Protein co-expression is a powerful alternative to the reconstitution in vitro of protein complexes, and is of help in performing biochemical and genetic tests in vivo. Here we report on the use of protein co-expression in Escherichia coli to obtain protein complexes, and to tune the mutation frequency of cells.
We report here that the expression of protein complexes in vivo in Escherichia coli can be more convenient than traditional reconstitution experiments in vitro. In particular, we show that the poor solubility of Escherichia coli DNA polymerase III ε subunit (featuring 3’-5’ exonuclease activity) is highly improved when the same protein is co-expressed with the α and θ subunits (featuring DNA polymerase activity and stabilizing ε, respectively). We also show that protein co-expression in E. coli can be used to efficiently test the competence of subunits from different bacterial species to associate in a functional protein complex. We indeed show that the α subunit of Deinococcus radiodurans DNA polymerase III can be co-expressed in vivo with the ε subunit of E. coli. In addition, we report on the use of protein co-expression to modulate mutation frequency in E. coli. By expressing the wild-type ε subunit under the control of the araBAD promoter (arabinose-inducible), and co-expressing the mutagenic D12A variant of the same protein, under the control of the lac promoter (inducible by isopropyl-thio-β-D-galactopyranoside, IPTG), we were able to alter the E. coli mutation frequency using appropriate concentrations of the inducers arabinose and IPTG. Finally, we discuss recent advances and future challenges of protein co-expression in E. coli.
De invoering van overexpressie technologieën gestimuleerd ofwel de biochemische studies van laag kopie-aantal enzymen en de industriële productie van farmacologisch werkzame eiwitten (bijvoorbeeld insuline). Sinds de komst van deze technologieën, aanzienlijke vooruitgang geboekt om de opbrengst en kwaliteit van recombinante eiwitten verhogen. Daarnaast werden prokaryotische 1,2 en eukaryotische 3 overexpressie systemen ontwikkeld door de jaren heen, het aanbieden van bruikbare alternatieven voor het "werkpaard" van eiwitten biotechnologie, dat wil zeggen, Escherichia coli. Vooral de beschikbaarheid van alternatieve platforms E. coli leidde tot de productie van recombinante peptiden of proteïnen dragen post-translationele modificaties. Er moet echter worden opgemerkt dat E. coli is nog het organisme gekozen voor recombinante eiwitproductie. Dit komt door verschillende factoren, waaronder de meest relevant kan zijnbeschouwd: i) de beschikbaarheid van een groot aantal overexpressie systemen (expressievectoren en stammen) voor E. coli 1,2; ii) de korte generatietijden en hoge opbrengsten biomassa, E. coli in een verscheidenheid van rijke en synthetische media; iii) de gemakkelijke manipulatie hetzij op biochemisch en op genetisch niveau van dit micro-organisme; iv) het isoleren van stammen in staat de productie van toxische eiwitten 4; v) de bouw van de stammen met homogene inductie bij de bevolking niveau 5,6. Bovendien is recent aangetoond dat expressie die geschikt zijn voor de productie in E. coli van post-translationeel gemodificeerde eiwitten kunnen worden bedacht en gebouwd 2.
Momenteel wordt eiwitoverexpressie vooral gebruikt om monomere of homo-oligomere eiwitten waarvan hypersynthesis kan worden uitgevoerd met een enkel gen gekloneerd in een geschikte plasmide te verkrijgen. Echter, aandacht werd onlangsbesteed aan de bouw van E. coli eiwit co-expressiesystemen, tegen de productie, in vivo, van hetero-oligomere complexen 2. Interessant is dat vroege experimenten van eiwit co-expressie gericht de inter-species assemblage van grote en kleine subeenheden van cyanobacteriën ribulose-1,5-bisfosfaatcarboxylase / oxygenase 7,8, en de vereniging van afgeknotte en full-length vormen van HIV-1 reverse transcriptase 9. Deze baanbrekende studies aangetoond dat eiwit co-expressie is een krachtig alternatief voor traditionele in vitro reconstitutie. Bovendien eiwit co-expressie in E. coli werd gebruikt om verschillende eiwitten dragen posttranslationele modificaties 2 produceren, eiwitten bevattende natuurlijke aminozuren 2 verkrijgen, en om de opbrengst van overexpressie membraaneiwitten 2 verhogen. Bovendien is het potentieel van proteïne co-expressie als een middel te verlenen aan E. coli concurrerennvu in eiwituitscheiding is onder actieve onderzoek 2.
