Multiplex in situ hybridization (ISH) blev ansat til samtidig at visualisere udskrifter for to G protein-koblede receptorer og en transskription faktor i hele vagal ganglionic kompleks af den voksne mus. Denne protokol kan bruges til at generere nøjagtige kort over de transskriptionelle profiler af vagale afferent neuroner.
Denne undersøgelse beskriver en protokol for multiplex in situ hybridisering (ISH) af musen jugular-nodose ganglier, med særlig vægt på at opdage udtrykket af G protein-koblede receptorer (GPCRs). Formalin-faste jugular-nodose ganglier blev behandlet med RNAscope teknologi til samtidig at opdage udtrykket af to repræsentative GPCRs (cholecystokinin og ghrelin receptorer) i kombination med en markør gen af enten nodose (parret-lignende homeobox 2b, Phox2b) eller halspulsåren afferent neuroner (PR domæne zink finger protein 12, Prdm12). Mærket ganglier blev afbildet ved hjælp af konfokal mikroskopi til at bestemme fordelingen og udtryk mønstre af de ovennævnte udskrifter. Kort, Phox2b afferent neuroner blev fundet at rigeligt udtrykke cholecystokinin receptor (Cck1r), men ikke ghrelin receptor (Ghsr). En lille delmængde af Prdm12 afferent neuroner blev også fundet at udtrykke Ghsr og / eller Cck1r. Potentielle tekniske forbehold i design, forarbejdning og fortolkning af multiplex ISH diskuteres. Den fremgangsmåde, der er beskrevet i denne artikel, kan hjælpe forskere med at generere nøjagtige kort over transskriptionsprofilerne for vagale afferent neuroner.
Cellekroppe af vagale afferents er indeholdt i jugular, petrosal, og nodose ganglier1,2,3. Deres axoner rejser sammen via flere grene af vagusnerven til craniocervical, thorax og abdominal territorier4,5,6,7. Fra deres viscerale slutninger kan vagale afferents reagere på en bred vifte af fysiologiske og skadelige stimuli8,9,10. Imidlertid er fordelingen af signalmolekyler og receptorer, der er involveret i vagal sensing, stadig dårligt karakteriseret. Dette skyldes til dels, at vagal ganglier, på trods af deres lille størrelse, udtrykker et bredt spektrum af receptorer, herunder et stort antal GPCRs8,11,12,13. Desuden er vagale afferent neuroner i sagens natur heterogene og viser forskellige molekylære profiler14. For at komplicere sagen er halspulsåren, petrosal og nodose ganglier fastgjort i musen og danner derved en enkelt ganglionic masse. Endelig er nodose ganglion i en delmængde af dyr knyttet til den sympatiske overlegne cervikal ganglion15.
Tidligere har efterforskere henvendt sig til immunohistochemistry for at studere den neurokemiske make-up af vagale afferent neuroner16,17,18. Mens immunohistochemistry ved hjælp af validerede antistoffer er nyttige, skal resultaterne af immunohistokemiske undersøgelser fortolkes med forsigtighed. For eksempel har talrige bestræbelser på at identificere specifikke antistoffer mod GPCR undladt19,20,21,22,23,24,25, førende efterforskere til at konkludere, at de fleste antistoffer mod GPCR er upålidelige. For at omgå disse spørgsmål er kvantitativ PCR (qPCR) blevet meget brugt til at vurdere genekspression i gnaverens vagale ganglionic masse26,27,28,29. Men, undersøgelse genekspression ved hjælp af qPCR sker på bekostning af et tab af rumlige oplysninger. Det kan især ikke forudsiges, hvor mange celler eller hvilken eller flere celletyper der udtrykker et bestemt interessegen (f.eks. nodose vs. halspulsårer). Tilbagevendende spørgsmål omfatter også forurening med tilstødende væv og inddragelse af variable længder af vagus nerve, overlegen cervikal, og halspulsåren ganglier under dissektion15. Som et resultat af ovenstående vanskeligheder, kontroverser omgiver udtryk og distribution af flere GPCRs i vagal afferent neuroner. Et særligt gådefuldt eksempel vedrører ghrelin receptor (Ghsr). Mens nogle undersøgelser har fundet udbredt udtryk for denne receptor i vagal afferent neuroner30,31,32, andre har fundet Ghsr mRNA at være næsten målbart i nodose ganglion11,14. En detaljeret kortlægning af Ghsr mRNA i den vagale ganglionic masse er derfor berettiget.
In situ hybridisering (ISH) er også blevet brugt til at vurdere genekspressionsmønstre i vagal ganglionic masse7,11,12,33,34,35. Fordi RNA-baserede teknikker forbliver mere pålidelige og specifikke end antistofbaserede teknikker under de fleste omstændigheder36,37, har ISH-undersøgelser vist sig værdifulde for bedre forståelse af den neurokemiske kodning af vagale afferent neuroner. Ikke desto mindre er traditionelle ISH-teknikker i sig selv ikke uden forbehold. Radioaktiv ish er følsom, men genererer baggrund og forbliver besværlig38. Ikke-radioaktiv ish er mindre kompliceret, men også mindre følsom38. I modsætning hertil er den nyligt udviklede RNAscope ISH-metode meget følsom og genererer minimal baggrund39. Den nuværende undersøgelse anvendt multiplex fluorescerende RNAscope til påvisning af GPCRs i vagal afferent neuroner af musen. Vi fokuserede på at kortlægge fordelingen af Ghsr og sammenlignede dens fordeling med fordelingen af cholecystokinin receptor (Cck1r), en anden GPCR kendt for at være udtrykt i nodose ganglion34. Endelig blev de to transskriptionsfaktorer, parret-lignende homeobox 2b (Phox2b) og PR domæne zink finger protein 12 (Prdm12), brugt som selektive markører for nodose og jugular afferent neuroner, henholdsvis14. Uden at visualisere Phox2b eller Prdm12, ville det være udfordrende at identificere jugular vs nodose afferents med sikkerhed. Potentielle tekniske faldgruber diskuteres også i hele artiklen.
Teknikken med ISH blev opfundet i slutningen af 1960’erne42. Det er dog først i midten af 1980’erne , at det blev ansøgt om påvisning af mRNAs i de centrale og periferenervesystemer 43,44. I betragtning af nervesystemets heterogenitet og tilbagevendende problemer med antistoffer forbliver lokalisering af en bestemt udskrift på cellulært niveau et uvurderligt værktøj. Ikke desto mindre er traditionelle ISH-metoder forblevet besværli…
