Integrering av ulike synaptiske innganger til nevroner måles best i et preparat som bevarer alle presynaptiske kjerner for naturlig timing og kretsplastisitet, men hjerneskiver som vanligvis bryte mange tilkoblinger. Vi utviklet en modifisert hjerneskive for å etterligne in vivo kretsaktivitet samtidig som in vitro eksperimenteringsevne.
In vitro skive elektrofysiologi teknikker måle encellede aktivitet med presis elektrisk og timelig oppløsning. Hjerneskiver må være relativt tynne for å visualisere og få tilgang til nevroner for patch-klemming eller bildebehandling, og in vitro-undersøkelse av hjernekretser er begrenset til bare det som er fysisk tilstede i den akutte skiven. For å opprettholde fordelene med in vitro skive eksperimentering samtidig bevare en større del av presynaptic kjerner, utviklet vi en ny skive forberedelse. Denne “kile skive” ble designet for patch-clamp elektrofysiologi opptak for å karakterisere de ulike monaural, lyddrevne innganger til mediale olivocochlear (MOC) nevroner i hjernestammen. Disse nevronene får sine primære afferent excitatory og hemmende innganger fra nevroner aktivert av stimuli i det kontralaterale øret og tilsvarende cochleakjerne (CN). En asymmetrisk hjerneskive ble designet som er tykkest i rostro-caudal domenet på sidekanten av en halvkule og deretter tynner mot den laterale kanten av motsatt halvkule. Denne skiven inneholder, på den tykke siden, den hørbare nerveroten som formidler informasjon om auditive stimuli til hjernen, de indre CN-kretsene, og både de disynaptiske eksitatoriske og trisynaptiske hemmende afferentbanene som konvergerer på kontralaterale MOC-nevroner. Opptak utføres fra MOC nevroner på den tynne siden av skiven, hvor de visualiseres ved hjelp av DIC optikk for typiske patch-klemme eksperimenter. Direkte stimulering av hørselsnerven utføres når den kommer inn i den hørbare hjernestammen, slik at iboende CN-kretsaktivitet og synaptisk plastisitet kan forekomme ved synapser oppstrøms av MOC-nevroner. Med denne teknikken kan man etterligne in vivo kretsaktivering så nært som mulig i stykket. Dette kileskivepreparatet gjelder for andre hjernekretser der kretsanalyser vil ha nytte av bevaring av oppstrøms tilkobling og langdistanseinnganger, i kombinasjon med de tekniske fordelene med in vitro skive fysiologi.
Observasjon av aktiviteten til nevrale kretser er ideelt utført med innfødte sensoriske innganger og tilbakemeldinger, og intakt tilkobling mellom hjerneregioner, in vivo. Men å utføre eksperimenter som gir encellede oppløsning av nevrale kretsfunksjon er fortsatt begrenset av tekniske utfordringer i den intakte hjernen. Mens in vivo ekstracellulær elektrofysiologi eller multiphoton bildemetoder kan brukes til å undersøke aktivitet i intakte nervesystemer, tolke hvordan ulike innganger integrere eller måle subthreshold synaptiske innganger er fortsatt vanskelig. In vivo hele celleopptak overvinner disse begrensningene, men er utfordrende å utføre, selv i hjerneregioner som er lett tilgjengelig. Tekniske utfordringer med encellede oppløsningseksperimenter forsterkes ytterligere i visse nevronpopulasjoner som ligger dypt i hjernen, eller i romlig diffuse populasjoner som krever enten genetiske verktøy for å lokalisere celler in vivo (f.eks. genetisk uttrykk for kanalrhodopsin sammen med optrode-opptak) eller post-hoc histokjemisk identifikasjon etter registrering av stedmerking (f.eks. med nevrotransmisjonsspesifikke markører). Å være plassert diffust nær ventral overflaten av hjernestammen, lider mediale olivocochlear (MOC) nevroner av de ovennevntebegrensningene 1,noe som gjør dem ekstremt vanskelige å få tilgang til for in vivo eksperimentering.
Hjerneskiver (~ 100-500 μm tykkelse) har lenge vært brukt til å studere hjernekretser, inkludert auditive hjernestammekretser, på grunn av fysisk segregering av tilkoblede nevroner som finnes i samme skive2,3,4,5,6,7,8,9. Eksperimenter ved hjelp av mye tykkere skiver (> 1 mm) har blitt brukt i andre laboratorier for å forstå hvordan bilaterale innganger integreres i områder av det overlegne olivary komplekset (SOC), inkludert medial overlegen oliven10,11. Disse skivene ble utarbeidet slik at aksoner av hørselsnerven (AN) forble intakt i skiven og ble elektrisk stimulert til å initiere synaptisk nevrotransmitterutgivelse i CN, etterligne aktiviteten til første orden auditive nevroner som de ville reagere på lyd. En stor ulempe med disse tykke skiver er synlighet av nevroner for patch-klemme elektrofysiologiske opptak (“patching”). Patching blir stadig vanskeligere som de mange aksoner i dette området blir myelinated medalderen 12,13,14,15, noe som gjør vevet optisk tett og skjule nevroner selv i en typisk, tynn hjerneskive. Vårt mål er å skape in vitro-preparater som ligner tettere på kretstilkoblingen til in vivo-opptak, men med høy gjennomstrømning og høyoppløselige opptaksevner av visuelt styrt patch-clamp elektrofysiologi i hjerneskiver.
