O presente protocolo destaca a aplicação da técnica de western blotting para estudar as funções e atividades do co-transportador neuronal K-Cl KCC2. O protocolo descreve a investigação da fosforilação de KCC2 em sítios reguladores de quinase Thr906/1007 via western blotting. Os cotransportadores de cloreto de potássio 2 (KCC2) são um membro da família de transportadores de solutos 12 (SLC12) dos cotransportadores de cloreto de catião (CCCs), encontrados exclusivamente no neurônio e são essenciais para o bom funcionamento da Cl-homeostase e, consequentemente, da inibição funcional do GABAérgico. A falha na regulação adequada da KCC2 é deletéria e tem sido associada à prevalência de várias doenças neurológicas, incluindo epilepsia. Houve avanços consideráveis no que diz respeito à compreensão dos mecanismos envolvidos na regulação do KCC2, credenciados ao desenvolvimento de técnicas que permitam aos pesquisadores estudar suas funções e atividades; por meio de investigações diretas (avaliando a fosforilação dos sítios reguladores da quinase) ou indiretas (observando e monitorando a atividade do GABA). Aqui, o protocolo destaca como investigar a fosforilação de KCC2 em locais reguladores de quinase – Thr906 e Thr1007 – usando a técnica de western blotting. Existem outros métodos clássicos usados para medir diretamente a atividade do KCC2, como o ensaio de absorção de íons rubídio e íons tálio. Outras técnicas, como patch-clamp-electrophysiology são usadas para medir a atividade GABA; portanto, refletindo indiretamente KCC2 ativado e/ou inativado, conforme informado pela avaliação da homeostase intracelular de íons cloreto. Algumas dessas técnicas adicionais serão brevemente discutidas neste manuscrito.
Os cotransportadores de cloreto de potássio 2 (KCC2) são membros da família de transportadores de solutos 12 (SLC12) dos cotransportadores de cloreto de cátions (CCCs), encontrados exclusivamente no neurônio e são essenciais para o bom funcionamento da Cl-homeostase e, consequentemente, para a inibição funcional do GABAérgico 1,2,3,4. A manutenção da baixa concentração intraneuronal de Cl- ([Cl-]i) a 4-6 mM pelo KCC2 facilita a hiperpolarização do ácido γ-aminobutírico (GABA)/glicina e a inibição sináptica no cérebro e na medula espinhal5. A falha na regulação adequada da KCC2 tem sido associada à prevalência de várias doenças neurológicas, incluindo a epilepsia4. Além disso, a diminuição da extrusão de Cl− mediada por KCC2 e as correntes hiperpolarizantes hiperpolarizantes GABAA e/ou mediadas pelo receptor de glicina têm sido implicadas na epilepsia, dor neuropática e espasticidade 6,7. O KCC2 neuronal é modulado negativamente via fosforilação de resíduos reguladores chave dentro de seu domínio intracelular C-terminal pelo complexode sinalização 1 da quinase com não-lisina (WNK)-STE20/SPS1-related proline/alanine-rich (SPAK)/Oxidative stress-responsive (OSR), o que facilita a manutenção da atividade despolarizada do GABA em neurônios imaturos 2,8,9 . O WNK-SPAK/OSR1 fosforila os resíduos de treonina 906 e 1007 (Thr906/Thr1007) e, posteriormente, regula negativamente a expressão gênica de RNAm do KCC2, levando a uma consequente deterioração de sua função fisiológica 8,10. Mais importante, no entanto, já é um fato que o complexo quinase WNK-SPAK/OSR1 é conhecido por fosforilar e inibir a expressão de KCC2 1,2,4,11,12, e que a inibição das vias de sinalização do complexo quinase para o fosforilato Thr906/Thr1007 tem sido associada ao aumento da expressão do gene mRNA KCC2 13,14,15 . É importante ressaltar que a regulação da expressão neuronal de KCC2 e Na+-K+–2Cl-cotransportadores 1 (NKCC1) via fosforilação proteica funciona concomitantemente e em padrões inversos 1,4,16.

O protocolo aqui apresentado destaca a aplicação de técnicas de western blotting para estudar as funções e atividades do co-transportador neuronal K+-Cl– KCC2, investigando a fosforilação do cotransportador em sítios reguladores de quinase Thr906/1007.
Western blot é um método usado para detectar proteínas específicas de interesse de uma amostra de tecido ou célula. Este método primeiro separa as proteínas por tamanho através de eletroforese. As proteínas são então transferidas eletroforeticamente para um suporte sólido (geralmente uma membrana) antes que a proteína-alvo seja marcada usando um anticorpo específico. Os anticorpos são conjugados a diferentes tags ou anticorpos conjugados com fluoróforos que são detectados usando métodos colorimétricos, quimioluminescência ou fluorescência. Isso permite que uma proteína-alvo específica seja detectada a partir de uma mistura de proteínas. Essa técnica tem sido utilizada para caracterizar sítios fosfoespecíficos de KCCs1 e tem sido utilizada para identificar inibidores de quinase que inibem a fosforilação de KCC3 Thr991/Thr104817. Seguindo este protocolo, pode-se detectar especificamente KCC2 total e fosforilado a partir de lisatos celulares / teciduais. Em princípio, a detecção de anticorpos conjugados com proteínas por esta técnica é altamente instrumental, pois ajuda a melhorar a compreensão das atividades cooperativas nos fosfo-locais do KCC2, o que lança luz sobre os mecanismos moleculares envolvidos em suas regulações fisiológicas. A análise quantitativa da expressão proteica total é representativa da função e atividade do KCC2. Existem outros métodos clássicos usados para medir diretamente a atividade do KCC2, como o ensaio de absorção de íons rubídio e íons tálio.
