Abstract

Introduction
内耳毛细胞是检测声音和隐藏电信号机械刺激的感官细胞,最终由我们的大脑解释。这些细胞有一个楼梯形的捆绑三行基于活性蛋白的细丝,称为立体丝,从它们的锥形区域1,2突出。机械刺激使立体丝向最长的行偏转,并触发机械转导(MET)通道3的打开。MET通道的开放导致阳离子的涌入,使细胞去极化,从而在头发细胞的基础区域发出突触囊泡的释放。
MET通道对听力至关重要的生物物理特性得到了广泛的描述。在其他属性中,这些通道是阳离子选择性的,具有相对较大的电导率(低 Ca2+中为 150±300 pS)4、5、6、7、8、9、10 。值得注意的是,大型荧光分子,如FM1-43和得克萨斯红标记氨胶苷是MET通道的渗透阻滞剂,导致其积累在头发细胞体,可以使用荧光显微镜11,12,13,14。相反,MET通道的分子特性和结构及其渗透途径仍然难以捉摸。越来越多的实验证据表明,跨膜样通道蛋白1(TMC1)是成熟毛细胞15、16、17、18、19中MET通道的组成部分。跨膜状通道1(TMC1)中的突变会改变MET通道特性19、20、21、22,并引起耳聋。此外,TMC1还本地化到MET通道18、23的站点,并与负责将机械力传输到MET通道24、25的尖端链路进行交互。此外,最近的生物信息学分析已经发现TMC蛋白是进化相关的对美甲敏感通道TMEM63/OSCA蛋白质和TMEM16蛋白质,钙活性氯化物通道和脂质scramblass26,27,28。基于这些蛋白质之间关系的TMC1结构模型揭示了在蛋白质-脂质界面27处存在一个大腔。该腔包含两个TMC1突变,导致常染色体显性听力损失(DFNA36)27、29、30、31、32,以及选择性地修饰半胱氨酸突变体,使其在腔内残留物改变MET通道特性28,表明它可以作为MET通道的渗透通路。TMC蛋白质中这种预测腔的较大尺寸可以解释大分子渗透MET通道的能力。为了测试MET通道包含异常大渗透通路的预测,并突破TMC1中观察到的腔体大小的极限,我们开发了一个协议,在Corti外植的器官中用更大的分子进行吸收实验,3 kDa dextran荧光标记为德州红。
Dextran是一种复杂的分枝多糖,由许多D-葡萄糖分子组成,由α-1,6糖碱基结结合。其高溶性在水中,低细胞毒性,和生物不性使其成为研究多种细胞过程的多功能工具。此外,dextran 有多种尺寸可供选择,并带有多种颜色的荧光标记。荧光标记的dextrans通常用于细胞和组织渗透性研究33,34,研究多细胞系统35,36的内分泌,以及神经追踪37,38。在听觉领域,dextran分子也被用来评估细胞-细胞结的中断和听觉感觉上皮完整性的丧失后暴露于Corti39,40的chinchilla器官的强烈噪音。
在这项工作中,我们利用一些最小的(3和10 kDa)荧光除变性器的特性,在鼠内耳毛细胞中进行吸收实验,并探索内耳毛细胞MET通道的渗透通路的大小。此外,我们使用激光扫描共聚焦显微镜(LSM)880配备了Airyscan探测器,以可视化和本地化荧光dextran在立体神经和听觉毛细胞的细胞体。

Protocol
动物护理和实验程序按照《实验室动物护理和使用指南》进行,该准则经国家神经疾病和中风研究所动物护理和使用委员会批准(动物协议#1336到KJS)。
1. 老鼠
- 设置一对C57BL/6J野生型的繁殖对在动物设施中繁殖,以控制幼崽的出生日期,并跟踪幼崽的年龄。
2. 耳蜗解剖
- 设置一个靠近立体显微镜的清洁空间以执行解剖(图1A)。使用 70% 乙醇清洁空间和周围环境,并放置干净的台垫。需要医疗病理废物(MPW)塑料袋才能丢弃动物尸体。
- 准备几个35毫米的菜肴与一些莱博维茨的L15介质。
注:莱博维茨的L-15细胞介质含有1⁄2 mM Ca2+,这是保持尖端链接完整性所必需的,并含有必需氨基酸、维生素和丙酸钠,以改善细胞健康和生存。血清被排除,以避免实验变异,由于其定义不良的成分和潜在的干扰dextran。 - 通过斩首使产后6天6(P6)小鼠安乐死。

