Dit protocol illustreert een veilige en efficiënte procedure voor het wijzigen van de CD133+ hematopoietische stamcellen. De voorgestelde niet-virale, magnetische polyplex gebaseerde benadering verschaffen een basis voor de optimalisatie van de cel van de stam van de therapeutische effecten ook monitoring van het product door de overheid gereguleerde cel via magnetische resonantie beeldvorming.
Terwijl CD133+ hematopoietische stamcellen (gcv) zijn bewezen om hoog potentieel op het gebied van regeneratieve geneeskunde, hun lage retentie tarieven na injectie in benadeelde weefsels, alsmede de waargenomen massale cel dood tarieven leiden tot zeer beperkte therapeutische effecten. Deze beperkingen wilt opheffen, we geprobeerd om een niet-virale gebaseerd protocol voor geschikt cel engineering voorafgaand aan hun administratie. De wijziging van menselijke CD133+ uiten van SCs met behulp van magnetische polyplexes microRNA (miR) geladen werd behandeld met betrekking tot de opname efficiëntie en veiligheid alsmede de targeting mogelijkheden van de cellen. Een beroep op ons protocol, kunnen wij hoge miR opname tarieven van 80-90% terwijl de CD133+ stamcel eigenschappen blijven onaangetast. Bovendien, deze gewijzigde cellen bieden de mogelijkheid van magnetische targeting. Hier beschrijven we een veilige en zeer efficiënte procedure voor de wijziging van de CD133+ SCs. Wij verwachten dat deze aanpak om een standaardtechnologie voor optimalisatie van de cel van de stam van de therapeutische effecten en voor de controle van het product door de overheid gereguleerde cel via magnetische resonantie beeldvorming (MRI).
CD133+ SCs vertegenwoordigen een heterogene stam en progenitor cel bevolking met het veelbelovende potentieel voor regeneratieve geneeskunde. Hun hematopoietische, endotheel en myogenic differentiatie potentiële1,2,3 maakt de CD133+ cellen, bijvoorbeeldbij te dragen tot neovascularization processen door middel van differentiatie in nieuw vorming van vaartuigen en activering van pro-angiogenic signalering door paracrine mechanismen4,5,6,7.
Ondanks hun hoge potentieel aangetoond in meer dan 30 erkende klinische proeven (ClinicalTrails.gov), is hun therapeutische resultaat nog steeds onder de controversiële discussie4. Inderdaad, een klinische toepassing van SCs wordt belemmerd door lage retentie in het orgel van belang en massale eerste cel dood5,8,9. Extra engineering van CD133+ SCs voorafgaande transplantatie kan helpen deze uitdagingen.
Een voorwaarde voor een efficiënte celtherapie zou de verlaging van de massale eerste celdood ter verbetering van de engraftment van therapeutische relevante cellen10. Huidige studies aangetoond een immense cel verlies van 90-99% in zeer geperfundeerd organen zoals de hersenen en het hart tijdens de eerste 1-2 h, onafhankelijk van de getransplanteerde celtype of toepassing route11,12,13 ,14,15,16,17,18,19,20,21. SC labelen met behulp van magnetische nanodeeltjes (MNPs) kan een niet-invasieve innovatiestrategie op target cellen op de site van belang22,23,24,25,26 en tegelijkertijd zorgt voor de cel opvolging met behulp van MRI27 en magnetische deeltjes imaging (MPI). De meest efficiënte in vivo studies toepassen gemagnetiseerde cel gericht op behoud van de gebruikte cel na lokale toediening boven cel begeleiding na intraveneuze injectie23,24,28 . Daarom is onze fractie ontworpen een levering systeem bestaande uit superparamagnetische ijzeroxide nanodeeltjes29. Met deze techniek, CD133+ SCs en menselijke navelstreng ader endotheliale cellen (HUVECs) kunnen efficiënt worden gericht, zoals blijkt uit in vitro probeert30,31.
Een andere hindernis voor SC therapieën is de vijandige inflammatoire omgeving van het getroffen weefsel na de transplantatie, die tot de eerste cel dood32 bijdraagt. Naast verschillende studies van de pre-conditionering was de toepassing van therapeutische relevante miRs geteste33; succesvol is gebleken dat anti-apoptotic miRs apoptosis in vitro remmen en cel engraftment in vivo33 verbeteren. Deze kleine molecules, samengesteld uit de 20-25 nucleotiden, spelen een cruciale rol als posttranscriptional modulatoren van messenger RNAs (mRNAs) en dus beïnvloeden stamcel lot en gedrag34. Bovendien voorkomen de exogene invoering van miRs de ongewenste stabiele integratie in de host genoom34.
Huidige pogingen voor efficiënte binnenbrengen van nucleic zuren (NAs) primaire SCs zijn meestal gebaseerd op recombinante virussen8,35. Ondanks de hoge transfectie efficiëntie presenteert recombinante virus manipulatie een groot obstakel voor een vertaling van de Bank-naar-bed, bijvoorbeeld, oncontroleerbare genexpressie, pathogeniteit, immunogeniciteit en dat mutagenese35 ,,36. Dus, niet-virale levering systemen zoals polymeer gebaseerde constructies zijn kritisch te ontwikkelen. Onder degenen, polyethylenimine (PEI) vertegenwoordigt een geldige leveringsvoertuig biedt voordelen voor miRs zoals nb condensatie te behoeden voor degradatie, cellulaire opname, en intracellulaire release via endosomal ontsnappen37,,38. MiR-PEI complexen blijkt bovendien een hoge biocompatibiliteit in klinische proeven39. Daarom onze levering systeem bestaat uit een biotinyleerd vertakte 25 kDa PEI gebonden aan een daar beklede MNP-core30,31,40.
In dit manuscript, presenteren wij een uitgebreide protocol beschrijven (i) de handmatige Isolatievan CD133+ SC van de donatie van menselijke beenmerg (BM) met een gedetailleerde karakterisering van het product van de SC en (ii) een efficiënte en zachte transfectie strategie van een Magnetisch niet-virale polymeer gebaseerde expresbezorgingssysteem voor genetische manipulatie van CD133+ SCs miRs gebruiken. CD133+ SCs zijn geïsoleerd en magnetisch verrijkt van menselijke sternale BM aangeblazen met behulp van een oppervlakte antilichaam gebaseerde magnetische-geactiveerde cel sorteren (MACS) systeem. Daarna worden de levensvatbaarheid van de cellen, alsook de zuiverheid van de cel geanalyseerd met behulp van stroom cytometry. Vervolgens, miR/PEI/MNP complexen zijn bereid en CD133+ SCs zijn transfected. 18 h na transfectie, de opname doeltreffendheid en het effect van transfectie op SC marker expressie en cel levensvatbaarheid worden geanalyseerd. Bovendien is evaluatie van de intracellulaire verdeling van de transfectie complexe verbindingen wordt uitgevoerd met behulp van vierkleuren labeling en gestructureerde verlichting microscopie (SIM).
