Bruker flertrinns kultur systemer, rapportere vi en i vitro B-celle til plasma celle differensiering modell.
Plasma celler (stk.) skiller ut store mengder av antistoffer og utvikle fra B-celler som har blitt aktivert. PCer er sjeldne celler finnes i benmarg eller mucosa og sikre humoral immunitet. Deres lav frekvens og plassering, studiet av PCer er vanskelig i menneskelig. Vi rapporterte en B til PC i vitro differensiering modellen med valgte kombinasjoner av cytokiner og aktivisering molekyler som kunne reprodusere sekvensiell celledifferensiering forekommer i vivo. I denne i vitro modellen, minne B-celler (MBCs) vil skille ut før plasmablasts (prePBs), plasmablasts (PBs), tidlig PCer og til slutt i varige nær PCer, med en fenotypen sine kolleger i friske individer. Vi har også bygget en åpen tilgang Bioinformatikk verktøy for å analysere den mest fremtredende informasjonen fra GEP data relatert til PC differensiering. Disse ressursene kan brukes til å studere human B til PC differensiering og i denne studien, vi undersøkt genet uttrykk regulering av epigenetic faktorer i human B til PC differensiering.
Differensiering av B-celler til plasma celler (stk.) er avgjørende for humoral immunitet og beskytte verten mot infeksjoner1. B å PC differensiering er forbundet med store endringer i transkripsjon kapasitet og metabolisme å imøtekomme til antistoff sekret. Transkripsjonsfaktorer som kontrollerer B til PC differensiering har vært grundig studert og avdekket eksklusive nettverk inkludert B – og PC-spesifikke transkripsjon faktorer (TFs)2. B celler er PAX5, BCL6 og BACH2 TFs voktere av B celle identitet2,3. Induksjon av IRF4, PRDM1 koding BLIMP1 og XBP1 PC TF vil slukke B celle gener og indusere en koordinert antistoff-sekresjon celle transcriptional programmet3,4,5. Koordinerte transcriptional endringene er forbundet med Ig gener transkripsjon aktivisering med en bryter fra skjemaet membran-bundet til secreted form av immunglobulin tunge kjeden2,3, 4. B til PC differensiering er knyttet til induksjon i gener involvert i endoplasmatiske retikulum og Golgi apparatet fungerer samtidig med oppslåtte protein svar (UPR) aktivisering kjent for å spille en nøkkelrolle i PC ved imøtekommende syntesen av utskilles immunglobuliner6,7. TF-XBP1 spiller en viktig rolle i denne mobilnettet tilpasning8,9,10.
B-celler og PCer er sentrale aktører i humoral immunitet. Forstå biologiske prosesser som kontrollerer produksjonen og overlevelse av normale plasma celler er kritisk i terapeutiske tiltak som skal sikre effektiv immunreaksjoner og forhindre autoimmunitet eller uimottakelig mangel. PC er sjeldne celler differensiering-tidligfasen som finner sted i anatomisk steder som hemmer full biologiske karakterisering, spesielt i menneskelig. Bruker flertrinns kultur systemer, har vi rapportert en i vitro B PC differensiering modellen. Denne modellen reproduserer den sekvensielle celledifferensiering og modning forekommer i forskjellige organer i vivo11,12,13. I et første skritt, minne B-celler er først aktivert for fire dager av CD40 ligand, oligodeoxynucleotides og cytokin kombinasjon og differensiere i preplasmablasts (PrePBs). Under det andre trinnet hjulpet preplasmablasts til å skille ut plasmablasts (PBs) ved å fjerne CD40L og oligodeoxynucleotides stimulering og endre cytokin kombinasjonen. I en tredje trinnet er plasmablasts overtalt til å skille ut tidlig PCer ved å endre de cytokin kombinasjon11,12. En fjerde trinnet ble innført for å få fullt ut modne PCer ved dyrking disse tidlige PCs med benmarg stromal celler betinget medium eller valgt vekstfaktorer13. Disse eldre PCer kan overleve flere måneder i vitro og skiller ut store mengder immunglobulin (figur 1). Interessant, viser vår i vitro modell koordinerte transcriptional endringene og fenotypen av ulike B til PC stadier som kan være oppdaget i vivo11,12,13,14 ,15. PCer er sjeldne cellene våre i vitro differensiering modellen kan studere human B til PC differensiering.
I menneskelige, PC er sjeldne celler differensiering stadier foregår anatomiske steder som hemmer full biologiske karakterisering. Vi har utviklet et i vitro B PC differensiering modellen bruker flertrinns kultur systemer der ulike kombinasjoner av aktivisering molekyler og cytokiner brukes deretter for å reprodusere sekvensiell celledifferensiering forekommer i den forskjellige organer/vev i vivo11,12,13.
