We developed an in vitro model of dormancy in the bone marrow for estrogen-sensitive breast cancer cells. The goal of this protocol is to demonstrate use of the model for the study of the molecular and cellular biology of dormancy and for generation of hypotheses for subsequent testing in vivo.
The study of breast cancer dormancy in the bone marrow is an exceptionally difficult undertaking due to the complexity of the interactions of dormant cells with their microenvironment, their rarity and the overwhelming excess of hematopoietic cells. Towards this end, we developed an in vitro 2D clonogenic model of dormancy of estrogen-sensitive breast cancer cells in the bone marrow. The model consists of a few key elements necessary for dormancy. These include 1) the use of estrogen sensitive breast cancer cells, which are the type likely to remain dormant for extended periods, 2) incubation of cells at clonogenic density, where the structural interaction of each cell is primarily with the substratum, 3) fibronectin, a key structural element of the marrow and 4) FGF-2, a growth factor abundantly synthesized by bone marrow stromal cells and heavily deposited in the extracellular matrix. Cells incubated with FGF-2 form dormant clones after 6 days, which consist of 12 or less cells that have a distinct flat appearance, are significantly larger and more spread out than growing cells and have large cytoplasm to nucleus ratios. In contrast, cells incubated without FGF-2 form primarily growing colonies consisting of >30 relatively small cells. Perturbations of the system with antibodies, inhibitors, peptides or nucleic acids on day 3 after incubation can significantly affect various phenotypic and molecular aspects of the dormant cells at 6 days and can be used to assess the roles of membrane-localized or intracellular molecules, factors or signaling pathways on the dormant state or survival of dormant cells. While recognizing the in vitro nature of the assay, it can function as a highly useful tool to glean significant information about the molecular mechanisms necessary for establishment and survival of dormant cells. This data can be used to generate hypotheses to be tested in vivo models.
Borstkanker metastaseren naar het beenmerg voordat de ziekte detecteerbaar 1, zodra kleine kankers bloedvaten 2,3. De metastatische proces is snel, maar inefficiënt. De studie slaaptoestand in vivo is buitengewoon moeilijk, omdat micrometastasen zeldzaam en worden overtroffen door hematopoietische cellen met meer dan 10 6-voudige. Daarom moet relevante modellen worden gegenereerd die voorzien in vitro data die mechanismen kunnen suggereren en het genereren van toetsbare hypothesen in vivo. Een aantal slaaptoestand modellen, waaronder matheuitkraging modellen 12,13, in vitro modellen 7,8,14,15, in vivo xenograft modellen 16 combinaties van in vitro en xenograft modellen 17,18 en spontane tumoren en metastase model 19, hebben enig inzicht in kankercel rusttoestand 20 opleverde . Elk van deze modellen hebben hun eigen beperkingen en zijn van zichzelf vooral nuttig voor het genereren van hypothesen over moleculaire signalering en interacties die bepalen slaaptoestand te testen in meer biologisch relevante modellen.
Met het algemene doel van het definiëren van de moleculaire mechanismen van slaaptoestand, de interacties met de micro-omgeving die resulteert in cyclus arrest, redifferentiatie en therapeutische weerstand en mechanismen die leiden tot een herhaling in ER + cellen, we een in vitro model dat zorgt voor geselecteerde relevante elementen van de ontwikkelde stromale micromilieu 7. Dit model, terwijl relatief schaars in zijn componenten, voldoende robuust is om onderzoekers mogelijk om specifieke moleculaire mechanismen die significant functies slaaptoestand invloed afleiden. Deze experimenten genereren hypotheses die direct kunnen worden getest in vivo. Het model is gebaseerd op een aantal belangrijke elementen die we aangetoond relevant rusttoestand zijn. Zij omvatten het gebruik van oestrogeen-afhankelijke borstkanker cellen, kweken van cellen in een klonale dichtheid waar hun interactie primair bij de ondergrond en oplosbare bestanddelen van het medium, een fibronectine substraat en de aanwezigheid van basische fibroblast groeifactor (FGF-2) in het medium.
We kenmerk mechanismen die het systeem regelen in vitro, waaronder de inductie van celcyclus arrestatie door FGF-2 21, gemedieerd door TGFp 22, overleving signalering door PI3 kinase ERK 7,8 tot 8 en morfogene differentiatie epitheliale fenotype, dat op afhing RhoA inactivering integrine45, 5β1 opregulatie en ligatie van stromale fibronectine voor overleving 7,15 (figuur 1). De in vitro celcyclus effecten van FGF-2 aan MCF-7 cellen beginnen bij concentraties ten minste één log dan 10 ng / ml 21,23. De grondgedachte was gebaseerd op de temporele controle van FGF-2 expressie betreffende mammaire ductale morfogenese, cyclische expansie en recessie in een aantal zoogdiersystemen 24-27. We toonden aan dat FGF-2 induceert, waaronder ductale morfogenese in 3D cultuur 28 en dat FGF-2 expressie algemeen verloren maligne transformatie van humane tumoren 29. De expressie van FGR1 bleef intact in borstcarcinomen ondervraagde 29 en MCF-7 cellen blijven alle 4 FGF-receptoren 30 te drukken. In de context van de slaaptoestand, is FGF-2 werd uitgevoerd door en zwaar afgezet op beenmerg stroma 31,32 wanneer het functioneert in het behoud van hematopoietische stamcellen 33. We demonstrated dat FGF-2 leidt tot een slapende toestand in ER + borstkankercellen gekweekt op fibronectine substraten, ook overvloedig in het beenmerg, waar het induceert morfogenisch differentiatie 7. In het model borstkankercellen zijn groei geremd, inactiveren Rho A via RhoGap GRAF, redifferentiate een epitheliaal fenotype en re-express integrinen α5β1 lost met maligne progressie. Ze binden fibronectine door middel van integrine α5β1 en activeer survival signaleren dat ze bestand zijn tegen cytotoxische therapie 7,8,15 (figuur 1) te maken. Remming van Rho GTPases klasse is eerder aangetoond dat een slapende fenotype 34 induceren.
Hier zullen we de specifieke procedures die onderzoekers zullen toelaten om het model vast te stellen en te bestuderen specifieke moleculaire en cellulaire mechanismen die rustperiode van ER + borstkankercellen schetsen. In de experimenten die hier ter illustratie van het gebruik van het modelWij gerichte PI3K (Figuur 1B) met een Akt-remmer en een PI3K remmer en alle leden van de Rho familie (Figuur 1B) met een pan-Rho inhibitor en een Rho kinase (ROCK) inhibitor.
