This protocol describes the use of excised intestinal tissue preparations or “intestinal sacs” as an ex vivo model of intestinal barrier function. This model may be used to assess integrity of both the epithelial barrier and the mucous gel layer at specific intestinal sites in animal models of digestive disease.
De epitheliale barrière de eerste aangeboren verdediging van het maagdarmkanaal en selectief regelt vervoer van het lumen aan het onderliggende weefsel compartimenten, beperkt het transport van kleine moleculen door het epitheel en bijna volledig verbod epitheliale macromoleculaire transport. Deze selectiviteit wordt bepaald door het slijmvlies gellaag, die het transport van lipofiele moleculen en zowel de apicale receptoren en strak junctional eiwitcomplexen van het epitheel beperkt. In vitro celcultuur modellen van het epitheel zijn handig, maar als voorbeeld, ze missen de complexiteit van de interacties tussen de microbiota, slijm-gel, epitheel en het immuunsysteem. Anderzijds kan in vivo evaluatie van intestinale absorptie of permeabiliteit worden uitgevoerd, maar deze assays meten totale gastrointestinale absorptie, zonder vermelding van plaatsgebondenheid. Ex vivo permeabiliteit assays met "intestinale sacs"; een snelle en gevoelige werkwijze voor het meten ofwel globaal intestinale integriteit of vergelijkende transport van een specifiek molecuul, met het extra voordeel van intestinale plaatsgebondenheid. Hier beschrijven we de bereiding van intestinale zakjes op doorlatendheid studies en de berekening van de schijnbare permeabiliteit (P app) van een molecuul in de darmwand. Deze techniek kan worden gebruikt als een methode voor de absorptie of regionale epitheliale barrière disfunctie in dierlijke modellen van gastro-intestinale aandoeningen te onderzoeken.
De intestinale epitheliale barrière van het maagdarmkanaal is een mucosaal oppervlak geraamd op 400 m 2 in de volwassen mens. Derhalve wordt voortdurend blootgesteld aan uitdaging van microben, opgenomen geneesmiddelen, nutriënten en bacteriële toxines. De host moet niet alleen onderscheid maken tussen het aanvaardbare commensale bacteriën en potentiële pathogenen, maar beletten dat deze soorten en hun uitgescheiden moleculen die de epitheliale barrière, terwijl tegelijkertijd waardoor opname van voedingsstoffen. Zo is de rol van het darmepitheel is om als een selectieve barrière voor het luminale inhoud 1. Dit gebeurt in hoofdzaak door de epitheliale aangeboren afweersysteem op het slijmvlies, die door een reagerend biologisch systeem bestaande uit constitutieve en induceerbare mechanismen 2 optreedt.
Verlies van epitheliale barrièrefunctie een pathologie die kenmerkend is voor een aantal gastro-intestinale ziekten. In vivoonderzoek van epitheliale barrièrefunctie kan worden beoordeeld door middel van orale sondevoeding van een tracer molecule en daaropvolgende serum analyse 3. Deze techniek geeft geen indicatie van de plaats van barrière disfunctie. In vitro en ex vivo bepaling van transepithele weerstand gebruik Transwell systemen 3 respectievelijk Ussing kamers 4,5, worden gewoonlijk gebruikt als surrogaat markers van epitheliale barrièrefunctie, maar missen de bijdragende ziekte fysiologie van diermodellen 6. In dit protocol beschrijven we een ex vivo model weefselbereiding die direct en gelokaliseerde evaluatie van intestinale integriteit en die kunnen worden gebruikt om mucosale barrièrefunctie beoordeeld op meerdere niveaus mogelijk maakt. Belangrijk is dat deze techniek worden toegepast op diermodellen voor de ziekte, of kan farmacologisch worden gemanipuleerd om grondig afvragen van mucosale barrière disfunctie.
Hier hebben we de isolatie en bereiding van intestinale zakjes gedetailleerde mucosale barrièrefunctie ex vivo te beoordelen. Intestinale sac voorbereidingen zijn in de eerste plaats zijn gebruikt in het farmaceutisch onderzoek, onderzoek naar de absorptie van kandidaat medicijnen over de darm. Echter, deze test even goed geschikt voor de studie van darmziekten. Darmdoorlaatbaarheid kan sterk verschillen per regio en site-specifieke beoordeling van de permeabiliteit zorgt voor een beter begrip van het regional…
