Vanwege de inherente lage signaal-ruisverhouding (SNR) van Fӧrster resonantie energieoverdracht (FRET) gebaseerde sensoren, is het meten van cAMP-signalen een uitdaging geweest, vooral in drie ruimtelijke dimensies. Hier beschrijven we een hyperspectrale FRET-beeldvormings- en analysemethodologie die het mogelijk maakt om de cAMP-verdeling in drie ruimtelijke dimensies te meten.
Cyclisch AMP is een tweede boodschapper die betrokken is bij een breed scala aan cellulaire en fysiologische activiteiten. Verschillende studies suggereren dat cAMP-signalen gecompartimenteerd zijn en dat compartimentering bijdraagt aan signaleringsspecificiteit binnen de cAMP-signaleringsroute. De ontwikkeling van op Fӧrster resonantie energieoverdracht (FRET) gebaseerde biosensoren heeft het vermogen bevorderd om cAMP-signalen in cellen te meten en te visualiseren. Deze metingen zijn echter vaak beperkt tot twee ruimtelijke dimensies, wat kan resulteren in een verkeerde interpretatie van gegevens. Tot op heden zijn er slechts zeer beperkte metingen van cAMP-signalen in drie ruimtelijke dimensies (x, y en z) geweest, vanwege de technische beperkingen bij het gebruik van FRET-sensoren die inherent een lage signaal-ruisverhouding (SNR) vertonen. Bovendien zijn traditionele filtergebaseerde beeldvormingsbenaderingen vaak niet effectief voor nauwkeurige meting van cAMP-signalen in gelokaliseerde subcellulaire gebieden vanwege een reeks factoren, waaronder spectrale overspraak, beperkte signaalsterkte en autofluorescentie. Om deze beperkingen te overwinnen en fret-gebaseerde biosensoren te kunnen gebruiken met meerdere fluoroforen, hebben we hyperspectrale FRET-beeldvormings- en analysebenaderingen ontwikkeld die spectrale specificiteit bieden voor het berekenen van FRET-efficiëntie en de mogelijkheid om FRET-signalen spectraal te scheiden van verstorende autofluorescentie en / of signalen van extra fluorescerende labels. Hier presenteren we de methodologie voor het implementeren van hyperspectrale FRET-beeldvorming, evenals de noodzaak om een geschikte spectrale bibliotheek te construeren die niet ondersampled of oversampled is om spectrale ontmenging uit te voeren. Terwijl we deze methodologie presenteren voor het meten van driedimensionale cAMP-verdelingen in pulmonale microvasculaire endotheelcellen (PMVECs), kan deze methodologie worden gebruikt om ruimtelijke verdelingen van cAMP in een reeks celtypen te bestuderen.
Cyclisch adenosinemonofosfaat (cAMP) is een tweede boodschapper die betrokken is bij belangrijke cellulaire en fysiologische processen, waaronder celdeling, calciuminstroom, gentranscriptie en signaaltransductie. Een groeiend aantal bewijzen suggereert het bestaan van cAMP-compartimenten in de cel waardoor signaleringsspecificiteit wordt bereikt1,2,3,4,5,6,7. Tot voor kort werd cAMP-compartimentering afgeleid op basis van verschillende fysiologische of cellulaire effecten geïnduceerd door verschillende G-gekoppelde receptoragonisten8,9,10,11. Meer recent hebben fret-gebaseerde fluorescentiebeeldvormingssondes nieuwe benaderingen geboden voor de directe meting en observatie van cAMP-signalen in een cel12,13,14.
Förster resonantie energieoverdracht (FRET) is een fysisch fenomeen waarbij energieoverdracht plaatsvindt tussen donor- en acceptormoleculen op een niet-stralingsve manier wanneer de moleculen zich in de nabijheid bevinden15,16. Met de ontwikkeling van FRET-gebaseerde fluorescerende indicatoren is dit fysische fenomeen gebruikt in biologische toepassingen om eiwit-eiwitinteracties17,eiwitcolokalisatie18,Ca+2-signalering 19,genexpressie20,celdeling21 en cyclische nucleotidesignalering te bestuderen. FRET-gebaseerde cAMP-indicatoren bestaan doorgaans uit een cAMP-bindingsdomein, een donorfluoofoor en een acceptorfluoofoor22. De H188 cAMP-sensor12,22 die in deze methodologie wordt gebruikt, bestaat bijvoorbeeld uit een cAMP-bindingsdomein verkregen van Epac, ingeklemd tussen Turquoise (donor) en Venus (acceptor) fluoroforen. Bij basale omstandigheden (ongebonden) bevinden Turkoois en Venus zich in een zodanige oriëntatie dat FRET optreedt tussen de fluoroforen. Bij binding van cAMP aan het bindingsdomein treedt een conformatieverandering op die zodanig dat Turquoise en Venus uit elkaar bewegen, wat resulteert in een afname van FRET.