- 将头骨分成两半,用小铲子去除大脑(图2C-D)。
- 用小剪刀,切周围的时间骨骼,并切除组织。
- 将两个临时骨骼放入 35 mm 的培养皿中,并确保它们被 L15 介质覆盖(图 2E)。
注:以下步骤在立体显微镜下执行。黑色背景通常有助于在精细解剖步骤期间可视化组织。 - 在配备宽场目镜(10倍放大功率(WF10X)和外部交流电(AC)卤素光源)的立体显微镜下,用手术钳切除周围的耳蜗组织、半圆形管道和前庭器官(图2F)。
- 为了让 dextran 和 L15 介质进入耳蜗管,请在解剖的耳蜗上执行两个穿刺边界,一个在圆窗上,另一个在耳蜗区域。
- 从 9 孔玻璃凹陷板中在每个孔中加入 300 mL 的 Leibovitz L15 介质。
- 在每个井上至少放置三个解剖的耳蜗。
3. 德特森标签
- 在汉克斯的平衡盐溶液中重组dextran,无Ca2+和Mg 2+(HBSS-CFM),最终浓度为10mg/mL。此库存溶液必须在不透明的黑色管中加报价(防止光线照射),并储存在 -30°C 下,直到使用。
注: 使用可莱辛可修复的dextran对于该协议的成功结果至关重要。 - 在莱博维茨的L15介质的500 mL中,以2mg/mL的最终浓度制备每个dextran。

4. 成像样品制备
- 使用 dextran 孵育后,用介质洗涤组织 2 分钟两次,用 HBSS 洗涤一次。
- 在室温下用4%的甲醛在汉克斯的平衡盐溶液(HBSS)中孵育组织30分钟。
注意:接触甲醛会刺激眼睛、鼻子和上呼吸道。在某些个体中,皮肤反复接触甲醛会导致过敏性皮肤炎。甲醛是已知的人类致癌物质,是一种疑似生殖危害。 - 用HBSS快速轻轻清洗固定组织两次,以去除甲醛。
注:在耳蜗的这个发育阶段,不需要对骨骼进行去化。 - 去除螺旋韧带和细尖钳膜,以解剖Corti的器官(图2G)。
- 取出所有小块组织,用哈佛商学院清洗组织。
- 在 PBS 中渗透 0.5% Triton X-100 中的组织,其中含有荧光标记的 phalloidin(在分别测试 TR-或 FITC 标记的 dextran 时与绿色或红色结合),在 1:200 稀释 30 分钟,以标记 F-actin 并可视化基于功能的立体性。
- 用HBSS缓冲液每次清洗组织2~3次2分钟,去除三吨和噬菌体素的过量,用PBS去除盐。
- 将 Corti 组织的器官安装在显微镜幻灯片上,并用安装介质覆盖它。
注:安装组织时,确保包含发细胞立体膜的组织侧朝向盖玻片。 - 清除在添加安装介质过程中产生的任何潜在气泡,并防止在盖玻片放置过程中产生新的气泡。
注:在添加安装介质期间产生任何气泡,用移液器吸气。为防止气泡卡在盖玻片下,请将盖玻片的边缘靠近样品,并使用钳子或移液器尖端小心并缓慢地将盖玻片降至组织上。 - 用玻璃盖玻片盖住安装介质中的组织 (图 2H)。
注: 数值孔径高于 0.4 的孔径设计用于使用 #1.5 盖玻片(0.17 mm 厚度)。使用错误的盖玻片可能会对图像的强度和质量产生严重影响。 - 在室温下孵育安装组织过夜,让安装介质干燥,并将幻灯片储存在 4°C,直到成像。
5. 图像采集和图像处理
注:共聚焦图像采用LSM 880共聚焦显微镜,在超分辨率(SR)模式43和63倍物镜中配备32通道Airyscan探测器。
- 在目标上加入一小滴浸入油。
- 将装有安装的组织样品的显微镜滑块放在显微镜阶段,玻璃盖玻片面向浸入油。
- 关注示例并使用图像采集软件设置成像参数。
- 为每个独立实验使用相同的图像采集设置和最佳参数,用于 x、y 和 z 分辨率。在获取 z 堆栈图像时,需要压电驱动对焦系统快速、精确地移动目标。
注: 要对毛细胞的整个杏子区域进行成像,请使用最佳设置从立体细胞体到头发细胞体的半张图像收集 z 堆栈图像。沿毛细胞收集一个大的 z 堆栈以确保囊泡状颗粒的成像至关重要。 - 使用图像采集软件使用 Airyscan 3D 重建算法使用自动默认反卷积滤波器设置来处理原始共聚焦图像。