Laboratoriet vårt undersøker fysiologien til nevroner i det auditive efferent-systemet, inkludert MOC-nevroner. Disse kolinerge nevronene gir efferent tilbakemelding til sneglehuset ved å modulere aktiviteten til ytre hårceller (OHCs)16,17,18,19,20. Tidligere studier har vist at denne moduleringen spiller en rolle i gevinstkontroll i sneglehuset21,22,23,24,25,26 og beskyttelse mot akustisk traumer27,28,29,30,31,32,33. Hos mus er MOC-nevroner diffust plassert i ventral kjernen i trapeskroppen (VNTB) i den auditive hjernestammen1. Vår gruppe har benyttet ChAT-IRES-Cre muselinjen krysset med tdTomato reporter muselinje for å målrette MOC nevroner i hjernestamme skiver under epifluorescent belysning. Vi viste at MOC nevroner får afferent hemmende innspill fra ipsilateral medial kjerne av trapeskroppen (MNTB), som er begeistret, i sin tur, av aksler fra kuleformede buskete celler (GBC) i den kontralaterale cochleakjernen (CN)34,35,36,37,38. I tillegg får MOC-nevroner sannsynligvis sine eksitatoriske innspill fra T-stellate celler i kontralaterale CN39,40,41. Samlet viser disse studiene moc nevroner får både excitatory og hemmende innganger avledet fra samme (kontralaterale) øre. Imidlertid er de presynaptiske nevronene, og deres aksoner som konvergerer på MOC-nevroner, ikke helt nær nok til hverandre til å være helt intakte i et typisk koronarskivepreparat. For å undersøke hvordan integrering av synaptiske innganger til MOC-nevroner påvirker deres virkningspotensiale avfyringsmønstre, med fokus på nylig beskrevet hemming, utviklet vi et preparat der vi kunne stimulere de ulike afferentene til MOC-nevroner fra ett øre på den mest fysiologisk realistiske måten mulig, men med de tekniske fordelene med in vitro hjerneskiver eksperimenter.
Kileskiven er et modifisert tykt stykkepreparat designet for undersøkelse av kretsintegrasjon i MOC-nevroner (skjematisert i figur 1A). På den tykke siden av skiven inneholder kilen de avkuttede aksonene til hørselsnerven (kalt “auditiv nerverot” heretter) når de kommer inn i hjernestammen fra periferien og synapse i CN. Den auditive nerveroten kan stimuleres elektrisk til å fremkalle nevrotransmitterutgivelse og synaptisk aktivering av celler i den helt intakte CN42,43,44,45,46. Dette stimuleringsformatet har flere fordeler for kretsanalyse. Først, i stedet for å direkte stimulere T-stellate og GBC axons som gir afferent inngang til MOC nevroner, stimulerer vi AN for å tillate aktivering av iboende kretser rikelig i CN. Disse kretsene modulere produksjonen av CN nevroner til sine mål i hele hjernen, inkludert MOC nevroner46,47,48,49,50,51. For det andre, polysynaptisk aktivering av afferent kretser fra AN gjennom CN oppstrøms av MOC nevroner gir mer naturlig aktivering timing og for plastisitet å skje på disse synapser som de ville in vivo under auditiv stimulering. For det tredje kan vi variere våre stimuleringsmønstre for å etterligne AN-aktivitet. Til slutt er både excitatory og hemmende monaural projeksjoner til MOC nevroner intakt i kilehæren, og deres integrasjon kan måles ved en MOC neuron med presisjonen av patch-clamp elektrofysiologi. Som helhet gir denne aktiveringsordningen en mer intakt krets til MOC-nevronene sammenlignet med et typisk hjerneskiverpreparat. Denne brainstem kile skive kan også brukes til å undersøke andre auditive områder som mottar hemmende innspill fra ipsilateral MNTB inkludert lateral overlegen oliven, overlegen olivary kjerne og mediale overlegen oliven10,11,52,53,54,55,56. Utover vårt spesifikke preparat kan denne kuttemetoden brukes eller endres til å evaluere andre systemer ved å opprettholde tilkobling av langdistanseinnganger og forbedre visualisering av nevroner for en rekke encellede oppløsning elektrofysiologi eller bildeteknikker.
Denne protokollen krever bruk av et vibratomstadium eller en plattform som kan vippes ca. 15°. Her bruker vi et kommersielt tilgjengelig 2-delt magnetisk stadium hvor “scenen” er en metallplate med en buet bunn plassert i en konkav magnetisk “scenebase”. Scenen kan deretter skiftes for å justere skivevinkelen. Konsentriske sirkler på scenebasen brukes til å estimere vinkelen reproduserbar. Scenen og scenebasen er plassert i kuttekammeret, hvor den magnetiske scenebasen også kan roteres.
Kutteprosedyren beskrevet her kalt en kile skive er kraftig for å opprettholde intakt presynaptic neuronal kretser, men med tilgjengeligheten av hjernen skive eksperimentering for analyse av nevronal funksjon. Stor forsiktighet må tas i flere innledende trinn for å maksimere nytten av preparatet for kretsanalyse. Dimensjonene på kilen bør bekreftes ved hjelp av histologisk undersøkelse, noe som er integrert for bekreftelse på at både presynaptiske kjerner og deres aksonale projeksjoner finnes i den tilberedte kil…