Twee belangrijke strategieën van eiwit co-expressie in E. coli kan worden nagestreefd: i) het gebruik van een enkel plasmide op de verschillende genen gastheer voor overexpressie; ii) het gebruik van meerdere plasmiden in enkele cellen co-expressie van de doelwit eiwitten. In het eerste geval zijn de criteria voor de keuze van plasmide niet van die van conventionele enkelvoudige eiwitoverexpressie experimenten, hoewel name plasmiden die tandem promoter / operator elementen zijn geconstrueerd voor co-expressie 10. Deze eerste benadering is derhalve vrij eenvoudig. Er moet echter worden opgemerkt dat het gebruik van een enkel plasmide co-expressie verschillende eiwitten voor twee belangrijke problemen: i) de molecuulmassa van de vector toeneemt met het aantal gastheer genen, waardoor het aantal co-expressie gebrachte eiwitten; ii) wanneer meerdere genen worden gekloneerd onder de controle van een enkele promoter, polariteit decrease de expressie van de genen distaal van de promoter. Het gebruik van dubbele of meervoudige plasmiden in enkele E. coli-cellen heeft om de verenigbaarheid van de vectoren van keuze te bereiken, dus het opleggen van beperkingen aan de in aanmerking komende combinaties van plasmiden. Echter, deze tweede co-expressie strategie biedt het voordeel dat met de molecuulmassa van vectoren, en beperkt polariteit. We hebben onlangs gebouwde een eiwit co-expressie systeem ontworpen om het pendelen van genen tussen de co-expressie plasmiden 11 te vergemakkelijken. In het bijzonder hebben we de pGOOD vectoren serie geconstrueerd, de relevante eigenschappen daarvan zijn: i) een p15A oorsprong van replicatie, de verenigbaarheid van de pGOOD plasmiden met de commerciële vectoren die de ColE1 oorsprong (bijvoorbeeld de pBAD serie 12) te verschaffen; ii) een tetracycline-resistentie cassette; iii) de aanwezigheid van lac -afgeleide regulerende elementen, dat wil zeggen, de promotor-Operator (O 1) paar en het lad </em> q-gen. Met behulp van een geschikte pBAD-pGOOD echtpaar, waren we in staat om de katalytische kern van E. overexpressie coli DNA polymerase III, samengesteld uit drie verschillende subeenheden, namelijk α (de 5'-3 'polymerase), ε (de 3'-5' exonuclease) en θ (stabiliserende ε) 13. In het bijzonder hebben we aangetoond dat de co-expressie van het αεθ complex strikt afhankelijk van de toevoeging aan de E. coli kweekmedium van zowel IPTG en arabinose, wat leidde tot overexpressie van pGOOD en pBAD respectievelijk (Figuur 1A).
In dit rapport, illustreren we hoe eiwit co-expressie efficiënt kunnen oplossen problemen in verband met de slechte oplosbaarheid van een eiwitcomplex subunit. Verder tonen we hoe in vivo eiwit complementatie tests kunnen worden uitgevoerd, en tenslotte beschrijven we het gebruik van eiwit co-expressie afstemmen mutatiefrequentie in E. coli. Om dit doel, gebruikten we pGOOD-pBAD koppels geschikt om relevante voorbeelden van elke casus illustreren.
Eiwitten kunnen intrinsiek ontvouwen, die gebieden waarvan de tertiaire structuur is niet beperkt tot een beperkt aantal conformaties 25. Deze ontvouwen eiwitten zijn meestal gevoelig voor aggregatie 25, en hun isolatie en karakterisering misschien een moeilijke taak te vertegenwoordigen.
