Modeling human brain development has been hindered due to the unprecedented complexity of neural epithelial tissue. Here, a method for the robust generation of brain organoids to delineate early events of human brain development and to model microcephaly in vitro is described.
The restricted availability of suitable in vitro models that can reliably represent complex human brain development is a significant bottleneck that limits the translation of basic brain research into clinical application. While induced pluripotent stem cells (iPSCs) have replaced the ethically questionable human embryonic stem cells, iPSC-based neuronal differentiation studies remain descriptive at the cellular level but fail to adequately provide the details that could be derived from a complex, 3D human brain tissue.
This gap is now filled through the application of iPSC-derived, 3D brain organoids, “Brains in a dish,” that model many features of complex human brain development. Here, a method for generating iPSC-derived, 3D brain organoids is described. The organoids can help with modeling autosomal recessive primary microcephaly (MCPH), a rare human neurodevelopmental disorder. A widely accepted explanation for the brain malformation in MCPH is a depletion of the neural stem cell pool during the early stages of human brain development, a developmental defect that is difficult to recreate or prove in vitro.
To study MCPH, we generated iPSCs from patient-derived fibroblasts carrying a mutation in the centrosomal protein CPAP. By analyzing the ventricular zone of microcephaly 3D brain organoids, we showed the premature differentiation of neural progenitors. These 3D brain organoids are a powerful in vitro system that will be instrumental in modeling congenital brain disorders induced by neurotoxic chemicals, neurotrophic viral infections, or inherited genetic mutations.
Humane neurologiske lidelser, såsom mikrocefali, kan kun dårligt undersøgt i dyremodeller på grund af det faktum, at den menneskelige hjerne har et udvidet kortikale overflade, en unik funktion adskiller sig fra ikke-humane dyr.

Takket være laboratorier, banebrydende 3D kultur teknologier og afmystificeret kompleksitet organdannelse, startende fra stamceller,vi udviklet en robust metode til generering hjernen organoider at afgrænse tidlige begivenheder af menneskelige hjerne udvikling og at modellere mikrocefali in vitro 1, 2, 3. Det er bemærkelsesværdigt, at vi tilpasset den oprindelige metode udviklet af Lancaster et al. at generere cerebrale organoider 1. Denne metode blev ændret i henhold til vores eksperimentelle krav.
Formålet med en undersøgelse fra Gabriel et al. var at analysere de cellulære og molekylære mekanismer i neural stamcelle vedligeholdelse under hjernens udvikling. For at gøre dette blev en mekanistisk forsøg udført med analyse neurale progenitorceller (NPCs) i 3D hjernen organoider afledt fra en mikrocefali patient 4. Denne patient bar en mutation i CPAP, en konserveret centrosomal protein, der kræves for centrosomreplikation biogenese 5. En bredt accepteret hypotese er, at mikrocefali er resultatet af en udtynding af NPC pool, og dette kan enten skyldes celledød eller for tidlig differentiering 1, 6, 7, 8, 9.
Ved at analysere de ventrikulære zoner (VZs) af mikrocephali hjerne organoider blev det vist, at et betydeligt antal NPC undergår asymmetrisk celledeling i modsætning hjernen organoider afledt fra en rask donor 4. Omfattende mikroskopiske og biokemiske analyser af microcephalic hjerne organoider afslørede en uventet rolle for CPAP i tide cilia demontering 4. Specifikt er muteret CPAP forbundet med retarderet cilium demontering og forsinket cellecyklus genindførsel, der fører til for tidlig differentiering af NPCs 4. Disse resultater tyder på en rolle for cilier i mikrocefali og thEIR inddragelse under neurogenese og hjerne størrelseskontrol 10.
Den første del af denne protokol er en beskrivelse af en tretrins metode til at generere homogene hjerne organoider. Som nævnt før, blev den oprindelige Lancaster protokol tilpasset og modificeret så de passer til vores formål 1. Første, humane iPSCs dyrkes i et defineret feeder-fri tilstand på Engelbreth-Holm-Swarm (EHS) matrix. Dette trin undgår variationer af feeder-afhængige pluripotente stamceller cellekulturer. I denne protokol, at induktionen af neural differentiering danne neurale epitel starter direkte fra iPSCs. Ved at springe embryoiddannelsesmuligheden legeme (EB) dannelse trin, neurale differentierings forløber i en mere kontrolleret og styret måde. Denne fremgangsmåde begrænser spontan og ikke-orienteret dannelse af andre kim cellelag, såsom mesoderm og endoderm. Ved at anvende denne protokol, kan neurosfærer indeholdende neurale rosetter høstes på dag 5 for EHS matrix indlejring og stationær suspensionskultur. Det organoide medium, der anvendes til det tredje trin i vores protokol er suppleret med dorsomorphin og SB431542. Dorsomorphin er et lille-molekyle inhibitor af knoglemorfogent protein (BMP), og SB431542 hæmmer TGFp / activin / nodal signalvej. Kombinationen af disse faktorer kan fremme neural differentiering mere effektivt end retinsyre alene 11, 12, 13, 14.
Tilsammen disse modifikationer muliggøre reproducerbar generation af hjernens organoider, med minimale variationer tværs organoider. Vigtigere, blev denne fremgangsmåde anvendt på robust generere microcephalic hjerne organoider fra patientens iPSCs, som bærer mutationer i gener, der påvirker centrosomer og celle-cyklus dynamik.
Den anden del af denne protokol giver instruktioner til at forberede brain organoider til analyse og fortolkning af cellulære defekter i mikrocefali. Dette omfatter fiksering, Fremstilling af frysesnit, immunfluorescensfarvning og konfokal mikroskopisk analyse. MCPH er en kompleks human neurologisk lidelse, der ikke kan rekapituleres i dyremodeller in vivo eller in simpel human cellekultur nærmer in vitro.
