Glaucoma is characterized by damage to retinal ganglion cells. Inducing glaucoma in animal models can provide insight into the study of this disease. Here, we outline a procedure that induces loss of RGCs in an in vivo rat model and demonstrates the preparation of whole-mount retinas for analysis.
Glaucoom is een ziekte van het centrale zenuwstelsel die retinale ganglioncellen (RGC's). RGC axonen die deel uitmaken van de oogzenuw te voeren visuele input naar de hersenen voor visuele waarneming. Schade aan RGC en hun axonen leidt tot verlies van het gezichtsvermogen en / of blindheid. Hoewel de specifieke oorzaak van glaucoom is niet bekend, de belangrijkste risicofactor voor de ziekte een verhoogde intraoculaire druk. Het protocol in dit document beschrijft een werkwijze voor het induceren glaucoom – achtige omstandigheden in een in vivo rattenmodel wanneer 50 gl 2 M hypertone zoutoplossing wordt geïnjecteerd in de episclerale veneuze plexus. De resultaten van dit lab blijkt dat ongeveer 25% van RGC verloren binnen een maand na de procedure in vergelijking met interne controles. Deze procedure maakt de kwantitatieve analyse van retinale ganglion celdood in een in vivo rat model glaucoom.
Glaucoom is een groep van oogziekten invloed neuronen in het netvlies, in het bijzonder, de retinale ganglioncellen 1-2. De axonen van deze cellen convergeren naar de oogzenuw uitvoering visuele informatie naar de hersenen waar het zicht wordt waargenomen worden. Schade aan RGC en hun axonen veroorzaakt daarom visuele gebreken.
De belangrijkste kenmerken die geassocieerd worden met glaucoom aandoeningen zijn RGC degeneratie en dood, verhoogde intra-oculaire druk (IOP) en optische schijf cupping en atrofie. Deze kenmerken leiden tot gezichtsveld verlies of volledige, onomkeerbare blindheid. Op dit moment, glaucoom heeft blindheid veroorzaakt in 70 miljoen mensen wereldwijd 3. Als zodanig is het 's werelds derde grootste oorzaak van blindheid 4.
Het exacte mechanisme van RGC dood in glaucoom blijft onbekend. Er is veel onderzoek gedaan om het mysterie te ontgrendelen. Het is echter bekend dat de primaire risicofactor glaucoom verhoging in intraoculaire druk door onregelmatige circulatie van waterig lichaamsvocht (AH) in de voorste kamer van het oog. AH fungeert als een transparante en kleurloze vervanging voor bloed in de avasculaire voorste kamer van het oog. Het voedt de omringende cellen, verwijdert uitgescheiden afvalproducten uit metabolische processen, transporten neurotransmitters, en maakt de verspreiding van drugs en inflammatoire cellen in het oog tijdens pathologische toestanden 1.
Het onderhoud van waterige humor omloop betreft het ciliaire lichaam en het trabeculaire netwerk. Kamerwater wordt geproduceerd door het corpus ciliare. Vervolgens stroomt in de voorste kamer naar de algehele gezondheid van het oogweefsel handhaven. 75-80% van de waterige humor uitstroom actief uitgescheiden via niet-pigment ciliaire epitheel wanneer het fluïdum wordt gefiltreerd door drie lagen sponsachtige weefsel in de ciliaire spier. De vloeistof verlaat via de trabeculaire netwerk en via het kanaal van Schlemm die emptven in het bloedsysteem 5 .De resterende 20-25% van uitstroom omzeilt het trabeculaire netwerk en passief uitgescheiden door ultrafiltratie en diffusie door de uveo-sclerale route. Deze route lijkt relatief onafhankelijk van de intraoculaire druk 1 zijn.
Wanneer waterige humor productie en uitstroom uit balans zijn, druk opbouwt in het oog. Zoals gezegd, deze toename van de intraoculaire druk is de belangrijkste risicofactor voor het ontwikkelen van glaucoom. Dergelijke druk veroorzaakt schade aan de ingewikkelde lagen van neuronen in het netvlies aan de achterkant van het oog. Beschadiging van de retinale ganglioncellen axonen van de oogzenuw veroorzaakt de hersenen om nauwkeurige visuele informatie niet meer ontvangen. Hierdoor wordt de perceptie van het gezichtsvermogen verloren en volledige blindheid kan optreden.
Tot op heden is er geen behandeling voor glaucoom. Verschillende behandelmethoden bestaan die in de eerste plaats gericht zijn op de intra-oculaire druk te verminderen. Deze omvatten actueelmedicatie klassen zoals beta1-adrenerge receptorblokkers of topische prostaglandinederivaten. Bèta blokkers verlagen de intraoculaire druk door het verlagen van de productie van kamerwater 7. Prostaglandinen functie om IOP te verminderen door de afvoer van kamerwater 8-14. Alfa- adrenergische agonisten en koolzuuranhydraseremmers worden ook gebruikt als secundaire behandelingsmethoden. Alpha adrenerge agonisten verhogen uitstroom door de uveosclerale route 15-17. Koolzuuranhydraseremmers vermindering van de productie van AH door enzymatische remming 18. Veel meer invasieve procedures worden ook gebruikt voor de behandeling van glaucoom. Lasertrabeculoplastie wordt gebruikt om de uitstroom van waterige humor 19 verhogen. Andere chirurgische therapie, genaamd trabeculectomie, creëert een alternatieve drainage site AH filteren wanneer de traditionele trabeculaire route geblokkeerd 20-21.
Deze behandelingen zijn bekend om effectief verminderen IOP. Echter, tot 40% van glaucoom patiënten vertonen normale IOP niveaus en de noodzaak van vollediger therapeutische werkwijzen. 22,23 Bovendien retinale ganglion celdood waargenomen bij glaucoom is onomkeerbaar wanneer het begint en de huidige behandelingen niet de progressie van de ziekte te stoppen 24-28. Dit heeft gewezen op de noodzaak van effectieve neuroprotectieve therapieën die het overleven van de neuronen zelf richten. Ontwikkeling van glaucoom modellen is cruciaal voor deze ontwikkeling.
In deze studie demonstreren een werkwijze voor het induceren glaucoom-achtige effecten in volwassen Long Evans ratten onder toepassing van een gemodificeerde werkwijze die oorspronkelijk beschreven door Morrison 29. In deze procedure injecties van 2 M hypertone zoutoplossing in de episclerale veneuze plexus induceert glaucoom-achtige omstandigheden door littekens weefsel waterige humor uitstroom verlagen in het trabeculaire netwerk leidt tot een toename van intraoculaire druk en een aanzienlijk verlies van RGC within een maand na de procedure 30-31. Glaucoom inducerende procedures, zoals hier beschreven, kan de sleutel tot nieuwe ontwikkelingen in glaucoom behandelingen.
Dit protocol beschrijft een werkwijze voor het induceren glaucoom-achtige omstandigheden in een in vivo rattenmodel.
