Analyzing the physiological properties of olfactory sensory neurons still faces technical limitations. Here we record them through perforated patch-clamp in an intact preparation of the olfactory epithelium in gene-targeted mice. This technique allows the characterization of membrane properties and responses to specific ligands of neurons expressing defined olfactory receptors.
Analysere de fysiologiske responser av olfactory sensoriske nevroner (OSN) når stimulert med spesifikke ligander er avgjørende for å forstå grunnlaget for duftdrevet atferd og deres modulering. Disse kode egenskaper avhenge sterkt på det første samspillet mellom luktmolekyler og lukte reseptor (OR) uttrykt i OSNs. Identiteten, spesifisitet og ligand spekteret av uttrykte eller er kritisk. Sannsynligheten for å finne den ligand med eller uttrykt i et OSN velges vilkårlig i epitelet er meget lav. For å løse denne utfordringen, bruker denne protokoll genetisk kodede mus som uttrykker den fluorescerende protein GFP under kontroll av promotoren av definerte ORs. OSNs ligger i en tett og organisert epitel fôr nesehulen, med nabocellene påvirke deres modning og funksjon. Olfactory sensoriske nevroner (OSN) representerer første trinn i olfactory oppfatning. Hver OSN uttrykker en olfaktorisk reseptor (OR) valgt fra et stort multigen familie 6. Det er ~ 1000 ORs uttrykt i hoved olfaktoriske epitel hos mus. På grunn av det store antallet av eller i villtype dyr, er sjansene for å registrere OSNs som uttrykker det samme eller er meget lav. For å overvinne disse begrensningene, Gene målrettet mus finnes der alle OSNs uttrykker en identifisert eller er merket med et fluorescent protein 7-9. Dessuten patch-clamp opptak av OSNs også tillate presis analyse av membranegenskaper, transduksjon pathway farmakologi, ligand / eller interaksjoner. Alle disse temaene kan neppe bli analysert ved hjelp av ekstracellulære opptak. Vi brukte denne teknikken til å overvåke svarene til OSNs uttrykker odorantreseptorer SR1 og MOR23 12,13. Gjennomførbarheten av teknikken ble bekreftet av andre grupper på MOR23 uttrykker OSNs 14 samt på andre ORs uttrykker nevroner 15,16. Overvåkingen av en definert populasjon av OSNs kan føre til analyse av deres egenskaper i mange ulike sammenhenger som for eksempel utvikling 14, aldring 17, odorant indusert plastisitet 18, og rollen til variasjoner i odoranten reseptoren sekvens i lukt kodende 15. Denne protokollen gir således et kraftig verktøy for å overvåke de funksjonelle egenskaper som er definert OSNs på membrannivå.
Muligheten av denne protokollen til riktig overvåke egenskapene til sunne OSNs er svært avhengig av kvaliteten på forberedelse. Derfor er de trinnene disseksjon er kritiske. Først er det viktig å ta hensyn til kvaliteten (pH, osmolaritet) oksygenering og temperatur (is-kald, men ikke frosset) av disseksjon medium. For det andre må manipulering av epitel med dissekere verktøy være så begrenset som mulig for å unngå skader. Endelig, er det avgjørende å oppnå et preparat så flat som mulig for å få tilgang …
