A convenient and powerful method for studying autophagy in Penicillium chrysogenum by using starvation pads (mixtures of agarose and tap water in a microscope slide containing a central cavity) is presented here.
The study of cellular quality control systems has emerged as a highly dynamic and relevant field of contemporary research. It has become clear that cells possess several lines of defense against damage to biologically relevant molecules like nucleic acids, lipids and proteins. In addition to organelle dynamics (fusion/fission/motility/inheritance) and tightly controlled protease activity, the degradation of surplus, damaged or compromised organelles by autophagy (cellular ‘self-eating’) has received much attention from the scientific community. The regulation of autophagy is quite complex and depends on genetic and environmental factors, many of which have so far not been elucidated. Here a novel method is presented that allows the convenient study of autophagy in the filamentous fungus Penicillium chrysogenum. It is based on growth of the fungus on so-called ‘starvation pads’ for stimulation of autophagy in a reproducible manner. Samples are directly assayed by microscopy and evaluated for autophagy induction / progress. The protocol presented here is not limited for use with P. chrysogenum and can be easily adapted for use in other filamentous fungi.
Filamenteuze schimmels zijn uitstekend modelsysteem voor het bestuderen van ontwikkelingsprocessen. Ze bieden verschillende experimentele voordelen zoals goedkope teelt, de hoge aantallen van de nakomelingen en genetische toegankelijkheid. Het laatste punt is van bijzonder belang voor de bouw van transformanten die het mogelijk maken het onderzoek naar het belang van zo-ver uncharacterized genen voor verschillende cellulaire mechanismen. Schimmels zijn voor het ophelderen van verschillende elementen en mechanismen van cellulaire kwaliteitscontrole trajecten protease activiteit voor de afbraak van afwijkende eiwitten, mitochondriale dynamiek lid voor het handhaven mitochondriale integriteit en autophagy voor het verwijderen van overtollige en / of disfunctionele celbestanddelen en onderhouden levensvatbaarheid van de cellen in tijden van hongersnood 1,2,3.
Er zijn verschillende experimentele technieken beschikbaar voor de studie van autofagie in draadschimmels 2: (i) onderzoek vacuolen if zij dichte autofagocytische lichamen vertonen wanneer proteasen worden geremd door transmissie elektronenmicroscopie 4, (ii) visualisatie van autofagosomen door toezicht GFP-Atg8 foci via fluorescentiemicroscopie 5,6 en (iii) detectie van aangezuurde autophagosomal structuren met de fluorescerende kleurstof monodansylcadaverine 7.
Hier wordt een nieuwe kweekmethode Penicillium chrysogenum voor autophagy studies gepresenteerd. Het belangrijkste element is de 'honger pad' die gewoon bestaat uit 1% agarose opgelost in gesteriliseerd kraanwater. Extra verbindingen (bijv stressoren, scavengers, autophagy modulatoren) worden aan de pad toegevoegd zolang zij niet automatisch fluorescentie geven. Het pad ligt in de microscoop dia's die een ondiepe centrale holte bevatten. Deze pad in geënt ofwel met een sporesuspensie of met kleine mycelium fragmenten. Dit laatste is geboden als de stam van belang niet sporulate efficiënt (bijv Δ atg1 stammen 8). Monsters worden geplaatst in vochtige kamers (deze kunnen gemakkelijk worden geconstrueerd met behulp lege pipetpunt dozen) verdroging voorkomen dat het monster en geïncubeerd bij kamertemperatuur. P. chrysogenum kan groeien enkele dagen onder deze omstandigheden. Autofagie kan microscopisch worden waargenomen door de vacuole uitbreiding, die een positieve marker voor schimmel autofagie. In deze bijdrage, een P. chrysogenum stam (Wisconsin 54-1255) gebruikt die groen fluorescent eiwit dat is gericht naar peroxisomen door zijn C-terminale 'SKL-sequentie 9 vormt. Daarom is het mogelijk om de afbraak van peroxisomen controleren. Het is mogelijk om ook labelen andere compartimenten van de cel (bijvoorbeeld mitochondria) waarbij geschikte lokalisatie signalen en hun degradatie analyseren. Hoewel gegevens van P. chrysogenum Ws54-1255 (GFP-SKL) wordt hier voorgesteld, is het zeker mogelijkde "honger pad methode ook voor andere filamenteuze schimmels (bijvoorbeeld Neurospora crassa, Sordaria macrospora, Aspergillus species, etc.).
De hier gepresenteerde methode maakt het handig en reproduceerbare studie van autofagie in P. chrysogenum. Zo kan het worden gebruikt voor het screenen van de effectiviteit van verschillende verbindingen of zij kunnen moduleren de autophagy respons van deze schimmel of niet. De resultaten met rapamycine bewijzen dat remming van TOR signaal leidt tot een uitgesproken inductie van autofagie in P. chrysogenum die is ook aangetoond voor andere organismen 11.