- 在图像处理软件中打开共聚焦图像以调整亮度和对比度,添加比例尺,并导出最终图形的图像。

Representative Results
我们观察到,在野生型产后6(P6)小鼠的Corti外体器官孵育2小时后,毛细胞的强健和特殊标记,3 kDa dextran荧光标记有德州红(dextran-TR)(图2A-B)。在Corti器官的基底、中、顶区域(图2B)的内侧和外发细胞(IHC和OHC)中观察到了Dextran标记。
荧光标记的phalloidin用于对抗丝状物(F-actin)和可视化基于功能素的头发细胞立体。我们还在较短的孵育时间进行了类似的实验,虽然我们观察到了头发细胞体内的dextran-TR积累,但信号比2小时孵育时的信号更弱,变化更大(图2C)。
接下来,我们影像了毛细胞的立体细胞和细胞体,并观察到,只有那些在其细胞体内加入3 kDa dextran-TR的细胞显示出其立体细胞的荧光标记(图3A)。在受损组织毛细胞中不存在立体细胞和细胞体标记之间的这种关系,在感觉上皮的几个细胞类型中也呈现了dextran的未特异性标记(图2B和图3B中的黄色方块)。重要的是,我们观察到沿立体音节的均匀荧光信号,在MET通道位于18、23的较短立体音节行的尖端处进行富集(图3C)。此外,我们注意到毛细胞和邻近支持细胞的细胞体中类似囊泡的结构。这些细胞中囊泡状的摄入模式表明,dextran-TR也可以通过内分泌(图3C)来接受。
此处描述的协议还允许检查较大除变性器的采用和不同除变器的组合。标有德州红(dextran-TR)或荧光素(dextran-FITC)的10 kDa的较大dextran,除了在毛细胞和支撑细胞的细胞体中产生囊泡状图案外,还以斑块模式在毛细胞膜周围积累(图4A,B)。10 kDa dextran-TR 也表面上标记了所有毛细胞的三个立体细胞行(图 4A,内插),可能是由于毛细胞血浆膜 44、45、46的负电荷表面。接下来,我们同时检查了 3 kDa dextran-TR 和 10 kDa dextran-FITC 在 Corti 外植物器官中的摄入量。对于 3 kDa dextran-TR,我们观察到了漫射和囊泡状图案。然而,10 kDa dextran-FITC 只显示一个囊泡状图案 (图 4C)。这些数据表明,dextran 的接受至少由两种依赖于 dextran 大小的不同机制发生。
接下来,我们评估是否需要功能 MET 通道来采用 3 kDa dextran-TR。为此,我们测试了在MET通道阻滞剂(新霉素和阿米利德)13、14、47或Ca2+夹层BAPTA的存在下,在Corti外植器官的毛细胞中加入的dextran,通过断开尖端链路和防止通道25、48、49的门控来消除MET电流。在这些实验中,组织外用新霉素(500 mM)、阿米洛里德(150 mM)或BAPTA(5mM)在加入3kDa dextran-TR之前,在相应的MET阻滞剂下孵育了30分钟。BAPTA或MET通道阻滞剂的存在阻止了立体细胞标签(图5A)和毛细胞中3kDa dextran-TR的吸收(图5B)。然而,对MET通道的封锁保留了囊泡状模式(图5B),表明这种接收模式与功能MET通道无关。在TMC1/TMC2双淘汰小鼠的毛细胞中也观察到了类似的结果,这些小鼠缺乏MET27。有趣的是,这些囊泡状结构在阿米洛里德的存在下是不可观察到的,众所周知,它抑制Na+-H+交换器,从而抑制内分泌物变50,51。这些结果表明,3kDa dextran-TR通过两种不同的途径进入毛细胞,一条是涉及囊泡状结构的大型分体体,另一条是依赖于功能MET通道的。

Discussion
