Dieses Papier stellt eine Methode, mit der Synthese und Charakterisierung von Monocyte-targeting Peptid Amphiphile Micellen und den entsprechenden assays für Biokompatibilität und Fähigkeit der Micelle binden an Monozyten zu testen.
Atherosklerose ist ein wesentlicher Faktor für kardiovaskuläre Erkrankungen, die häufigste Todesursache weltweit, der jährlich 17,3 Millionen Todesopfer fordert. Atherosklerose ist auch die häufigste Ursache des plötzlichen Tod und Myokardinfarkt, angestiftet von instabilen Plaques, die platzen und verdecken das Blutgefäß ohne Vorwarnung. Aktuelle bildgebende Modalitäten kann nicht unterscheiden zwischen stabilen und instabilen Plaques, die platzen. Peptid amphiphilen Micellen (PAMs) können diesen Nachteil überwinden, wie sie können, mit einer Vielzahl geändert werden von targeting Moieties, die speziell auf erkranktes Gewebe zu binden. Monozyten haben gezeigt, zu frühe Marker für Arteriosklerose, während große Ansammlung von Monozyten Plaques anfällig für Bruch zugeordnet ist. Daher können Nanopartikel, die Monozyten abzielen kann verwendet werden, verschiedene Stadien der Arteriosklerose zu unterscheiden. Zu diesem Zweck hier beschreiben wir ein Protokoll für die Vorbereitung des Monocyte-targeting PAMs (Monocyte Lockstoffgradient Protein-1 (MCP-1) PAMs). MCP-1 PAMs sind selbst zusammengebaut durch Synthese unter milden Bedingungen, Form Nanopartikel von 15 nm im Durchmesser mit in der Nähe von neutralen Oberflächenladung. In Vitro, PAMs erwiesen sich biokompatible und hatte eine hohe Bindungsaffinität für Monozyten. Die hier beschriebenen Methoden zeigen, Versprechen für ein breites Anwendungsspektrum in Atherosklerose sowie anderen entzündlichen Erkrankungen.
Herz-Kreislauf-Krankheiten bleiben die führenden Todesursachen weltweit mit etwa 17,3 Millionen Todesfälle weltweit1. Herz-Kreislauf-Krankheiten sind durch Arteriosklerose, eine Bedingung trugen in denen Plaques in den Arterien, dadurch hemmende Blut- und Sauerstoffzufuhr zu den Zellen des Körpers2,3bauen. Die Progression der Atherosklerose beinhaltet die Verdickung und Verhärtung der Arterien durch eine entzündliche Reaktion, unregelmäßige Fettstoffwechsel und Plaquebildung, führt zu Plaque Ruptur und Myokardinfarkt4,5. Endothelzellen express Zytokine und Adhäsion Moleküle, die MCP-1, die an den C-C Chemokin-Rezeptor (CCR2 enthalten) auf der Oberfläche der Monozyten6,7,8bindet. Oxidiertes Cholesterin wandelt Monozyten zu Makrophagen in der frühen Phase der Plaquebildung, die verstärkt der entzündlichen Reaktion in der Region und führt zu Gewebeschäden und die Bildung von Plaques instabil oder gefährdeten9, 10.
Traditionell wird Arteriosklerose durch die Beurteilung der luminalen Stenose durch anatomische Bildgebung mit Ultraschall oder Angiographie11,12ausgewertet. Diese Methoden können aber nur feststellen starke Verengung der Arterienwand und nicht die frühen Stadium der Arteriosklerose, wie erste Gedenktafel Wachstum führt zu arteriellen Umbau um Arterie Größe beizubehalten und blood Flow Rate12,13, 14. Daher underrepresent Angiogramme Atherosklerose Prävalenz. Darüber hinaus nicht-invasive bildgebende Verfahren wie single Photonenemission berechnet Tomographie, Positronen-Emissions-Tomographie und Magnetresonanz-Bildgebung vor kurzem wurden verwendet, um Plaque-Morphologie zu charakterisieren, da sie erste Details bieten können und Charakterisierung von Plaques. Aber diese Modalitäten sind oft begrenzt durch den Mangel an Sensibilität, räumliche Auflösung, oder erfordern die Verwendung von ionisierender Strahlung, machen bildgebende Plaque Fortschreiten in den verschiedenen Phasen viel mehr herausfordernde15,16, 17. Ein bildgebendes System der Lieferung, das speziell Plaques in den verschiedenen Phasen der Atherosklerose identifizieren würde bleibt entwickelt werden.
