Dit document stelt een methode waarbij de synthese en de karakterisering van monocyt-targeting peptide Amfifiel micellen en de corresponderende testen om te testen voor biocompatibiliteit en het vermogen van de micel binden aan monocyten.
Atherosclerose is een belangrijke bijdrage aan hart-en vaatziekten, de belangrijkste oorzaak van overlijden wereldwijd, die 17,3 miljoen doden per jaar beweert. Atherosclerose is ook de belangrijkste oorzaak van plotselinge dood en myocardinfarct, aangespoord door unstable plaques die scheuren en occlude van het bloedvat zonder waarschuwing. Huidige imaging modaliteiten kan niet differentiëren tussen stabiele en de instabiele plaques die scheuren. Peptide amfifielen micellen (PAMs) kunnen dit nadeel overwinnen als ze kunnen worden gewijzigd met een verscheidenheid van targeting wordt die specifiek aan zieke weefsels binden. Monocyten zijn gebleken te zijn van vroege markers van atherosclerose, terwijl grote opeenstapeling van monocyten geassocieerd met plaques vatbaar wordt voor scheuren. Vandaar, nanodeeltjes die zijn op monocyten gericht kan kan worden gebruikt om te discrimineren verschillende stadia van atherosclerose. Te dien einde, hier, beschrijven we een protocol voor de bereiding van monocyt-targeting PAMs (monocyte chemoattractant protein-1 (MCP-1) PAMs). MCP-1 PAMs zelf worden geassembleerd door synthese onder milde voorwaarden aan formulier nanodeeltjes van 15 nm in diameter met in de buurt van neutrale oppervlakte lading. In vitro, PAMs bleken te zijn biocompatibel en had een hoge bindende affiniteit voor monocyten. De hierin beschreven methoden tonen belofte voor een brede waaier van toepassingen in atherosclerose, alsmede andere ontstekingsziekten.
Cardiovasculaire aandoeningen moeten nog worden de belangrijkste doodsoorzaken wereldwijd met ongeveer 17,3 miljoen doden wereldwijd1. Cardiovasculaire aandoeningen zijn bijgedragen door atherosclerose, een toestand waarin plaques in de slagaders, waardoor remmende bloed en zuurstof stroom aan de cellen van het lichaam,2,3 opbouwen. De progressie van aderverkalking houdt de verdikking en de verharding van de slagaders door een ontstekingsreactie, onregelmatige lipide metabolisme en plaque-opbouw, leidt tot plaque breuk en myocardiaal infarct4,5. Endotheliale cellen express cytokines en hechting moleculen, waaronder MCP-1, dat zich aan de C-C chemokine receptor (CCR2 bindt) gevonden op het oppervlak van de monocyten6,7,8. Geoxideerde cholesterol omgezet in monocyten macrofagen in de vroege fase van de vorming van de plaque, die de ontstekingsreactie in de regio versterkt en leidt tot weefsel schade en de vorming van onstabiele of kwetsbare plaques9, 10.
Traditioneel, wordt atherosclerose geëvalueerd door de beoordeling van luminal stenose door de anatomische geschikt zijn met behulp van angiografie of echografie11,12. Deze methoden kunnen echter alleen bepalen ernstige vernauwing van de arteriële muur en niet het beginstadium van atherosclerose, als eerste plaque groei arteriële remodelleren veroorzaakt voor het onderhouden van de grootte van de slagader en bloed stroom tarief12,13, 14. Daarom, angiograms underrepresent atherosclerose prevalentie. Bovendien berekend noninvasive beeldvormingstechnieken zoals één foton emissie tomografie, positron emissie tomografie en magnetische resonantie beeldvorming zijn onlangs gebruikt om plaque morfologie karakteriseren als zij eerste details bieden kunnen en karakterisering van plaques. Echter, deze modaliteiten zijn vaak beperkt door het gebrek aan gevoeligheid, ruimtelijke resolutie, of vereisen het gebruik van ioniserende straling, waardoor imaging plaque progressie in verschillende stadia veel meer uitdagende15,16, 17. Een imaging bezorgingssysteem dat specifiek plaques in verschillende stadia van atherosclerose zou identificeren moet nog worden ontwikkeld.
