Verarbeitung von Bronchoalveolar Lavage Fluid und Matched Blood für Alveolar Macrophage und CD4+ T-Zell Immunophenotypisierung und HIV Reservoir Assessment
Wir beschreiben eine Methode zur Verarbeitung von bronchoalveolarer Spülflüssigkeit und abgestimmtem peripherem Blut von chronisch HIV-infizierten Personen auf antiretrovirale Therapie zur Beurteilung von lungenen HIV-Reservoirs. Diese Methoden führen zum Erwerb von hochreinen CD4-T-Zellen und Alveolar-Makrophagen, die anschließend zur Immunphenotypisierung und HIV-DNA/RNA-Quantifizierungen durch ultraempfindliche Polymerase-Kettenreaktion eingesetzt werden können.
