Dit protocol beschrijft de methode, materialen, apparatuur en stappen voor bottom-up voorbereiding van RNA en eiwit produceren synthetische cellen. Het binnenste waterige compartiment van de synthetische cellen bevatte de S30 bacteriële lysaat ingekapseld in een lipide tweelaag (dat wil zeggen, stabiele liposomen), met behulp van een water-in-olie emulsie overdracht methode.