Nanodeeltjes hebben aangetoond een opkomende platform voor in-vivo plaque targeting en diagnostiek18,19,20,21. In het bijzonder zijn PAMs voordelig vanwege hun chemische diversiteit en de mogelijkheid om een verscheidenheid van wordt, composities, maten, vormen en oppervlakte functionalization22tegemoet te komen. Peptide amfifielen (BK) bestaan uit een hydrofiele, peptide “headgroup” gekoppeld aan een hydrofobe staart, die doorgaans lipiden; deze structuur amfifiele verleent zelfassemblerende mogelijkheden en zorgt voor een multivalent weergave van peptiden op het oppervlak van het deeltje22,23,24. De headgroups peptide kan invloed hebben op de vorm van de deeltjes via vouwen en waterstof binding tussen peptiden25. Peptiden die spatel door β-sheet interactie is aangetoond dat het vormen van een verlengde micellen, terwijl α-Helix bevestiging beide bolvormig en verlengde micellen22,23,24, vormen kan 25,26,27. Polyethyleenglycol (PEG) linkers die schild van de oppervlakte lading van de peptide kunnen worden geplaatst tussen de hydrofiele peptide en de hydrofobe staart van PAMs, verbetering van de beschikbaarheid van de nanoparticle in de systemische circulatie28, 29 , 30 , 31. PAMs zijn ook voordelig omdat ze biocompatibel en is aangetoond dat ze hebben een breed scala aan toepassingen32,33. De oplosbaarheid in water van micellen biedt een voordeel ten opzichte van andere nanoparticle-gebaseerde systemen zoals bepaalde polymere nanodeeltjes die niet oplosbaar is in water en moeten worden opgeschort in solubilizers voor injecties34. Bovendien, maakt de mogelijkheid om PAMs die demonteren maken in reactie op een specifieke stimuli PAMs een aantrekkelijke kandidaat voor gecontroleerde intracellulaire drug delivery35.
Door binding aan de receptor CCR2 en zich ophopen in de aortaboog, werden PAMs eerder ontwikkeld voor monocyt gericht op voor het controleren van de verschillende stadia van atherosclerotische laesies in de aorta9. In ApoE– / – muizen, monocyt accumulatie verhoogt proportioneel plaque progressie36. Bovendien bleek dat patiënten met breuk-gevoelig, alsnog plaques hogere hoeveelheden monocyten37 bevatten. De wijziging van PAMs te nemen MCP-1 is daarom nuttig, omdat het zorgt voor meer doelgerichtheid specificiteit en differentiatie tussen atherosclerotische laesies van de vroege – en late-stadium. Deze proof-of-concept studies ook verifiëren dat PAMs zijn veilig genoeg om te worden pre-clinically gebruikt en worden leeggemaakt renally38. Aangezien monocyten en ontsteking karakteristiek voor andere ziekten zijn, hebben MCP-1 PAMs het potentieel om te worden gebruikt voor therapeutische en diagnostische toepassingen in andere ziekten buiten atherosclerose8,39,40 , 41.
Hierin, rapporteren we de fabricage van uiterst schaalbare en zelf gemonteerd MCP-1 PAMs dat aangetoond van het deeltje optimale omvang, oppervlakte lading en selectieve richten aan monocyten voor verbeterde beeldbewerkingstoepassingen in atherosclerose.
MCP-1 PAMs zijn een veelbelovende moleculaire beeldvorming platform, bestaande uit een hydrofiele targeting peptide en hydrofobe staart die de zelf samengestelde aard van de nanoparticle drijft. Deze micel monocyt-targeting kan bereid worden door eenvoudige synthese en zuivering stappen van het MCP-1 peptide en DSPE-PEG (2000)-MCP-1.
