Gewebekomplexitäten multizellulärer Systeme verwirren die Identifizierung des kausalen Zusammenhangs zwischen extrazellulären Hinweisen und individuellen zellulären Verhaltensweisen. Hier stellen wir eine Methode vor, um den direkten Zusammenhang zwischen kontaktabhängigen Cues und Divisionsachsen mit C. elegans embryo blastomeres und klebenden Polystyrolperlen zu untersuchen.
In mehrzelligen Systemen sind einzelne Zellen von den verschiedenen physikalischen und chemischen Hinweisen umgeben, die aus benachbarten Zellen und der Umgebung stammen. Diese Gewebekomplexität verwirrt die Identifizierung des kausalen Zusammenhangs zwischen extrinsischen Cues und zellulärer Dynamik. Ein synthetisch rekonstituiertes mehrzelliges System überwindet dieses Problem, indem es Forschern ermöglicht, auf einen bestimmten Hinweis zu testen und andere zu eliminieren. Hier stellen wir eine Methode zur Rekonstituierung von Zellkontaktmustern mit isolierten Caenorhabditis elegans blastomer und klebenden Polystyrolperlen vor. Die Verfahren umfassen die Entfernung von Eierschalen, blastomere Isolierung durch Unterbrechung der Zell-Zell-Adhäsion, Die Herstellung von klebenden Polystyrolperlen und die Rekonstitution von Zellzell- oder Zellperlenkontakt. Schließlich stellen wir die Anwendung dieser Methode vor, um die Ausrichtung zellulärer Teilungsachsen zu untersuchen, die zur Regulierung der räumlichen zellulären Musterung und Zellschicksalsspezifikation bei der Entwicklung von Embryonen beiträgt. Diese robuste, reproduzierbare und vielseitige In-vitro-Methode ermöglicht die Untersuchung direkter Zusammenhänge zwischen räumlichen Zellkontaktmustern und zellulären Reaktionen.
Während der multizellulären Entwicklung werden die zellulären Verhaltensweisen (z.B. Divisionsachse) einzelner Zellen durch verschiedene chemische und physikalische Hinweise spezifiziert. Zu verstehen, wie einzelne Zellen diese Informationen interpretieren und wie sie die mehrzellige Baugruppe als auftauchende Eigenschaft regulieren, ist eines der ultimativen Ziele von Morphogenesestudien. Der Modellorganismus C. elegans hat wesentlich zum Verständnis der zellulären Regulation der Morphogenese wie Zellpolarität1,Zellteilungsmuster1, Zellschicksalsentscheidung2und Gewebe-Skala-Vorschriften wie neuronale Verdrahtung3 und Organogenese4,5beigetragen. Obwohl es verschiedene genetische Werkzeuge zur Verfügung, Gewebe-Engineering-Methoden sind begrenzt.
Die erfolgreichste Tissue-Engineering-Methode in der C. elegans-Studie ist die klassische Blastomer-Isolation6; Da C. elegans Embryo von einer Eierschale und einer Durchlässigkeitsbarriere7umgeben ist, ist ihre Entfernung eines der hauptverfahren dieser Methode. Während diese blastomere Isolationsmethode eine vereinfachte Rekonstitution des Zell-Zell-Kontakts ermöglicht, erlaubt sie nicht die Eliminierung unerwünschter Hinweise; Der Zellkontakt stellt nach wie vor sowohl mechanische (z. B. Haftung) als auch chemische Hinweise dar, wodurch unsere Fähigkeit, den kausalen Zusammenhang zwischen dem Cue und dem zellulären Verhalten vollständig zu analysieren, eingeschränkt wird.
Die in diesem Papier vorgestellte Methode verwendet Carboxylat modifizierte Polystyrolperlen, die kovalent an alle amin-reaktiven Moleküle binden können, einschließlich Proteine wie Liganden. Insbesondere haben wir eine amin-reaktive Form von Rhodamine Red-X als Liganden verwendet, um Perlen sowohl visuell nachvollziehbar als auch klebend an der Zelle zu machen. Die Carboxylgruppen der Perlenoberfläche und der primären Amingruppen von Ligandenmolekülen werden durch wasserlösliches Carbodiimid 1-ethyl-3-(Dimethylaminopropyl)-Carbodiimid (EDAC)8,9gekoppelt. Erhaltene Klebeperlen ermöglichen die Auswirkungen des mechanischen Hinweises auf die Zelldynamik10.
