Hier presenteren we een praktische gids voor het bouwen van een geïntegreerde microscopie-systeem, dat wordt samengevoegd conventionele epi-belichting fluorescerende imaging, single-molecuul detectie gebaseerde Super resolutie beeldvorming en Multi-Color single-molecuul detectie, met inbegrip van single-molecuul fluorescentie resonantie energieoverdracht imaging, in één set-up op een kostenefficiënte manier.
Fluorescentie microscopie is een krachtig hulpmiddel om biologische moleculen in situ ontdekken en hun dynamiek en interactie in real-time controleren. Naast conventionele epi-fluorescentie microscopie, zijn verschillende beeldvormende technieken ontwikkeld om specifieke experimentele doelen te bereiken. Enkele van de meest gebruikte technieken omvatten single-molecuul fluorescentie resonantie energieoverdracht (smFRET), die conformationele veranderingen en moleculaire interacties met angstrom resolutie en single-molecuul detectie gebaseerde rapporteren kan Super resolutie (SR) imaging, die de ruimtelijke resolutie van ongeveer tien tot twentyfold in vergelijking met diffractie-limited microscopie verbeteren kan. Hier presenteren we een klant ontworpen geïntegreerd systeem, dat wordt samengevoegd meerdere beeldvormende methoden in een Microscoop, met inbegrip van conventionele epi-belichting fluorescerende imaging, single-molecuul detectie gebaseerde SR imaging en Multi-Color single-molecuul detectie, inclusief smFRET imaging. Verschillende beeldvormende methoden kunnen gemakkelijk en reproducibly worden bereikt door over te schakelen van optische elementen. Deze set-up is eenvoudig te nemen door een onderzoekslaboratorium dat in de biologische wetenschappen met een behoefte aan routine en gevarieerde beeldvorming experimenten op een lagere kosten en ruimte ten opzichte van de bouw van aparte microscopen voor individuele doeleinden.
Fluorescentie microscopen zijn belangrijke hulpmiddelen voor de moderne biologische wetenschap onderzoek en fluorescerende imaging wordt routinematig uitgevoerd in vele biologie laboratoria. Door biomoleculen van belang met fluorophores te labelen, kunnen we direct visualiseren ze onder de Microscoop en de tijd-afhankelijke wijzigingen vastleggen in lokalisatie, conformatie, interactie, en vergadering staat in vivo of in vitro. Conventionele fluorescentie microscopen hebben een ruimtelijke resolutie van diffractie-limited, die daarom ~ 200-300 nm in de laterale richting en ~ 500-700 nm in de axiale richting1,2, en zijn beperkt tot de beeldvorming op de 100s van nanometer-naar-micron schaal. Om te onthullen fijnere details in de moleculaire assembly of organisatie, hebben verschillende SR-microscopies die de diffractie-limiet breken kunnen ontwikkeld. Strategieën gebruikt om SR omvatten niet-lineaire optische effecten, zoals de gestimuleerde emissie uitputting (STED) microscopie3,4 en gestructureerde verlichting microscopie (SIM)5,6, 7, stochastische detectie van afzonderlijke moleculen, zoals stochastische optische wederopbouw microscopie (STORM)8 en photoactivated lokalisatie microscopie (PALM)9, en een combinatie van beide, zoals MINFLUX10. Onder deze microscopies SR, kunnen single-molecuul detectie gebaseerde SR microscopen relatief moeiteloos worden gewijzigd van een single-molecuul Microscoop set-up. Met de activering van de repetitieve en beeldvorming van photoactivatable fluorescente proteïnen (FPs) of foto-schakelbare kleurstoffen getagd op biomoleculen van belang, is ruimtelijke resolutie 10-20 nm11bereikbaar. Om te krijgen informatie over de moleculaire interacties en conformationele is dynamiek in real-time, angstrom-naar-nanometer resolutie vereist. smFRET12,13 is een benadering om deze oplossing te bereiken. In het algemeen, zijn afhankelijk van de biologische kwesties van belang, beeldvormende methoden met verschillende ruimtelijke resoluties nodig.
Meestal is voor elk type voor imaging, specifieke excitatie en/of emissie optische configuratie nodig. Bijvoorbeeld, is een van de meest gebruikte verlichting methoden voor single-molecuul detectie door middel van totale interne reflectie (TIR), waarin een specifieke excitatie hoek moet worden bereikt door middel van een prisma of door middel van het objectief. Voor de detectie van de smFRET moeten emissies van zowel donor als acceptor kleurstoffen worden ruimtelijk gescheiden en gericht op verschillende delen van het elektron-te vermenigvuldigen, charge – coupled apparaat (EMCCD), die kan worden bereikt met een set van spiegels en dichroïde beam splitters Geplaatst in het pad van de emissie. Voor driedimensionale (3-D) is SR imaging, een optische component, zoals een cilindrische lens14, nodig om een astigmatisme effect te veroorzaken in het pad van de emissie. Daarom, EIGENBOUW of verkrijgbare geïntegreerde microscopen, meestal, functioneel gespecialiseerd zijn voor elk type van imaging methode en zijn niet flexibel om te schakelen tussen verschillende beeldvormende methoden op de zelfde set-up. Hier presenteren we een kosteneffectieve, hybridesysteem waarmee verstelbaar en reproduceerbare Hiermee schakelt u tussen drie verschillende beeldvormende methoden: conventionele epi-belichting fluorescerende beeldbewerking met diffractie-beperkte resolutie, single-molecuul detectie gebaseerde SR beeldbewerking en Multi-Color single-molecuul detectie, met inbegrip van smFRET imaging (figuur 1A). In het bijzonder de hier gepresenteerde set-up bevat vezel-gekoppelde ingang lasers voor Multi-Color excitatie en een commerciële verlichting arm in het pad van excitatie, waardoor geprogrammeerd controle van de excitatie-hoek, om te schakelen tussen epi-modus en TIR. In het pad van de emissie, een verwisselbare EIGENBOUW cilindrische lens cassette is geplaatst binnen het lichaam van de Microscoop voor 3D-SR imaging en een commerciële balk splitter is geplaatst voor een EMCCD-camera die kan selectief worden ingeschakeld om te detecteren van meerdere kanalen van de emissie tegelijkertijd.
Deze hybride Microscoop systeem elimineert de noodzaak voor de aankoop van meerdere microscopen. De totale kosten voor alle onderdelen, inclusief de optische tabel, tabel installatie arbeid, software en werkstation, is ongeveer 230.000 dollar.
