Here, we present a protocol based on c-FOS protein immunohistological detection, a classical technique used for the identification of neuronal populations involved in specific physiological responses in vivo and ex vivo.

Twee benaderingen beschreven om mee neuronale populaties te identificeren in vivo bij dieren en ex vivo in deafferented hersenstam preparaten. In vivo dieren werden blootgesteld aan hypercapnische of hypoxische gasmengsels. Ex vivo werden deafferented preparaten superfused met hypoxische of hypercapnische kunstmatige cerebrospinale vloeistof. In beide gevallen, zowel in vivo controle dieren of ex vivo preparaten werden onder normoxische en normocapnic voorwaarden gehandhaafd. De vergelijking van deze twee benaderingen maakt de bepaling van de oorsprong van de neuronale dwz perifere en / of centrale. In vivo en ex vivo, brainstems verzameld, gefixeerd en in secties gesneden. Zodra secties werden bereid, werd immunohistochemische detectie van het c-fos eiwit gemaakt om de hersenstam groepen cellen geactiveerd door hypoxische of hypercapnische stimulaties identificeren. Gelabelde cellen werden geteld in hersenstam respiratoire structuren. In vergelijking met de controlegroep, hypoxie of hypercapnie verhoogde het aantal c-FOS gemerkte cellen in verschillende specifieke hersenstam sites die aldus constitutief van de zenuwbanen die bij de aanpassing van de centrale ademhalingscentrum zijn.
Het c-fos gen werd geïdentificeerd voor het eerst aan het begin van 1980 1,2 en het product werd gekarakteriseerd in 1984 als nucleair eiwit met gen-activator eigenschappen 3,4. Hij neemt deel aan de lange termijn mechanismen geassocieerd met neuron stimulatie. Inderdaad, veranderingen in neuronale activiteit tot second messenger signalerende cascades die de expressie van het directe vroege gen c-fos, die de productie van de transcriptiefactor c-FOS induceert induceren. De laatste initieert de expressie van late genen en derhalve deel aan adaptieve responsen van het zenuwstelsel van vele verschillende soorten stimuli 4. Aldus, sinds eind 1980 5,6, c-FOS eiwitdetectie is vaak gebruikt om de effecten van exogene factoren op gentranscriptie in het algemeen 4 en op de activiteit van het centrale zenuwstelsel (CNS) studeren kaart brengen zenuwbanen betrokken bij verschillende fysiologischeal omstandigheden.
Basale c-fos-expressie is onderzocht in diverse species waaronder muizen, ratten, katten, apen en humane 4. Daardoor wordt de kinetiek van de expressie vrij goed bekend. De transcriptie-activering is snel (5 tot 20 min) 7,8 en de mRNA accumulatie bereikt een maximum tussen 30 en 45 minuten na het begin van stimulatie 9 en afneemt met een korte halfwaardetijd van 12 min. De c-FOS eiwitsynthese volgt accumulatie van mRNA en door immunohistochemie kan worden gedetecteerd bij 20 tot 90 min na stimulatie 6.
Analyse van c-fos-expressie is klassiek gebruikt in in vivo studies om de centrale respiratoire netwerk betrokken zijn bij de ademhaling responsen op hypoxie of hypercapnie 10-14 identificeren. Meer recent werd dit instrument ook gebruikt in ex vivo hersenstam voorbereidingen naar het centrum van de luchtwegen netwerk aanpassingen aan hypoxie of h te verkennenypercapnia 15-18. Sterker nog, deze voorbereidingen genereren een ritmische activiteit klassiek gelijkgesteld aan de centrale luchtwegen aandrijving 19. Dus dit soort preparaat heeft het voordeel dat volledig deafferented en derhalve resultaten met betrekking tot c-fos expressie de gevolgen van een centrale stimulatie geven alleen zonder tussenkomst van perifere structuren.
De c-FOS detectie daarin van immunohistochemische of immunohistofluorescence benaderingen. Indirecte immunodetectie vereist het gebruik van een primair antilichaam tegen c-FOS en een secundair antilichaam gericht tegen de soorten waarbij het primaire antilichaam werd geproduceerd. Voor de immunohistochemische methode wordt het secundaire antilichaam geconjugeerd met een enzym (peroxidase, bijvoorbeeld) die inwerkt op een substraat (H 2 O 2 voor peroxidase). Het product van de enzymatische reactie wordt ontwikkeld door een chromogeen (3,3-diaminobenzidine tetrahydrochloride), die vlekken en onder lichtmicroscopie worden waargenomen. De reactie kan worden versterkt met behulp van nikkel ammoniumsulfaat. Deze werkwijzen maken de detectie van actieve neuronen in verschillende fysiologische problemen en dus de identificatie en / of het in kaart brengen van perifere en centrale routes betrokken bij de opeenvolgende fysiologische reacties.
C-fos is een directe vroege gen, en de detectie van het product, de c-fos-eiwit, is klassiek gebruikt om neuronale populaties betrokken bij specifieke respiratoire responsen in vivo 11,13,25,28 identificeren en ex vivo 16-18, 27,32,33.
