verder verbetert hun tissue technische toepassingen door de gecontroleerde verdeling van de cellen die potentieel kunnen differentiëren in verschillende weefsels. In aanvulling op het zaaien van stamcellen, kunnen nanovezels worden ingekapseld met signaalstoffen voor de regulatie van cellulaire respons. Koppeling nanoencapsulation de organisatorische verloop van deze scaffolds maakt voor de studie van cellulaire gedrag of mogelijke implantaatontwerpen en coatings. Inkapseling van functionele moleculen zoals bone morphogenetic protein 2 (BMP2), waarvan is aangetoond dat osteoblastdifferentiatie 15,16 induceren, zou verdere versterking van de tissue engineering toepassingen van deze scaffolds 10.
The most critical part of the protocol is generation of the gradient scaffold. It is imperative that the mask covering the collector moves at a constant velocity so there is a gradual change within the fiber scaffold. The correct preparation of PCL solution is also important to ensure electrospinning success. Checking the fiber morphology prior to electrospinning is recommendable, especially after the encapsulation of Coumarin-6, which may require a higher voltage to electrospin correctly.
