EPR og HJORT signaler oppstår fra en paramagneticspin etikett (SL) (dvs., methanethiosulfonate) knyttet til en enkelt-cystein rester i protein. Spin-etikettene rapportere tre typer strukturinformasjon: bevegelse, accessibilities og avstander. Denne informasjonen kan avgjøre om rester er gravlagt i protein eller utsettes for membran eller vandig miljø i apo og ligand binding stater13,17,18,19. Formålet her er å beskrive metoder for uttrykket, rensing, merking og rekonstituering av en funksjonell minimal cystein mindre rotte TRPV1 (eTRPV1) konstruere for spectroscopic analyser.