Nanopartikel haben eine neue Plattform für in Vivo Plaque-targeting und Diagnostik18,19,20,21gezeigt. Insbesondere sind PAMs vorteilhaft aufgrund ihrer chemischen Vielfalt und die Fähigkeit, eine Vielzahl von Moieties, Kompositionen, Größen, Formen und Oberflächen Funktionalisierung22unterbringen. Peptid amphiphilen (PAs) bestehen aus einem hydrophilen, Peptid “Headgroup” befestigt eine hydrophobe Schwanz, die in der Regel sind Lipide; Diese amphiphile Struktur verleiht selbstorganisierende Funktionen und ermöglicht eine multivalente Anzeige der Peptide auf der Oberfläche der Partikel22,23,24. Die Peptid-Headgroups beeinflussen die Kornform durch Falt- und Wasserstoffbindung zwischen Peptide25. Peptide, die durch die Interaktion der β-Blatt Falten haben gezeigt, dass längliche Micellen zu bilden, während α-helikale Bestätigung sowohl sphärische und längliche Micellen22,23,24, bilden kann 25,26,27. Polyethylenglykol (PEG) linker, die Oberflächenladung des Peptids schützen können zwischen der hydrophilen Peptid und das hydrophobe Ende der PAMs, Verbesserung der Verfügbarkeit der Nanopartikel im Körperkreislauf28, platziert werden 29 , 30 , 31. PAMs sind auch vorteilhaft, weil sie biokompatibel sind und gezeigt haben, dass ein breites Spektrum von Anwendungen32,33. Die Wasserlöslichkeit von Micellen bietet einen Vorteil gegenüber anderen Nanopartikel-basierten Systemen wie z. B. bestimmte Polymere Nanopartikel, die nicht in Wasser löslich und in Lösungsvermittler für Injektionen34ausgesetzt werden. Darüber hinaus macht die Fähigkeit zum Erstellen von PAMs, das Zerlegen in Reaktion auf bestimmte Reize PAMs einen attraktive Kandidaten für kontrollierte intrazellulären Droge Lieferung35.
Durch Bindung an den Rezeptor CCR2 und akkumulieren in den Aortenbogen, wurden zuvor PAMs für Monocyte gezielt zur Überwachung der verschiedener Phasen von atherosklerotischen Läsionen in der Aorta9entwickelt. ApoE– / – Mäusen Monocyte Ansammlung steigt proportional zu Plaque Fortschreiten36. Darüber hinaus wurde festgestellt, dass Patienten mit Ruptur neigende, Spätstadium Plaques höhere Mengen an Monozyten37 enthalten. Daher ist die Änderung der PAMs, MCP-1 einzubeziehen nützlich, denn es größere targeting Spezifität und Differenzierung zwischen frühen und späten Stadium atherosklerotische Läsionen ermöglicht. Diese Proof-of-Concept-Studien überprüft auch, ob PAMs sind sicher genug vorklinisch verwendet werden und renally38deaktiviert sind. Da Monozyten und Entzündungen charakteristisch für andere Krankheiten sind, haben MCP-1 PAMs das Potenzial, für therapeutische und diagnostische Anwendungen in anderen Krankheiten über Arteriosklerose8,39,40 verwendet werden , 41.
Hier berichten wir über die Herstellung von hoch skalierbare und selbst-zusammengebauten MCP-1 PAMs, die optimale Größe, Oberflächenladung und gezielte Ausrichtung auf Monozyten für erweiterte imaging-Anwendungen in Atherosklerose des Teilchens nachgewiesen.
MCP-1 PAMs sind eine vielversprechende molekulare Bildgebung-Plattform, bestehend aus einer hydrophilen targeting Peptid und hydrophobe Endstück, das treibt die selbstgebaute Natur der Nanopartikel.