该协议描述了如何在3 kDa dextran德州红的Corti外植植物的鼠器官中执行接受实验。这种方法的目的是测试之前测试的分子是否也能够专门标记听觉毛细胞,并渗透到MET通道。类似的实验方案以前曾用于评估毛细胞与其他荧光染料的渗透性,如FM1-43(0.56 kDa)12、19、20和得克萨斯州红标记的氨糖苷(1.29~1.43 kDa)13、52。使用这些荧光分子标记毛细胞所需的时间随大小而变化。最大标签需要几分钟的FM1-43 13,53,30-60分钟为得克萨斯州红标记氨基糖苷13,和1.5小时为得克萨斯州红标签3 kDa dextran。因此,长孵育时间对于这个实验至关重要,因为我们只看到头发细胞体在短孵育时间的最低标记(图2C)。我们选择研究P6小鼠进行这些实验,因为在这个年龄段,大多数的尖毛细胞和基底毛细胞携带转导电流,并表达TMC1和TMC2蛋白质12,18,54,55。应该可以研究年轻的动物,因为MET电流比P6早存在,特别是在耳蜗12的基部附近。研究年长的动物可能更具挑战性,因为解剖骨耳蜗变得更加困难,因为这种结构继续使56。
此协议中有几个关键步骤。最关键的一个是成功解剖和制备Corti组织器官(图1)。科尔蒂器官的微妙结构,包裹在骨性耳蜗中,对组织学和生化分析提出了挑战。成功解剖这种组织的几个详细方案已经发表在本期刊56,57,58和其他59。细胞-细胞结的中断和听觉感觉上皮完整性的丧失可能导致吸收高达40 kDa的dextran和受影响细胞的标签39,40。同意这一点,我们观察到毛细胞和周围支持细胞的未特异性标记时,组织在分部孵育前的解剖过程中被钳子意外损坏(图2B和图3B)。除了练习和耐心之外,没有技巧或准则可以熟练地解剖 Corti 的完整器官。然而,在组织解剖和摄取实验期间存在1⁄2 mM Ca2+对于保持将机械力传输到MET通道的尖端链路的完整性至关重要,从而保持MET通道功能。一旦组织用4%PFA固定,缓冲液或介质中钙的存在就变得微不足道。
这项研究的局限性之一在于dextran分子的异构性。由 Leuconostoc细菌所阐述的 dextran 是一种非氢葡萄糖的聚合物,由大约 95% 的 α-(1⁄6) D-连结组成(图 2A)。其余的联系占dextran的分支,这与它的长度相关,所以低分子量的dextran更像杆状,有一个较窄的分布,而较大的dextrans是多分散60。因此,分子的实际分子量在3kDadextran的每个制剂中范围为1.5至3.0kDa,包括荧光分子61。此外,荧光标记的 dextran 在附着在 dextran(标记程度)上的得克萨斯红色分子的数量上有所不同。建议使用贴标程度至少为0.4的dextran,因为较低的标签程度会妨碍通过显微镜检测荧光标记的dextran的能力。荧光染料耦合到dextran和可固定版本中的莱辛残留物,将决定dextran的总体电荷。这是一个重要的考虑因素,因为MET通道的阳离子选择性最好采用阳离子dextran27。最后,要达到这些实验中显示的分辨率,需要一种可莱辛可修复的dextran。不可修复的dextran将继续扩散到组织中,妨碍其准确的定位和可视化。
LSM 880 共聚焦显微镜配备了 Airyscan 探测器,用于成像 Corti 样品的器官,与传统的共聚焦显微镜62相比,它为我们提供了两倍的分辨率和更好的信噪比 (SNR)。因此,这种显微镜不仅使我们能够观察细胞体内的强健的dextran积累,而且还可以精确定位立体细胞尖端的dextran的积累和细胞体体类似囊泡的图案。传统的共聚焦显微镜可以可视化和量化细胞体荧光dextran的积累,但很难区分三行立体纤和在较短的立体细胞行的尖端的精确定位。使用传统的共聚焦显微镜具有过采样和去卷积,可以使用与 LSM 880 Airyscan 共聚焦实现的分辨率相类似,尽管 SNR 比 Airyscan 探测器的分辨率低。
除了使用3 kDa dextran-TR对功能MET通道进行检测外,该协议还可以进一步用于研究毛细胞的内分泌过程,因为所有经测试的分泌体都揭示了一种类似于内分泌的细胞内囊泡状模式。这一进程不是我们工作的重点,还需要进行更多的研究,以充分描述这一现象。
