Reaktive Sauerstoff-Spezies (ROS)-Ansammlung ist ein Markenzeichen der Pflanze abiotischen Stress-Reaktion. ROS spielen eine Doppelrolle in Pflanzen als Signalmoleküle auf einem niedrigen Niveau und Moleküle auf einem hohen Niveau zu beschädigen. Anhäufung von ROS in gestresste Pflanzen kann Stoffwechselprodukte, Enzyme, Lipide und DNA, was zu einer Reduzierung des Pflanzenwachstums und der Ertrag beschädigen. Die Fähigkeit der Cerium-oxid-Nanopartikel (Nanoceria), katalytisch ROS in Vivo Aufräumen bietet ein einzigartiges Werkzeug zu verstehen und Biotechnologe Pflanzenverträglichkeit abiotischem Stress. Hier präsentieren wir Ihnen ein Protokoll zu synthetisieren und Poly (Acryl) Säure beschichteten Nanoceria (PNC) zu charakterisieren, Schnittstelle die Nanopartikel mit Pflanzen über Blatt-Lamina-Infiltration und Überwachung ihrer Verteilung und ROS Aufräumvorgang in Vivo konfokale mit Mikroskopie. Aktuelle Molekulare Werkzeuge zur Manipulation der ROS-Akkumulation in Pflanzen sind nur die Modell-Arten und mühsamer Wandel Methoden erfordern. Dieses Protokoll für in Vivo ROS Aufräumvorgang hat das Potenzial, zu Wildtyp-Pflanzen mit breiten Blättern und Blattstruktur wie Arabidopsis Thalianaangewendet werden.
Cerium-oxid-Nanopartikel (Nanoceria) sind in lebenden Organismen, von der Grundlagenforschung zur Biotechnologie, aufgrund ihrer unterschiedlichen katalytischen reaktiven Sauerstoffspezies (ROS) Aufräumvorgang Fähigkeit1,2,3verbreitet. Nanoceria haben ROS Aufräumvorgang Fähigkeiten aufgrund einer großen Anzahl von Oberfläche Sauerstoff stellen, die abwechselnd zwei Oxidation Staaten (Ce3 + und CE-4 +) 4,5,6. Nanoceria bieten Bioingenieure und Biologen eine Nanotechnologie-basierte Tool, um abiotischem Stress-Reaktionen zu verstehen und zu verbessern Pflanzenverträglichkeit abiotischem Stress zu Pflanzen. Nanoceria der in Vivo ROS Aufräumvorgang Fähigkeiten sind unabhängig von Pflanzenarten und einfache Lieferung ins Pflanzengewebe hat das Potenzial, breite Anwendung außerhalb von Modellorganismen zu ermöglichen. Das übergeordnete Ziel dieses Protokolls ist es, (1) die Synthese und Charakterisierung von negativ geladenen Poly (Acryl) Säure Nanoceria (PNC), (2) die Lieferung und Verfolgung von PNC im gesamten Blattzellen und (3) die Kontrolle der PNC-fähigen ROS Aufräumvorgang beschreiben Vivo.
