Este protocolo ilustra um procedimento seguro e eficiente para modificar CD133+ células-tronco hematopoiéticas. A apresentado não-virais, magnética polyplex abordagem pode fornecer uma base para a otimização dos efeitos terapêuticos células-tronco, bem como acompanhamento do produto celular administrado via ressonância magnética.
Enquanto CD133+ células-tronco hematopoiéticas (SCs) tem sido comprovadas para fornecer alto potencial no campo da medicina regenerativa, suas taxas de retenção baixa após a injeção em tecidos feridos, bem como as taxas de mortalidade observadas células maciça levam a muito restrito de efeitos terapêuticos. Para superar essas limitações, nós procurou estabelecer um protocolo baseado em não-viral para engenharia celular adequada antes da sua administração. A modificação de CD133 humana+ expressar SCs usando os polyplexes magnético microRNA (miR) carregado foi abordada em relação a eficiência de absorção e de segurança, bem como o alvo potencial das células. Confiando em nosso protocolo, podemos conseguir taxas de absorção alta miR de 80-90%, enquanto o CD133+ Propriedades de células-tronco permanecem inalteradas. Além disso, essas células modificadas oferecem a opção de segmentação magnético. Descrevemos aqui um procedimento seguro e altamente eficiente para a modificação de CD133+ SCs. Esperamos que esta abordagem para fornecer uma tecnologia padrão para otimização dos efeitos terapêuticos de células estaminais e para acompanhamento do produto celular administrado através de ressonância magnética (MRI).
CD133+ SCs representam uma população de célula de tronco e progenitoras heterogénea com promissor potencial para a medicina regenerativa. Sua diferenciação hematopoiéticas, endoteliais e miogênico potencial1,2,3 permite o CD133+ células, por exemplo, contribuir para processos de neovascularização através da modulação em recém formando vasos e ativação de pro-angiogênico sinalização parácrina mecanismos4,5,6,7.
Apesar de seu alto potencial demonstrado em ensaios clínicos aprovados mais de 30 (ClinicalTrails.gov), seu resultado terapêutico é ainda sob polêmica discussão4. Na verdade, uma aplicação clínica de SCs é dificultada pela baixa retenção no órgão de interesse e enorme pilha inicial morte5,8,9. Produção adicional de CD133+ transplante prévio de SCs ajudasse a superar esses desafios.
Um pré-requisito para uma terapia celular eficiente seria a redução da morte maciça de pilha inicial para melhorar a enxertia de células relevantes terapêutico10. Estudos atuais demonstraram uma perda imensa pilha de 90 – 99% em órgãos altamente perfundidos, como o cérebro e o coração durante as primeiras horas de 1 – 2, independente do tipo de células transplantadas ou aplicativo rota11,12,13 ,14,15,16,17,18,19,20,21. SC rotulagem usando nanopartículas magnéticas (MNPs) permite uma estratégia inovadora de não-invasivo para células-alvo para o site de interesse22,23,24,25,26 e simultaneamente permite que a célula de monitoramento usando MRI27 e partícula magnética (MPI) de imagem. O mais eficiente na vivo estudos aplicando magnetizada célula alvo de retenção célula usada após a administração local, em detrimento da orientação celular após injeção intravenosa23,24,28 . Portanto, o nosso grupo projetado um sistema de entrega consiste de nanopartículas de óxido de ferro superparamagnético29. Com esta técnica, CD133+ SCs e células endoteliais da veia umbilical humana (HUVECs) com eficiência poderiam ser alvo, como demonstrado em vitro tentativas de30,31.
Um outro obstáculo para terapias de SC é o ambiente hostil e inflamatório do tecido afetado após o transplante, o que contribui para a morte de pilha inicial32. Além de vários estudos de pré-condicionamento, a aplicação de terapêuticas miRs relevantes foi testado33; foi com sucesso demonstrado que anti-apoptotic miRs inibem a apoptose em vitro e realçam a enxertia de celular na vivo33. Estas pequenas moléculas, compostas por 20-25 nucleotides, desempenham um papel crucial como moduladores posttranscriptional de RNA mensageiro (mRNAs) e, portanto, afetam células estaminais destino e comportamento34. Além disso, a introdução exógena de miRs evitar a integração estável indesejada para o anfitrião do genoma34.
Atuais tentativas para introdução eficiente de ácidos nucleicos (NAs) em SCs primárias baseiam-se principalmente a vírus recombinante8,35. Apesar da eficiência elevada do transfection, manipulação de vírus recombinante apresenta um grande obstáculo para uma tradução de banco-de-cabeceira, por exemplo, expressão gênica incontrolável, patogenicidade, imunogenicidade e mutagênese insercional35 ,36. Portanto, sistemas de entrega não-virais como construções baseadas em polímero são fundamentais para desenvolver. Entre esses, o polyethylenimine (PEI) representa um veículo de entrega válido, oferecendo benefícios para miRs como condensação de ND para proteger da degradação, absorção celular, e liberação intracelular através de CDDP escapar37,38. Além disso, complexos de miR-PEI demonstraram uma alta biocompatibilidade em ensaios clínicos39. Portanto, nosso sistema de entrega consiste de um biotinilado ramificada 25 kDa PEI acoplado a um streptavidin-revestido MNP-núcleo30,31,40.
Neste manuscrito, apresentamos um protocolo abrangente descrevendo (i) o isolamento manual de CD133+ SC de doação de medula óssea humana (BM) com uma caracterização detalhada do produto SC e (ii) uma estratégia de transfeccao eficiente e suave de um sistema baseado em polímero magneticamente não-virais entrega por engenharia genética de CD133+ SCs usando miRs. CD133+ SCs são isoladas e magneticamente enriquecido de humanos aspirados BM esternais, usando uma superfície baseado em anticorpos magnético-ativado da pilha classificação sistema (MACS). Depois disso, a viabilidade celular, bem como a pureza de célula é analisada usando citometria de fluxo. Posteriormente, miR/PEI/MNP complexos são preparados e CD133+ SCs são transfectadas. 18 h após a transfeccao, a eficiência de absorção e o impacto do transfection na viabilidade de expressão e célula de marcador de SC são analisados. Além disso, avaliação da distribuição dos compostos complexos transfeccao intracelular é executada usando rotulagem quatro cores e iluminação estruturada microscopia (SIM).
