I denne protokol, vi beskriver teknikker til ordentlig dissektion af Arabidopsis blomster og siliques, nogle grundlæggende clearing teknikker, og valgt Farvning procedurer for hele-mount observationer af reproduktive strukturer.
På grund af dets formidable værktøjer til molekylær genetiske undersøgelser er Arabidopsis thaliana en af de mest fremtrædende model arter i plantebiologi og navnlig plante reproduktiv biologi. Dog inddrage plante morfologiske, anatomiske og ultrastrukturelle analyser traditionelt tidskrævende indlejring og skæring procedurer til lysfelt, scanning og elektronmikroskopi. De seneste fremskridt i Konfokal Fluorescens mikroskopi, state-of-the-art 3D-computer-aided mikroskopiske analyser og løbende forfinelse af molekylære teknikker skal anvendes på minimalt forarbejdede hele-mount prøver, har ført til øget efterspørgsel efter udvikle effektive og minimal prøve forarbejdningsteknik. I denne protokol, vi beskriver teknikker til korrekt dissekere Arabidopsis blomster og siliques, grundlæggende clearing-teknikker, og nogle farvning procedurer for hele-mount observationer af reproduktive strukturer.
Blomster blandt de vigtigste definere organer af dækfrøede planter. Blomsterplanter dukkede op omkring 90-130 millioner år siden1, og diversificeret så hurtigt, at deres hurtige udseende blev beskrevet som en “afskyelige mysterium” af Charles Darwin2. Plante forskere interesser i blomst udvikling er forskellige. Nogle forskning har fokuseret på at forstå den evolutionære oprindelse af blomsten som helhed, eller udviklingen af særlige anatomiske, strukturelle og funktionelle egenskaber af blomster3,4,5,6 . Den høje variation i blomstermotiver form og struktur, samt tilstande af kønnet og ukønnet reproduktion påberåbe dem, gøre blomsten en meget kompleks struktur. Dette har ført til forskellige bestræbelser på at karakterisere de anatomi og strukturelle elementer i blomstermotiver organer, ved hjælp af lys og elektron mikroskopiske teknikker, der kan kombineres med genetiske og molekylære undersøgelser7. Desuden, som kilde af frugter og frø, er blomster af afgørende betydning for menneskers og dyrs ernæring. Derfor, karakterisering af blomst og frugt udvikling har mange konsekvenser for anvendt forskning, herunder fødevaresikkerhed for en voksende befolkning og økologisk bevarelsesstrategier under et skiftende miljø8 , 9 , 10.
Blomst udvikling i Arabidopsis starter med blomst induktion og omdannelsen af den vegetative meristem til en blomsterstand (gruppe af blomster) meristem. Blomst primordia initieres lateralt på flanken af blomsterstand meristem11. Floral orgel primordia danne gradvis i koncentriske hvirvler udefra til midten af blomsten, og i sidste ende udvikle sig til bægerblade, kronblade, støvdragere og frugtblade7. Disse blomsteragtige organer opfylde særskilte nærende, beskyttende og funktionelle (fxbestøver tiltrækning) roller i forskellige plantearter, med kønsorganer fastholde udviklingen af mandlige og kvindelige gametofytter, henholdsvis12 , 13. gametofytter, til gengæld hver skelne et par mand (sæd) og kvindelige kønsceller (æg og central celle), der forener på dobbelt befrugtning danner den næste generation, de befrugtede æg, og den primære frøhvide, en terminal væv støtte den udviklingen af fosteret14,15. Frugt og frø udvikling støtter vækst, modning og opbevaring af embryoner og i sidste ende, dens spredning. Omfattende forskning er blevet udført for at karakterisere blomst og embryo udvikling i forskellige plantearter, især i arternes model Arabidopsis7,16,17.
Tidlig mikroskopiske analyser af blomst udvikling var baseret på tidskrævende prøve behandling og observation teknikker, såsom paraffin eller harpiks indlejring og skæring, kombineret med lys eller Elektron Mikroskopi. Disse traditionelle mikroskopiske teknikker blev ofte brugt i kombination med molekylær genetiske undersøgelser, som mikroskopiske analyser af mutanter, lokalisering af RNA ved i situ hybridisering eller immuno-påvisning af proteiner. De seneste fremskridt i wide-felt og konfokal Fluorescens mikroskopi, state-of-the-art 3-D computer-aided billede analyser og løbende forfinelse af molekylære metoder, der kan bruges på minimalt forarbejdede hele-mount prøver, har ført til et behov for effektiv og minimal prøve databehandling, der er fortrinsvis indstillet til kvantitative analyser. I de seneste år, er der sket betydelige fremskridt i udviklingen af clearing teknikker på hele-mount animalske modellen. De render prøven gennemsigtig enten ved hjælp af vandig urea – eller sukker-baserede reagenser (fxSCALE, SeeDB, CUBIC)18,19,20, eller ved selektivt at fjerne lipider (ved hjælp af vaskemiddel SDS) efter indlejring prøver i stabile hydrogels; fjernelse af lipider kan opnås enten ved passiv diffusion (f.eks.ændret klarhed protokol21, PAGTEN-PARS-fælge22) eller aktivt ved elektroforese (oprindelige klarhed protokol23 og ACT-PRESTO24). Opmuntret af denne hurtige fremskridt, nogle afledte teknikker også vej til brug i planter.
I denne metoder papir fokuseret på model Arabidopsis, beskrive vi proceduren for ordentlig dissektion af blomsterknopper, blomster, og unge siliques og clearing af hele-mount prøver til forskellige farvning og observation procedurer ved hjælp af klassisk eller en nylig SDS-baserede clearing metode. Gives eksempler for stivelse, callose og kromatin farvning. Selvom disse procedurer kan have yderligere forbedringer og tilpasninger når det bruges med andre arter, håber vi, de vil sætte scenen for yderligere forskning på disse simple men kritiske metoder, der er udgangspunktet for mange forskningsprojekter.
Eksistensen af mange blomsterknopper inden for en enkelt blomsterstand af Arabidopsis, der spænder over alle blomst udviklingsstadier, tilbyder en unik mulighed for undersøgelser rettet mod kendetegner en effekt af en behandling eller en udviklingsmæssig funktion samtidigt på tværs af de forskellige stadier af blomst udvikling.
