Att främja studien av preantral follikulogenes kräver effektiva metoder för follikelisolering från enstaka äggstockar. Här presenteras ett strömlinjeformat, mekaniskt protokoll för follikelisolering från nötkreatur äggstockar med hjälp av en vävnadshackare och homogenisator. Denna metod möjliggör insamling av ett stort antal livskraftiga preantrala folliklar från en enda äggstock.
Att förstå hela processen för däggdjursfollikulogenes är avgörande för att förbättra assisterad reproduktionsteknik hos boskap, människor och hotade arter. Forskningen har mestadels begränsats till antrala och stora preantrala folliklar på grund av svårigheter att isolera mindre preantrala folliklar, särskilt hos stora däggdjur som nötkreatur. Detta arbete presenterar ett effektivt tillvägagångssätt för att hämta ett stort antal små preantrala folliklar från en enda bovin äggstock. Cortex hos enskilda nötkreaturs äggstockar skivades i 500 μm kuber med hjälp av en vävnadshackare och homogeniserades i 6 minuter vid 9 000-11 000 rpm med en 10 mm sond. Stora skräp separerades från homogenatet med hjälp av en ostduk, följt av seriell filtrering genom 300 μm och 40 μm cellsilar. Innehållet som fanns kvar i 40 μm-silen sköljdes i en sökskål, där folliklar identifierades och samlades in i en droppe medium. Livskraften hos de uppsamlade folliklarna testades via trypanblå färgning. Denna metod möjliggör isolering av ett stort antal livskraftiga små preantrala folliklar från en enda nötkreaturs äggstock på cirka 90 minuter. Det är viktigt att denna metod är helt mekanisk och undviker användning av enzymer för att dissociera vävnaden, vilket kan skada folliklarna. Folliklarna erhållna med användning av detta protokoll kan användas för nedströms applikationer såsom isolering av RNA för RT-qPCR, immunolokalisering av specifika proteiner och in vitro-odling .
Äggstocksfolliklar är de funktionella enheterna i äggstocken, som ansvarar för produktionen av könscellen (äggcellen) samt hormoner som är kritiska för reproduktiv funktion och allmän hälsa. Primordiala folliklar bildas i äggstocken under fostrets utveckling eller under neonatalperioden beroende på art1, och de utgör en kvinnas äggstocksreserv. Follikulär tillväxt börjar med aktivering av primordiala folliklar som lämnar vilopoolen och går in i odlingsfasen. Preantral follikulogenes, som omfattar alla follikelstadier före antrumutveckling, är en mycket dynamisk process som kräver synkrona morfologiska och metaboliska förändringar i äggcellen och de omgivande granulosacellerna, drivna av tät kommunikation mellan dessa två celltyper 2,3. Preantrala folliklar utgör majoriteten av de follikulära enheter som finns i äggstocken vid varje given tidpunkt4. Utveckling genom de preantrala stadierna av follikulogenes uppskattas vara flera veckor längre än antral utveckling 5,6, och denna tid är nödvändig för att oocyten och somatiska celler ska få tillräcklig mognad för att gå in i det sista utvecklingsstadiet (dvs. antralstadiet) och förbereda sig för ägglossning, befruktning och embryonal utveckling 7,8,9.
Mycket av den nuvarande kunskapen om ovariell preantral follikulogenes kommer från musmodeller10,11,12,13, delvis på grund av lättheten att återhämta ett stort antal av dessa folliklar från en mindre och mindre fibrös äggstock. Även om rapporter om isolering av ett stort antal preantrala folliklar från nötkreaturs äggstockar går tillbaka cirka 30 år14, har en mer fullständig förståelse för de processer som reglerar utvecklingen av dessa folliklar i tidigt stadium förblivit orealiserad, till stor del på grund av bristen på optimerade, effektiva och repeterbara metoder för att hämta tillräckligt antal livskraftiga preantrala folliklar, särskilt i tidiga utvecklingsstadier. Med det ökande intresset för att bevara äggstocksreserven för framtida användning vid assisterad befruktning hos människor blir kor en attraktiv modell på grund av deras mer liknande äggstocksstruktur15. Den bovina äggstocken är dock markant rikare på kollagen jämfört med musens äggstock16, vilket gör mekanisk isolering med metoder som beskrivs för musen mycket ineffektiva. Ansträngningar för att utöka fertilitetsbevarande tekniker inkluderar fullständig in vitro-tillväxt av preantrala folliklar till antralstadiet, följt av in vitro-mognad (IVM) av de slutna oocyterna, in vitro-befruktning (IVF) och embryoproduktion och överföring17. Hittills har hela denna process endast uppnåtts hos möss18. Hos nötkreatur är framstegen mot follikeltillväxt in vitro begränsad till några rapporter med varierande follikelstadier i början av kulturen, liksom varierande kulturlängd mellan protokoll17,19.
De metoder som beskrivs i litteraturen för skörd av preantrala folliklar från nötkreaturs äggstock har mestadels använt mekaniska och enzymatiska tekniker, antingen isolerade eller i kombination 2,14,17,20. Den första rapporten från ett protokoll för isolering av bovin preantral follikel använde en vävnadshomogenisator och seriell filtrering för att bearbeta hela äggstockarna20. Denna studie följdes av rapporter som kombinerar mekaniska och enzymatiska procedurer som använde kollagenas14. Ett återkommande tema när man använder kollagenas för att smälta äggstocksvävnaden är den potentiella risken för skador på det follikulära källarmembranet, vilket kan äventyra follikelns livskraft 14,21,22,23. Därför har olika kombinationer av mekaniska metoder använts, såsom användning av en vävnadshackare och upprepad pipettering eller en vävnadshackare i kombination med homogenisering20,24,25,26. En annan mekanisk teknik som har beskrivits använder nålar för att dissekera preantrala folliklar direkt från äggstocksvävnaden, vilket är särskilt användbart för att isolera större (>200 μm) sekundära folliklar. Denna process är dock tidskrävande, ineffektiv för att isolera mindre preantrala folliklar och är kompetensberoende vid försök i nötkreaturs äggstockar 19,27,28.
Genom att dra nytta av de olika teknikerna som beskrivs i litteraturen syftade detta protokoll till att optimera isoleringen av preantrala folliklar från enstaka nötkreatur äggstockar på ett enkelt, konsekvent och effektivt sätt som undviker inkubation i enzymatiska lösningar. Förbättring av metoderna för att isolera preantrala folliklar kommer att ge möjlighet att förbättra förståelsen för detta stadium av follikulogenes och möjliggöra utveckling av effektiva odlingssystem för att utveckla preantrala folliklar till antralstadiet. De detaljerade förfaranden som beskrivs häri för isolering av preantrala folliklar från ett stort däggdjur som nötkreatur kommer att vara avgörande för forskare som syftar till att studera tidig follikulogenes hos en icke-murin art som kan översättas till människor.
Detta protokoll beskriver en reproducerbar metod för att hämta preantrala folliklar i tidigt stadium, särskilt i primära och tidiga sekundära stadier, från bovin äggstock.
