The design-of-experiments procedure presented here allows the evaluation of different flocculants in terms of their ability to aggregate dispersed particles in plant extracts, thus reducing turbidity and the costs of downstream processing.
Plants are important to humans not only because they provide commodities such as food, feed and raw materials, but increasingly because they can be used as manufacturing platforms for added-value products such as biopharmaceuticals. In both cases, liquid plant extracts may need to be clarified to remove particulates. Optimal clarification reduces the costs of filtration and centrifugation by increasing capacity and longevity. This can be achieved by introducing charged polymers known as flocculants, which cross-link dispersed particles to facilitate solid-liquid separation. There are no mechanistic flocculation models for complex mixtures such as plant extracts so empirical models are used instead. Here a design-of-experiments procedure is described that allows the rapid screening of different flocculants, optimizing the clarification of plant extracts and significantly reducing turbidity. The resulting predictive models allow the identification of robust process conditions and sets of polymers with complementary properties, e.g. effective flocculation in extracts with specific conductivities. The results presented for tobacco leaf extracts can easily be adapted to other plant species or tissues and will thus facilitate the development of more cost-effective downstream processes for commodities and plant-derived pharmaceuticals.
Planten worden wijd gebruikt om levensmiddelen zoals vruchtensappen produceren, maar kunnen ook worden ontwikkeld als platforms voor de vervaardiging van hogere waarde biofarmaceutische producten 1-3. In beide gevallen, downstream processing (DSP) begint vaak met het verwijderen van vloeistoffen uit weefsels zoals bladeren en vruchten, gevolgd door de verduidelijking van deeltjes beladen extracten 4,5. Voor de vervaardiging van geneesmiddelen, kunnen de kosten van DSP oplopen tot 80% van de totale productiekosten 6,7 en dit gedeeltelijk weerspiegelt de hoge belasting deeltjes aanwezig in extracten bereid met werkwijzen zoals verstorende-blade gebaseerde homogenisering 8,9 . Hoewel de rationele selectie filterlagen de deeltjesgrootteverdeling overeen in het extract kan filtercapaciteit en lagere kosten 10,11, kan de verbetering niet meer dan een maximum van bepaald door het aantal deeltjes dat per behouden moeten blijven absolute capaciteit-eenheid van het filter gebied om opheldering te bereiken.
Het plafond kan worden opgeheven indien minder deeltjes het oppervlak van de beste filters in de filtratietrein bereiken en dit kan worden bereikt als gedispergeerde deeltjes zijn gemengd met polymeren bekend als vlokmiddelen die aggregatie bevorderen grote vlokken 12 vormen. Dergelijke vlokken kunnen verder stroomopwaarts van grovere en goedkoper zakkenfilters worden gehandhaafd, waardoor de deeltjes belasting bereiken hoe fijner en duurder dieptefilters. De polymeren moeten veiligheid profielen die geschikt zijn voor hun applicaties, bijvoorbeeld voor biofarmaceutische producten ze moeten voldoen aan Good Manufacturing Practice (GMP) te hebben, en meestal moeten ze een molmassa> 100 kDa hebben en kan zowel neutraal of geladen 13. Overwegende neutrale flocculatoren het algemeen werken door het verknopen van verspreide deeltjes waardoor hun aggregatie en de vorming van vlokken met een diameter> 1 mm 11, geladen polymeren neutraliseren van de lading van dispersed deeltjes, waardoor hun oplosbaarheid en derhalve precipitatie 14 veroorzaakt.
Flocculatie kan worden verbeterd door het instellen van parameters zoals pH of geleidbaarheid, en het polymeer type of concentratie op de eigenschappen van het extract 15,16 passen. Tabak extracten voorbehandeld met 0,5-5,0 g L -1 polyethyleenimine (PEI), een meer dan 2-voudige toename dieptefilter capaciteit werd gerapporteerd in een 100-L proefschaal proces. De kosten van dit polymeer is minder dan € 10 kg -1, zodat de invoering ervan in het proces heeft geleid tot een kostenbesparing van ongeveer € 6.000 voor filters en verbruiksmaterialen per batch 16 of zelfs meer in combinatie met cellulose gebaseerde filter aids 17. Toch zijn voorspellende modellen nodig om de a priori economische voordelen van flocculanten evalueren, omdat hun opname greep stappen van 15-30 min 16,18 kan eisen, wat resulteert in verdere investeringen kosten voor opslagtanks. Echter, er zijn geen mechanistische modellen beschikbaar die de uitkomst van dergelijke experimenten kunnen voorspellen als gevolg van de complexe aard van flocculatie. Daarom is een meer passend ontwerp-of-experimenten (DoE) benadering 19 werd ontwikkeld zoals beschreven in dit artikel. Een protocol voor de algemene DoE procedure is onlangs verschenen 20.
Kleinschalig apparaten zijn nu beschikbaar voor de high-throughput screening van flocculatie omstandigheden 21. Echter, deze apparaten niet realistisch simuleren tijdens het uitvlokken van plantenextracten vanwege de afmetingen van het reactievat (~ 7 mm voor putten op een 96-wells plaat) en de deeltjes of vlokken kleiner dan een orde van grootte van elkaar kunnen zijn. Dit kan invloed mengen patronen en aldus de voorspellende kracht van het model. Bovendien kan het moeilijk zijn om de omvang van processen waarbij precipitatie als gevolg van niet-lineaire veranderingen in de menggedrag en neerslag stabaarheid 22. Daarom is dit artikel schetst een bench-top-scale screening systeem met een debiet van 50-75 monsters per dag, waardoor de resultaten die schaalbaar van de eerste 20 ml reactie volume op een 100 liter pilot-schaal proces 16 zijn. In combinatie met een DoE benadering, dit maakt de voorspellende modellen worden gebruikt voor procesoptimalisatie en documentatie van een kwaliteit-van-concept.
De hieronder beschreven methode kan ook worden aangepast om biofarmaceutica geproduceerd in celkweek gebaseerde processen, waarbij flocculanten ook als een kostenbesparende gereedschap 23 overwogen. Het kan ook worden gebruikt voor het neerslaan van doeleiwitten modelleren van een ruw extract als onderdeel van een zuiveringsstrategie, zoals aangetoond voor β-glucuronidase geproduceerd in canola, maïs en sojaboon 24,25. Een gedetailleerde beschrijving van flocculant eigenschappen kunnen elders worden gevonden 16,26 en het is belangrijk dat het polymeer concentraties ofwel niet giftig of minder schadelijke niveaus in het eindproduct 11.
Het belangrijkste aspect om te overwegen bij het opzetten van een hinde naar deeltje flocculatie karakteriseren is dat het ontwerp moet in principe in staat zijn om de verwachte of mogelijke effecten 36,38, bijvoorbeeld de invloed van de pH, het type polymeer en polymeerconcentratie 16 detecteren en te beschrijven.
