Measuring the barrier to the interspecies transmission of prion diseases is challenging and typically involves animal challenges or biochemical assays. Here, we present an in vitro prion protein conversion assay with the ability to predict species barriers.
Studies to understanding interspecies transmission of transmissible spongiform encephalopathies (TSEs, prion diseases) are challenging in that they typically rely upon lengthy and costly in vivo animal challenge studies. A number of in vitro assays have been developed to aid in measuring prion species barriers, thereby reducing animal use and providing quicker results than animal bioassays. Here, we present the protocol for a rapid in vitro prion conversion assay called the conversion efficiency ratio (CER) assay. In this assay cellular prion protein (PrPC) from an uninfected host brain is denatured at both pH 7.4 and 3.5 to produce two substrates. When the pH 7.4 substrate is incubated with TSE agent, the amount of PrPC that converts to a proteinase K (PK)-resistant state is modulated by the original host’s species barrier to the TSE agent. In contrast, PrPC in the pH 3.5 substrate is misfolded by any TSE agent. By comparing the amount of PK-resistant prion protein in the two substrates, an assessment of the host’s species barrier can be made. We show that the CER assay correctly predicts known prion species barriers of laboratory mice and, as an example, show some preliminary results suggesting that bobcats (Lynx rufus) may be susceptible to white-tailed deer (Odocoileus virginianus) chronic wasting disease agent.
Overdraagbare spongiforme encefalopathieën (TSE's, prionziekten) zijn een groep van fatale neurodegeneratieve ziekten met uitgebreide incubatie periodes die een verscheidenheid aan dieren en mensen beïnvloeden. De vermeende etiologische agens van TSE bestaat uit een verkeerd gevouwen isomeer van de gastheer prioneiwit (PrP), dat in staat is zichzelf voortplant sjabloongerichte omzetting van de normale cellulaire vorm van PrP (PrP C) in een besmettelijke, ziekte-geassocieerde vorm (PrP TSE) dat zich ophoopt in het centrale zenuwstelsel weefsels van de geïnfecteerde gastheer 1. Besmettelijke zoogdieren prionen algemeen uitzenden van host-to-host in een soort-specifieke manier, die aanleiding heeft gegeven tot het concept van de "TSE soort barrière" het beperken van interspecies transmissie gebeurtenissen 2. De biologische determinanten van het prion soort barrière zijn niet goed begrepen. Aminozuurvolgorde gelijkenis tussen de besmettelijke PrP TSE en de gastheer PrP C ceen sterke invloed op de vraag of conversie plaatsvindt 3-5, maar blijft onvoldoende om alle prion transmissie gebeurtenissen waargenomen in vivo 6,7 verklaren.
Zo heeft de karakterisering van TSE soorten barrières grotendeels ingeroepen dier challenge studies: blootstellen naïeve dieren van een bepaalde soort te prionen van een ander en het meten van de resulterende incubatietijd van de ziekte ontstaan en de attack rate als indicatoren van de transmissie-efficiëntie. Muizen die PrP C van heterologe species worden ook gebruikt voor dit type onderzoek 8. De kosten in verband met transgene muis productie en langdurige prion bioassays, evenals ethische overwegingen van het gebruik van dieren, zijn belemmeringen voor experimenteel onderzoek van TSE soorten barrières. Beoordeling van de menselijke soort barrière voor TSE's is gebaseerd op gemanipuleerde muizen met menselijke PrP C te drukken. Deze muizen vereisen lange incubatieperiode te bezwijken voor de menselijke TSE of aanpassingen aan tHet menselijke PrP C molecule voor snelle ziektebegin 9. Incubatieperiode van de niet-menselijke TSE's bij deze muizen kunnen buiten de normale muis levensduur waardoor interpretatie van negatieve resultaten uitdagend. Niet-menselijke primaten zijn ook gebruikt als proxies voor het bestuderen soortbarrières mens, maar deze studies zijn beladen met dezelfde problemen als andere soorten dierproeven en niet-menselijke primaten niet precies ziekte herhalen terwijl het zich in de menselijke gastheer.
Animal bioassays blijven de "gouden standaard" methode voor het meten van de gevoeligheid van een soort van een TSE, maar de obstakels, kosten en ethische deze levende dierstudies onderzoek naar alternatieven gedwongen. Een aantal in vitro assays, gebaseerd op de beoordeling van de omzetting van PrP c gastheer een proteïnase K (PK) resistente toestand (PrP res) na enting van PrP TSE, ontwikkeld en gebruikt om TSE species barrières 10-12. Voorbeelden van in vitro assays omvatten celvrije conversie testen eiwit misfolding cyclische amplificatie (PMCA) en de conversie efficiency ratio (CER) test 10-14. Hoewel geen van deze tests rekening worden gehouden met betrokken bij soorten barrières na natuurlijke infectie perifere factoren, kan alle nuttige om potentieel gevoelig hosts identificeren voor TSE's zijn.
Hier presenteren we het protocol voor de CER assay, waarbij twee gedenatureerd PrP C substraten afgeleid van normaal hersenweefsel homogenaten worden in tafelmodel prion omzettingsreacties (figuur 1) 13. PrP c in het substraat gedenatureerd bij pH 7,4 kan alleen PrP res worden omgezet PrP TSE gezaaid in de reactie in de afwezigheid van een soort barrière 14. Daarentegen denaturatie van PrP c in het andere substraat bij pH 3,5 het geschikt maakt omgezet PrP res na incubatiemet PrP TSE van elke soort en dient als controle voor conversie. De verhouding van de omzetting van PrP c naar PrP res in pH 7.4 substraat ten opzichte van die van de pH 3,5 substraat een maat van de soort barrière. Wij hebben gevonden dat de CER test voorspelt bekende species barrières van laboratoriummuizen verschillende TSE en de assay gebruikt in pogingen om de soort dam van vele zoogdiersoorten, waaronder bighornschapen, chronische wasting disease (CWD) en andere TSE 14 voorspellen, 15. Onderzoekers geïnteresseerd in een tool waardoor een snelle screening van TSE soorten barrières of beoordeling van PrP C to-PrP-res conversie zal deze methodiek nuttig vindt.
Voor een succesvolle afronding van dit protocol, moet aandacht worden besteed aan PrP C niveaus in niet-geïnfecteerde hersenweefsel gebruikt voor voorbereiding van de ondergrond (stap 2.1.2) en het zaad tot ratio voor conversie reacties (stap 4.4.2) substraat. In onze ervaring, kunnen de hersenen worden gewonnen voor gebruik als CER substraat na een aanzienlijke periode post-mortem, zolang PrP C aanwezig door immunoblotting (figuur 4) is.
