Denne protokollen presenterer en metode for å utføre kvantitativ, enkelt-celle in situ analyse av protein ekspresjon for å studere avstamning spesifikasjon i mus preimplantation embryoer. De prosedyrer som er nødvendige for innsamling av blastocyster, hel-mount immunofluorescent påvisning av proteiner, avbildning av prøvene på en konfokalmikroskop, og kjernefysisk segmentering og bildeanalyse er beskrevet.
