Here, a method that enables quick, efficient, and inexpensive preparation of polyacrylamide gels in a multiwell plate format is described. The method does not require any specialized equipment and could be easily adopted by any research laboratory. It would be particularly useful in research focused on understanding stiffness-dependent cell responses.
Currently, most of the in vitro cell research is performed on rigid tissue culture polystyrene (~1 GPa), while most cells in the body are attached to a matrix that is elastic and much softer (0.1 – 100 kPa). Since such stiffness mismatch greatly affects cell responses, there is a strong interest in developing hydrogel materials that span a wide range of stiffness to serve as cell substrates. Polyacrylamide gels, which are inexpensive and cover the stiffness range of all soft tissues in the body, are the hydrogel of choice for many research groups. However, polyacrylamide gel preparation is lengthy, tedious, and only suitable for small batches. Here, we describe an assay which by utilizing a permanent flexible plastic film as a structural support for the gels, enables the preparation of polyacrylamide gels in a multiwell plate format. The technique is faster, more efficient, and less costly than current methods and permits the preparation of gels of custom sizes not otherwise available. As it doesn’t require any specialized equipment, the method could be easily adopted by any research laboratory and would be particularly useful in research focused on understanding stiffness-dependent cell responses.
De flesta vävnader i kroppen är mjuka viskoelastiska material med en Youngs modul som sträcker sig från 0,1 kPa för hjärnan till 100 kPa för mjukt brosk, men ändå, de flesta in vitro cellforskning bedrivs på vävnadskulturpolystyren (TCP) som har en modul av ~ 1 GPa . 1 Denna stelhet obalans påverkar i hög grad hur celler reagerar på sin omgivning. Systematiska, flerdimensionella studier om effekten av substrat styvhet för en rad celltyper eller miljöförhållanden är sällsynta. 12
Flera lovande hög kapacitet hydrogel teknik har utvecklats, bland annat PEG-baserade microarrays, 13 mikrofluidikanordningar för produktion av agaros hydrogel mikropärlor, 14 eller mikro och nanostavar där styvhet moduleras av diametern och höjden på microrods. 15 Dock , den teknik för att förbereda sådana substrat är sofistikerade och tillgängliga för ett begränsat antal laboratorier. Mycket forskning involverar styvhet modulerad cellsvar utnyttjar polyakrylamid (PA) geler som inte bara är billig och enkel att genomföra, men uppvisar också ett fysiologiskt relevant utbud av Youngs modul, nämligen 0,3 -. 300 kPa 16-22 Men befintliga metoder för att tillverka PA geler för cellodling är arbetskrävande och därmed prepared i små satser. Några av de svårigheter som är förknippade med beredningen av PA geler som cellsubstrat härrör från kravet att gelerna måste vara beredda: 1) i frånvaro av syre för att möjliggöra fullständig polymerisation, 2) med en plan och jämn yta för att möjliggöra en enhetlig cell fastsättning och spridning, och 3) som är permanent fäst på botten av cellodlingsskål för att förhindra flytande.
Flera grupper har försökt att producera PA geler för cellodling i stora serier. Semler et al., Beredda tjocka ark av PA-geler vilka därefter "cut" med en hålstans och placerades i 96-brunnsplattor. 23 Emellertid är denna metod begränsad till styvare geler, dvs,> 1 kPa i Youngs modul, eftersom mjukare geler är "klibbiga", svårt att skära, och skadas lätt. Mih et al. Utvecklat en mer sofistikerad teknik som tillåter gelema att bli direkt polymeriseras i en glasbottnad flerkällsplatta. <supp> 6 Detta uppnåddes genom att hälla gellösningarna i funktionglasbottnade plattor och bildar geler med "sandwich" dem med en egen coverglass array. 6 Även om mycket lovande, små kanteffekter observerades fortfarande med denna teknik. Detta dokument beskriver ett enkelt och billigt sätt att montera PA geler i en flerkällsplatta som lätt kan antas av alla laboratorier. Här är en flexibel plaststöd utnyttjas, som har två sidor – en hydrofob en, som är motbjudande till PA geler, och en hydrofil en, som kovalent binder PA gel vid deponering. När PA gel ark deponeras och permanent fäst på flexibla plaststöd, möjliggör det hanterar geler av valfri tjocklek eller styvhet och skär dem i någon önskad form. Denna ca.oach inte bara producerar anpassade plast 'täck' i storlekar som inte på annat sätt är kommersiellt tillgängliga, utan även undanröjer behovet av att förbehandla glasytor, antingen glas täckglas eller brunnarna i kostsamma glasbottnade flerbrunnsplattor, med ett PA-bindande lösning, som är en mödosam och tidskrävande steg. Slutligen kan enhetliga PA geler ark framställas i stora satser och lagras de-hydrerad under flera månader.
Sammanfattningsvis analysen presenteras här är en förbättring jämfört med befintliga metoder i flera aspekter. Först, är processen för flerkällsplatta aggregatet effektiv, och den totala kostnaden för de material som behövs är låg. För det andra är de hydrogeler framställs i stora satser i en enda homogen gelfilm. Slutligen är endast material som är kommersiellt tillgängliga som krävs. Nyttan av analysen illustreras genom att utforska effekten av substrat styvhet på cellmorfologi och spridningsareal.
