Intercellulære kryds er krav til brystkirtelspecifikke funktioner og udvikling. Dette manuskript giver en detaljeret protokol til undersøgelsen af protein-protein-interaktioner (PPI'er) og co-lokalisering ved anvendelse af murine brystkirtler. Disse teknikker tillader undersøgelse af dynamikken i den fysiske forbindelse mellem intercellulære krydsninger på forskellige udviklingsstadier.
Cellecelleinteraktioner spiller en afgørende rolle for bevarelse af vævsintegriteten og barrieren mellem de forskellige rum i brystkirtlen. Disse interaktioner tilvejebringes af forbindelsesproteiner, der danner sammenfald mellem tilstødende celler. Junctional protein mislokalisering og reducerede fysiske associationer med deres bindende partnere kan resultere i tab af funktion og følgelig til organ dysfunktion. Identifikation af proteinlokalisering og protein-protein-interaktioner (PPI'er) i normale og sygdomsrelaterede væv er således afgørende for at finde nye beviser og mekanismer, der fører til udviklingen af sygdomme eller ændringer i udviklingsstatus. Dette manuskript præsenterer en to-trins metode til evaluering af PPI'er i murine brystkirtler. I protokolafsnit 1 beskrives en fremgangsmåde til at udføre co-immunofluorescens (co-IF) ved anvendelse af antistoffer hævet mod proteiner af interesse efterfulgt af sekundære antistoffer mærket med fluorochromer. Selvom co-IF alleOws til demonstration af nærhed af proteinerne, gør det muligt at studere deres fysiske interaktioner. Derfor gives en detaljeret protokol for co-immunopræcipitation (co-IP) i protokolafsnit 2. Denne metode anvendes til at bestemme de fysiske interaktioner mellem proteiner uden at bekræfte, om disse interaktioner er direkte eller indirekte. I de sidste par år har co-IF og co-IP teknikker vist, at visse komponenter i intercellulære krydsninger samlokaliserer og interagerer sammen, hvilket skaber trinafhængige knudepunkter, der varierer under udvikling af brystkirtlen.
Mammekirtlen vokser og udvikler sig primært efter fødslen. Dette organ ombygger sig selv hele tiden i et pattedyr 1 's reproduktive liv. Det voksne brystkirtleepitel består af et indre lag af luminale epithelceller og et ydre lag af basale celler, hovedsageligt sammensat af myopiteliale celler omgivet af en kældermembran 2 . For en god gennemgang af brystkirtels struktur og udvikling kan læseren henvise til Sternlicht 1 . Cellecelleinteraktioner via mellemrum (GJ), stramme (TJ) og adherens (AJ) krydsninger er nødvendige for den normale udvikling og funktion af kirtlen 1 , 3 , 4 , 5 , 6 . Hovedkomponenterne i disse kryds i den murine brystkirtlen er Cx26, Cx30, Cx32 og Cx43 (GJ); Claudin-1, -3, -4 og -7 ogZO-1 (TJ); Og E-cadherin, P-cadherin og β-catenin (AJ) 7 , 8 . Ekspressionsniveauerne af disse forskellige forbindelsesproteiner varierer på en trinafhængig måde under udvikling af brystkirtler, hvilket foreslår differentialcellecelleinteraktionskrav 9 . GJ, TJ og AJ er strukturelt og funktionelt forbundet og sammenkobler andre strukturelle eller regulatoriske proteiner til de tilstødende steder i tilstødende celler og derved skaber en forbindelsesnekke 10 . Sammensætningen af den forbindelsesmæssige nexus kan påvirke brodannelsen med det underliggende cytoskelet såvel som nexuspermeabilitet og stabilitet og kan følgelig påvirke funktionen af kirtlen 8 , 9 , 10 , 11 . Komponenterne i intercellulære krydsninger, der befinder sig i krydsede nexuser eller interagerer med hinanden ved difHjerteudviklingsstadier af udvikling af brystkirtler blev analyseret for nylig ved anvendelse af co-immunofluorescens (co-IF) og co-immunpræcipitation (co-IP) 9 . Mens andre teknikker tillader evaluering af den funktionelle association mellem proteiner, er disse metoder ikke præsenteret i dette manuskript.
Da proteiner kun virker alene for at fungere, er det vigtigt for mange forskere at studere protein-protein-interaktioner (PPI'er), såsom signaltransduceringer og biokemiske kaskader, og kan give væsentlig information om proteinernes funktion. Co-IF og mikroskopisk analyse hjælper med at evaluere et par proteiner, der deler det samme subcellulære rum. Antallet af mål er imidlertid begrænset af antistofferne, som skal opdrættes i forskellige dyr og ved adgangen til et konfokalt mikroskop udstyret med forskellige bølgelængderlasere og en spektral detektor til multiplexering. Co-IP bekræfter eller afslører høj affinitet fysiske interaktioner betwEn to eller flere proteiner, der ligger inden for et proteinkompleks. På trods af udviklingen af nye teknikker, såsom fluorescensresonansenergioverførsel (FRET) 12 og nærhedsligeringsassay (PLA) 13 , som samtidig kan detektere lokalisering og interaktioner af proteiner, forbliver co-IP en passende og overkommelig teknik til undersøgelse af interaktioner mellem Endogene proteiner.
Den trinvise fremgangsmåde beskrevet i dette manuskript vil lette undersøgelsen af proteinlokalisering og PPI'er og påpege faldgruber for at undgå, når man studerer endogene PPI'er i brystkirtlerne. Metoden starter med præsentationen af de forskellige bevarelsesprocedurer for de krævede væv til hver teknik. Del 1 præsenterer, hvordan man studerer protein co-lokalisering i tre trin: i) snittet af brystkirtler, ii) dobbelt- eller tredobbelt mærkning af forskellige proteiner ved hjælp af co-IF-teknikken, og iii) billeddannelsen afProtein lokalisering. Del 2 viser, hvordan man udfælder et endogent protein og identificerer dets interagerende proteiner i tre trin: i) lysatpræparation, ii) indirekte proteinimmunpræcipitation og iii) bindingspartneridentifikation ved SDS-PAGE og Western blot. Hvert trin i denne protokol er optimeret til gnavere af brystkirtler og genererer højkvalitetsspecifikke og reproducerbare resultater. Denne protokol kan også bruges som udgangspunkt for PPI-studier i andre væv eller cellelinier.
Cell-celle interaktioner via kryds er nødvendige for den korrekte funktion og udvikling af mange organer, såsom brystkirtlen. Undersøgelser har vist, at forbindelsesproteiner kan regulere funktionen og stabiliteten af hinanden og aktivere signaltransduktion ved at tinde hinanden ved cellemembranen 10 .
