A hipersensibilidade de contato (CHS) é um modelo experimental de murina de dermatite de contato alérgico (ACD). O CHS baseia-se na sensibilização com hapten reativo pintando a pele raspada do peito e abdômen, com um subsequente desafio da pele do ouvido com um hapten diluído, causando uma reação de inchaço que é avaliada de várias maneiras.
A hipersensibilidade de contato (CHS) é um modelo experimental de dermatite de contato alérgico (ACD) que pode ser estudado em camundongos. Este estudo tem como objetivo apresentar um método laboratorial objetivo que possa ajudar a estudar a reação do CHS em camundongos, que pode ser medido e quantificado por vários testes. Para induzir o CHS, no dia “0”, os camundongos foram sensibilizados em um local previamente raspado por pintura de pele abdominal com a mistura hapten 2,4,6-trinitrcloobenzeno (TNCB) em uma mistura acetona-etanol, enquanto os ratos de controle negativo foram falsos sensibilizados com a mistura veículo sozinho-acetona-etanol. No dia “4”, a espessura da orelha da linha de base foi medida com um micrômetro antes da obtenção de CHS (desafio) pintando ambas as orelhas com TNCB diluído tanto nos grupos de teste quanto de controle. Após 24h, o inchaço da orelha foi medido com um micrômetro. O CHS é um exemplo de resposta imune mediada por células T que causa inchaço no tecido inflamado, atingindo 24 h após o desafio da pele com o mesmo hapten. Aumento do edema de ouvido correlacionado com aumento do peso do ouvido, atividade mieloperoxidase (MPO), concentração pró-inflamatório de citocina nos extratos de ouvido, aumento do espessamento do edematoso no exame histológico e permeabilidade vascular do ouvido. Houve também um aumento na concentração de anticorpos IgG1 específicos do TNP na sera do grupo de teste quando comparado com os camundongos de controle. Além disso, o CHS pode ser transferido com sucesso com as células efeito CHS obtidas de doadores previamente sensibilizados com tncb. As células com efeito CHS foram administradas por via intravenosa em camundongos receptores ingênuos, que foram posteriormente desafiados com o mesmo hapten diluído. O inchaço da orelha foi medido com um micrômetro 24 h depois.
A dermatite de contato alérgica (ACD) é uma doença inflamatória da pele comum em países industrializados causada por uma reação de hipersensibilidade tipo IV resultante da exposição a produtos químicos de baixo peso molecular chamados haptens. As substâncias que causam sensibilização de contato em humanos incluem íons de metal pesado (cromo, níquel, ferro, cobalto), terebintina, fragrâncias, corantes e conservantes presentes em cosméticos (por exemplo, p-fenilediamina), algumas drogas (por exemplo, neomicina, benzocaína), antibióticos β-lactam (ou seja, penicilina), produtos químicos produzidos por plantas (pentadecacatechol, substância presente na hera venenosa), bem como hidroquinona utilizada na indústria fotográfica 1,2 . Os agentes etiológicos ACD são muito altos, pois mais de 100.000 produtos químicos são usados apenas na indústria, e 2.000 novos são sintetizados a cada ano. Até o momento, mais de 3.700 moléculas foram identificadas que podem ser contato haptens/alergênicos3. A reação de hipersensibilidade de contato (CHS) é um modelo experimental de ACD que pode ser estudado em camundongos, cobaias e ratos e pode ser induzido pela aplicação da pele tópica de haptens químicos reativos dissolvidos em solventes orgânicos 4,5,6. Este estudo tem como objetivo descrever um método de laboratório objetivo que pode ajudar a estudar a reação do CHS em camundongos, que pode ser medido e quantificado por vários testes.
O CHS consiste em fases de sensibilização (indução) e efeito (desafio). Em modelos animais, os haptens primeiro se ligam covalentemente às proteínas do corpo para criar neoantígenos. Durante a fase de sensibilização, os queratinócitos ativados promovem a migração e a maturação das células dendríticas da pele (SDCs) produzindo o fator de necrose pró-inflamatório citocinas-tumora α (TNF-α) e interleucina 1β (IL-1β)7. As células epidérmicas langerhans (LCs) apresentam antígenos durante as fases 8 de indução e efeito do efetordo CHS. As LCs expostas ao hapten durante a sensibilização promovem a indução tanto das células regulatórias quanto das células9. Evidências crescentes de vários estudos sugerem que as respostas do CHS podem ser mediadas por células Th1 da classe 2 do CD4+ MHC, liberando localmente interferon-γ (IFN-γ) para empregar um infiltrado inflamatório característico, linfócitos Tc1 classe I restritos cd8+ que também podem liberar ifn-γ mas principalmente mediar danos citotóxicos para queratinócitos, e agora também interleucina 17 (IL-17)-produzindo células Th1710, 11 anos.
Vários modelos chs diferentes que empregam várias espécies1 2,13,14 e haptens foram desenvolvidos (uma comparação detalhada de diferentes haptens, solventes e tempo de aplicação é resumida na Tabela 1). O camundongo, uma espécie de laboratório frequentemente utilizada, oferece algumas vantagens no estudo do CHS. Há mais cepas, nocautes (KO) e animais transgênicos entre camundongos em comparação com outras espécies, o que os torna um animal muito atraente15. Além disso, o modelo CHS requer muitos animais, e os ratos são mais econômicos aqui. Modelos animais não imitam ACD em todos os aspectos; em particular, eles mostram crostas e desquamation, o que não é comum para humanos16,17. As características da doença crônica são desafiadoras para se reproduzir, principalmente porque o modelo descrito não assume a aplicação do hapten por um longo período de tempo. No entanto, foi confirmado aqui que muitos dos aspectos significativos da ACD são reproduzidos. Também foi demonstrado que, como em humanos, essas características estão associadas a reações alérgicas locais. A escolha do hapten, seu solvente e sua aplicação delineada neste protocolo foram ditadas pelo fato de que os resultados foram confirmados por inúmeros testes in vitro e que foi testado e modificado em laboratório por muitos anos até que a versão atual fosse estabelecida. Os modelos murinos permitem a análise dos subconjuntos celulares ou citocinas que estão envolvidos no desenvolvimento da ACD e são essenciais para avaliações pré-clínicas de novos tratamentos.
O CHS é induzido através de haptens, que se ligam a antígenos auto-proteína na pele, criando neoantígenos. O CHS é mediado pelo recrutamento extravascular local de células T com efeito de antígeno circulante, o que resulta em inchaço no tecido desafiado, atingindo 24 h após a exposição da pele secundária ao mesmo hapten6.