FRET-gebaseerde beeldvormingsbenaderingen bieden een veelbelovend hulpmiddel voor het onderzoeken en visualiseren van cAMP-signalen in een cel. De huidige op FRET gebaseerde microscopische beeldvormingstechnieken zijn echter vaak slechts gedeeltelijk succesvol in het bereiken van voldoende signaalsterkte om FRET met subcellulaire ruimtelijke helderheid te meten. Dit komt door verschillende factoren, waaronder de beperkte signaalsterkte van veel FRET-verslaggevers, de hoge mate van precisie die nodig is om veranderingen in FRET-efficiëntie nauwkeurig te kwantificeren en de aanwezigheid van verstorende factoren, zoals cellulaire autofluorescentie23,24. Het resultaat is vaak een FRET-beeld dat wordt geplaagd door zwakke SNR, waardoor visualisatie van subcellulaire veranderingen in FRET erg moeilijk is. Bovendien is onderzoek naar ruimtelijk gelokaliseerde cAMP-signalen bijna uitsluitend uitgevoerd in slechts twee ruimtelijke dimensies en is de axiale cAMP-verdeling zelden beschouwd25. Dit komt waarschijnlijk omdat een lage SNR de mogelijkheid belemmerde om cAMP-gradiënten in drie ruimtelijke dimensies te meten en te visualiseren. Om beperkingen van het gebruik van FRET-sensoren met een lage SNR te overwinnen, hebben we hyperspectrale beeldvormings- en analysebenaderingen geïmplementeerd om FRET in afzonderlijke cellen te meten25,26,27.
Hyperspectrale beeldvormingsbenaderingen werden ontwikkeld door NASA om terrestrische objecten te onderscheiden die aanwezig zijn in satellietbeelden28,29. Deze technieken zijn sindsdien vertaald naar het fluorescentiemicroscopieveld30, met verschillende commerciële confocale microscoopsystemen die spectrale detectoren aanbieden. In traditionele (niet-spectrale) fluorescentiebeeldvorming wordt het monster geëxciteerd met behulp van een banddoorlaatfilter of een laserlijn en wordt de emissie verzameld met behulp van een tweede banddoorlaatfilter, vaak geselecteerd om overeen te komen met de piekemissiegolflengte van de fluorofoor (en). Hyperspectrale beeldvormingsbenaderingen daarentegen proberen een volledig spectraal profiel te bemonsteren van ofwel de fluorescentie-emissie26,31,32 of excitatie33,34 op specifieke golflengte-intervallen. In onze eerdere studies toonden we aan dat hyperspectrale beeldvormings- en analysebenaderingen een verbeterde kwantificering van FRET-signalen in cellen kunnen bieden in vergelijking met traditionele filtergebaseerde FRET-beeldvormingstechnieken26. Hier presenteren we een methodologie voor het uitvoeren van 4-dimensionale (x, y, z en λ) hyperspectrale FRET-beeldvorming en -analyse om cAMP-verdelingen in drie ruimtelijke dimensies te meten en te visualiseren. Deze benaderingen hebben visualisatie van agonist-geïnduceerde cAMP ruimtelijke gradiënten in afzonderlijke cellen mogelijkgemaakt 25. Interessant is dat, afhankelijk van de agonist, cAMP-gradiënten zichtbaar kunnen zijn in cellen. De hier gepresenteerde methodologie maakt gebruik van spectrale ontmenging van niet-uniforme achtergrond en cellulaire autofluorescentie om de nauwkeurigheid van de FRET-metingen te verbeteren. Hoewel deze methodologie wordt gedemonstreerd in pulmonale microvasculaire endotheelcellen (PMVECs) met behulp van een cAMP FRET-biosensor, kan de methodologie gemakkelijk worden aangepast voor gebruik met alternatieve FRET-reporters of alternatieve cellijnen.
De ontwikkeling van FRET-biosensoren heeft de meting en visualisatie van cyclische nucleotidesignalen in afzonderlijke cellen mogelijk gemaakt, en er is een grote belofte voor het visualiseren van subcellulaire signaleringsgebeurtenissen13,22,37,38.
