Ce protocole illustre une procédure sûre et efficace pour modifier CD133+ des cellules souches hématopoïétiques. Présenté non-virale, magnétique polyplex approche fondée sur le peut fournir une base pour l’optimisation des effets thérapeutiques des cellules souches, aussi bien en ce qui concerne le suivi du produit cellule administré par imagerie par résonance magnétique.
Tandis que CD133+ de cellules souches hématopoïétiques (SCs) ont fait leurs preuves à fournir à fort potentiel dans le domaine de la médecine régénérative, leur taux de rétention faible après une injection à l’endommagement des tissus ainsi que les taux de mortalité cellulaire massive observée conduire à très effet thérapeutique limité. Pour surmonter ces limitations, nous avons cherché à établir un protocole de base non viraux d’ingénierie cellulaire appropriée avant leur administration. La modification des CD133 humaine+ exprimant SCs à l’aide de microARN (miR) chargé polyplexes magnétique a été adressée en ce qui concerne l’efficacité d’absorption et de sécurité, mais aussi le potentiel de ciblage des cellules. S’appuyant sur notre protocole, nous pouvons atteindre les vitesses d’absorption miR haute de 80 à 90 %, tandis que le CD133+ propriétés de cellules souches ne sont pas affectées. En outre, ces cellules modifiées offrent l’option de ciblage magnétique. Nous décrivons ici une procédure sûre et très efficace pour la modification des CD133+ SCs. Nous espérons que cette approche visant à fournir une technologie standard pour l’optimisation des effets thérapeutiques des cellules souches ainsi que la surveillance du produit administré cellulaire par l’intermédiaire de l’imagerie par résonance magnétique (IRM).
CD133+ SCs représentent une population de cellules souches et progénitrices hétérogène avec promettant potentiel pour la médecine régénérative. Leur différenciation hématopoïétique et endothéliales myogène potentiels1,2,3 permet le CD133+ cellules, par exemple, de contribuer au processus de néovascularisation par différenciation dans nouvellement formant des navires et l’activation de pro-angiogénique signalisation paracrine mécanismes4,5,6,7.
Malgré leur fort potentiel démontré dans plus de 30 essais cliniques approuvés (ClinicalTrails.gov), leur résultat thérapeutique est encore en discussion controversée4. En effet, une application clinique des SCs est entravée par la faible rétention dans l’organe d’intérêt et de la cellule initiale massif mort5,8,9. Supplémentaires d’ingénierie de CD133+ SCs avant transplantation pourrait aider à surmonter ces défis.
Une condition sine qua non pour une thérapie cellulaire efficace serait la réduction de la mort massive de cellules initiales afin d’améliorer la prise de greffe de cellules pertinentes thérapeutique10. Les études en cours ont démontré une perte immense cellule de 90 à 99 % dans fortement perfusés organes comme le cerveau et le coeur pendant les premières heures de 1 – 2, indépendamment du type de cellules transplantées ou application itinéraire11,12,13 ,14,15,16,17,18,19,20,21. SC étiquetage utilisant des nanoparticules magnétiques (MNPs) permet une stratégie novatrice et non invasif pour cibler les cellules sur le site d’intérêt22,23,24,25,26 et permet simultanément de cellule de surveillance à l’aide de MRI27 et des particules magnétiques d’imagerie (MPI). Le plus efficace in vivo études appliquant cellule aimantée ciblant la rétention de la cellule utilisée après administration locale plutôt que de l’orientation de la cellule après injection intraveineuse23,24,28 . Par conséquent, notre groupe a conçu un système de livraison consistant de nanoparticules d’oxyde de fer superparamagnétique29. Avec cette technique, CD133+ SCs et les cellules endothéliales de veine ombilicale humaine (HUVEC) pourraient être efficacement ciblées, comme l’a montré in vitro tente30,31.
Un autre obstacle pour les thérapies de SC est l’environnement hostile inflammatoire des tissus touchés après la transplantation, qui contribue à la cellule initiale mort32. En plus de plusieurs études de préconditionnement, l’application des thérapeutiques pertinentes miRs était testé33; Il a été démontré avec succès qu’anti-apoptotique miRs inhibent l’apoptose in vitro et d’améliorent la prise de greffe cellulaire in vivo33. Ces petites molécules, composés de 20 à 25 nucléotides, jouent un rôle crucial en tant que modulateurs post-transcriptionnelle des ARN messagers (ARNm) et affectent donc cellules souches sort et le comportement34. En outre, l’introduction exogène de miRs éviter l’intégration stable indésirable dans le génome de hôte34.
Les tentatives actuelles pour la mise en place efficace des acides nucléiques (NAs) dans SCs primaires reposent principalement sur des virus recombinants8,35. Malgré l’efficacité de transfection haute, manipulation virus recombinant présente un obstacle majeur pour un chevet à traduction, par exemple, l’expression des gènes incontrôlable, pathogénicité, immunogénicité et mutagenèse insertionnelle35 ,,36. Par conséquent, les vecteurs non viraux tels que les constructions à base de polymères sont essentiels d’élaborer. Parmi ceux-ci, polyéthylènimine (PEI) représente un véhicule de livraison valide offrant des avantages pour miRs tels que de la condensation de NA à protéger d’une dégradation, assimilation, et intracellulaire par endosomale échapper37,38. En outre, des complexes de miR-PEI a démontré une biocompatibilité élevée dans les essais cliniques39. Par conséquent, notre système de livraison comprend une biotinylé ramifiés 25 kDa PEI lié à une streptavidine MNP-core30,31,40.
Dans ce manuscrit, nous présentons un protocole complet décrivant (i) l’isolement manuelle des CD133+ SC de don de moelle osseuse humaine (BM) avec une caractérisation détaillée du produit SC et (ii) une stratégie de transfection efficace et doux d’une système de livraison magnétiquement non-virales base de polymères pour le génie génétique de CD133+ SCs à l’aide de miRs. CD133+ SCs sont isolées et magnétiquement enrichi de humaine ponction sternale de BM en utilisant une surface axée sur les anticorps magnétique-lancée de cellules (MACS) système de tri. Par la suite, la viabilité des cellules ainsi que la pureté de la cellule est analysée à l’aide de cytométrie en flux. Par la suite, miR/PEI/MNP complexes sont préparés et CD133+ SCs sont transfectées. 18 h après la transfection, l’efficacité de l’absorption et l’impact de la transfection sur SC marqueur expression et cellule de viabilité sont analysés. En outre, l’évaluation de la distribution intracellulaire des composés complexes transfection est effectuée illumination structurée microscopes (SIM) et de quatre couleurs d’étiquetage.
