Este protocolo ilustra un procedimiento seguro y eficaz para modificar CD133+ las células madre hematopoyéticas. El presentado no virales, magnético polyplex enfoque puede proporcionar una base para la optimización de los efectos terapéuticos de células madre, así en cuanto a la supervisión del producto celular administrada a través de la proyección de imagen de resonancia magnética.
Mientras CD133+ las células madre hematopoyéticas (SCs) se han demostrado para proporcionar alto potencial en el campo de la medicina regenerativa, sus tasas de retención baja después de la inyección en los tejidos lesionados, así como las tasas de muerte celular masiva observada conducen a muy efectos terapéuticos restringidos. Para superar estas limitaciones, intentamos establecer un protocolo basado no-virales para la ingeniería celular adecuado antes de su administración. La modificación de CD133 humano+ expresando SCs utilizando microARN (miR) cargada magnética polyplexes fue tratado con respecto a la eficiencia de absorción y seguridad así como el potencial de segmentación de las células. Confiando en nuestro protocolo, podemos lograr tasas de absorción miR alta de 80 – 90% mientras el CD133+ propiedades de células madre siguen siendo inafectadas. Además, estas células modificadas ofrecen la opción de orientación magnética. Aquí describimos un procedimiento seguro y altamente eficiente para la modificación de CD133+ SCs. Esperamos que este enfoque para proporcionar una tecnología estándar para la optimización de los efectos terapéuticos de células madre y para el seguimiento del producto celular administrada a través de la proyección de imagen de resonancia magnética (MRI).
CD133+ SCs representan una población heterogénea de células de vástago y del progenitor con prometedor potencial para la medicina regenerativa. Su diferenciación Hematopoyética, endoteliales y miógeno potencial1,2,3 permite la CD133+ de las células, por ejemplo, para contribuir a los procesos de neovascularización a través de la diferenciación en la formación de nuevos vasos y la activación de pro-angiogénica señalización paracrina mecanismos4,5,6,7.
A pesar de su alto potencial demostrado en más de 30 ensayos clínicos aprobados (ClinicalTrails.gov), su resultado terapéutico todavía está bajo discusión controversial4. De hecho, una aplicación clínica de SCs se ve obstaculizada por la baja retención en el órgano de interés y de la célula inicial masiva muerte5,8,9. Adicional de CD133+ SCs trasplante previo podría ayudar a superar estos desafíos.
Un requisito para una terapia celular eficiente sería la reducción de la muerte de la célula inicial masiva para mejorar el engraftment de células pertinentes terapéutica10. Los estudios actuales demuestran una pérdida inmensa de la célula del 90 – 99% en órganos altamente perfundidos como el cerebro y el corazón durante las primeras 1-2 h, independientemente del tipo de célula trasplantado o aplicación ruta11,12,13 ,14,15,16,17,18,19,20,21. SC etiquetado utilizando nanopartículas magnéticas (MNPs) permite una estrategia invasiva innovadora a las células blanco para el sitio de interés22,23,24,25,26 y al mismo tiempo permite el celular control usando MRI27 y partículas magnéticas (MPI) la proyección de imagen. Más eficiente en vivo estudios aplicando células magnetizadas a retención celular usado después de la administración local preferentemente dirección de celda después de inyección intravenosa23,24,28 . Por lo tanto, nuestro grupo diseñó un sistema de entrega que consta de óxido de hierro superparamagnético nanopartículas29. Con esta técnica, CD133+ SCs y las células endoteliales de vena umbilical humano (HUVECs) eficiente podrían ser objetivo, como lo demuestra en vitro trata de30,31.
Otro obstáculo para las terapias de la SC es el hostil entorno inflamatorio del tejido afectado después del trasplante, que contribuye a la muerte celular inicial del32. Además de varios estudios de acondicionamiento previo, la aplicación de terapéuticas relevantes miRs fue probado33; se ha demostrado con éxito que la anti-apoptotic miRs inhiben apoptosis en vitro y el engraftment de la célula en vivo33. Estas moléculas pequeñas, compuesto por 20-25 nucleótidos, desempeñan un papel crucial como moduladores postranscripcional de mensajero RNAs (mRNAs) y así afectan el destino y el comportamiento de la célula de vástago34. Por otra parte, la introducción exógena de miRs evitar la no deseada integración estable en el anfitrión genoma34.
Intentos actuales para la introducción eficiente de los ácidos nucleicos (NAs) en SCs primarios en su mayoría se basan en virus recombinantes8,35. A pesar de la eficacia de transfección alta manipulación de virus recombinante presenta un gran obstáculo para una traducción de banco a cabecera, p. ej., expresión génica incontrolable, patogenicidad, inmunogenicidad y mutagénesis de insertional35 ,36. Por lo tanto, son críticos para desarrollar sistemas de vectores no virales como construcciones basados en polímeros. Entre los, polietilenimina (PEI) representa un vehículo de entrega válida que ofrece beneficios para la miRs como condensación de NA para proteger de la degradación, absorción celular, y liberación intracelular a través de endosomal escapar37,38. Además, complejos de miR-PEI demostraron una alta biocompatibilidad en ensayos clínicos39. Por lo tanto, nuestro sistema consiste en un biotinilado ramificado 25 kDa PEI enlazado a un cubierto de streptavidin MNP-core30,31,40.
En este manuscrito, presentamos un protocolo completo que describe (i) el aislamiento manual de CD133+ SC de donación de médula ósea humana (BM) con una caracterización detallada del producto de SC y (ii) una estrategia de transfección eficiente y suave de un sistema de entrega basados en polímeros magnéticamente no-virales para la ingeniería genética de CD133+ SCs utilizando miRs. CD133+ SCs son aislados y magnéticamente enriquecido de humano aspira BM esternal utilizando una superficie basados en anticuerpos activada por el magnético celular (MACS) sistema de clasificación. Luego, la viabilidad de las células, así como la pureza de la célula se analiza mediante citometría de flujo. Posteriormente, se preparan el miR/PEI/MNP complejos y CD133+ SCs son transfectadas. 18 h después de la transfección, la eficiencia de la absorción y el impacto de la transfección en SC marcador expresión celular la viabilidad y se analizan. Por otra parte, la evaluación de la distribución intracelular de los compuestos complejos de transfección se realiza utilizando etiquetado del cuatro-color y microscopía de iluminación estructurada (SIM).
