Her presenterer vi en metode for å visualisere opptaket av 3 kDa Texas Red-merket dextran i auditive hårceller med funksjonelle mechanotransduction kanaler. I tillegg kan dextrans på 3-10 kDa brukes til å studere endocytose i håret og støtte celler i orgelet Corti.
Hårcellen mechanotransduction (MET) kanal spiller en viktig rolle i hørselen. Imidlertid forblir den molekylære identiteten og strukturelle informasjonen til MET ukjent. Elektrofysiologiske studier av hårceller viste at MET-kanalen har en stor ledning og er gjennomtrengelig for relativt store fluorescerende kationiske molekyler, inkludert noen styrylfargestoffer og Texas Red-merket aminoglykosid antibiotika. I denne protokollen beskriver vi en metode for å visualisere og evaluere opptaket av fluorescerende dekstrans i hårceller i organet corti planter som kan brukes til å analyse for funksjonelle MET-kanaler. Vi fant ut at 3 kDa Texas Red-merket dextran spesifikt merker funksjonelle auditive hårceller etter 1-2 h inkubasjon. Spesielt 3 kDa dextran etiketter de to kortere stereocilia rader og akkumuleres i cellekroppen i et diffust mønster når funksjonelle MET kanaler er til stede. Et ekstra vesicle-lignende mønster for merking ble observert i cellekroppen av hårceller og omkringliggende støtteceller. Våre data tyder på at 3 kDa Texas-Red dextran kan brukes til å visualisere og studere to veier for cellulær hellopptak; en hårcellespesifikk inngangsrute gjennom funksjonelle MET-kanaler og endocytose, et mønster som også er tilgjengelig for større dextran.
Hårcellene i det indre øret er sensoriske celler som oppdager lyd og skjulte de mekanisk stimuli i elektriske signaler, som til slutt tolkes av hjernen vår. Disse cellene har en trappformet bunt av tre rader med actin-baserte filamenter, kjent som stereocilia, som stikker ut fra deres apikale region1,2. De mekaniske stimuli avlede stereocilia filamenter mot den lengste raden og utløse åpningen av mechanotransduction (MET) kanaler3. Åpningen av MET-kanalene fører til en tilstrømning av kationer som depolariserer cellen og signaliserer dermed frigjøring av synapsevesikler i hårcellens basalområde.
De biofysiske egenskapene til MET-kanalen som er avgjørende for hørselen, har blitt mye preget. Blant andre egenskaper er disse kanalene kationiske selektive og har en relativt stor ledning (150–300 pS i lav Ca2 +)4,5,6,7,8,9,10. Bemerkelsesverdig, store fluorescerende molekyler som FM1-43 og Texas Red-merket aminoglykosider er permente blokkere av MET-kanalen, noe som resulterer i akkumulering i håret cellekroppen som kan visualiseres ved hjelp av fluorescens mikroskopi11,12,13,14. Omvendt har den molekylære identiteten og strukturen til MET-kanalen og dens permeasjonsvei vært unnvikende. Økende eksperimentelle bevis indikerer at transmembrane-lignende kanal protein 1 (TMC1) er en del av MET-kanalen i modne hårceller15,16,17,18,19. Mutasjoner i transmembrane-lignende kanal 1 (TMC1) endre MET kanal egenskaper19,20,21,22 og forårsake døvhet. I tillegg lokaliserer TMC1 til stedet for MET-kanalen18,23 og samhandler med tips-linken som er ansvarlig for å overføre den mekaniske kraften til MET-kanalen24,25. Videre har nyere bioinformatikkanalyse identifisert TMC-proteinene som evolusjonære relatert til mechanosensitive kanaler TMEM63/OSCA proteiner og TMEM16 proteiner, en familie av kalsiumaktiverte klorid kanaler og lipid scramblases26,27,28. En strukturell modell av TMC1 basert på forholdet mellom disse proteinene avslørte tilstedeværelsen av et stort hulrom ved protein-lipid-grensesnittet27. Dette hulrommet har de to TMC1 mutasjoner som forårsaker autosomal dominerende hørselstap (DFNA36)27,29,30,31,32, og selektiv modifisering av cysteinmutanter for rester i hulrommet endre MET kanalegenskaper28, noe som indikerer at det kan fungere som permeation banen til MET-kanalen. Den store størrelsen på dette spådde hulrommet i TMC-proteiner kan forklare evnen til store molekyler til å gjennomsyre MET-kanalen. For å teste spådommen om at MET-kanalen inneholder en uvanlig stor permeasjonsvei og for å presse grensene for størrelsen på hulrommet observert i TMC1, utviklet vi en protokoll for å utføre opptakseksperimenter i orgelet corti explants med et større molekyl, 3 kDa dextran fluorescerende merket med Texas Red.
Dextran er et komplekst forgrenet polysakkarid bestående av mange D-glukosemolekyler bundet av alfa-1,6 glykosidiske koblinger. Denhøye løseligheten i vann, lav celletoksisitet og bioinertitet gjør det til et allsidig verktøy for å studere flere cellulære prosesser. I tillegg er dextran tilgjengelig i et bredt spekter av størrelser og fluorescerende merket med fluoropforer av flere farger. Fluorescerende merket dextrans brukes vanligvis i celle- og vevpermeabilitetsforskning33,34, for å studere endocytose i flere cellulære systemer35,36, og for neural sporing37,38. I hørselsfeltet har dextran molekyler også blitt brukt til å vurdere forstyrrelsen av cellecellekrysset og tap av auditiv sensorisk epitelintegritet etter eksponering for intens støy i chinchillaorganet Corti39,40.
I dette arbeidet utnyttet vi egenskapene til noen av de minste (3 og 10 kDa) fluorescerende dextrans for å utføre opptakseksperimenter i murine indre øre hårceller og utforske størrelsen på permeasjonsbanen til den indre ørehårcellen MET kanal. I tillegg brukte vi et laserskannings konfokalmikroskop (LSM) 880 utstyrt med en Airyscan-detektor for å visualisere og lokalisere fluorescerende dextran ved stereocilia og cellekroppen av auditive hårceller.
Denne protokollen beskriver hvordan man utfører opptakseksperimenter i murine organ av Corti explants med 3 kDa dextran Texas Red. Målet med denne metoden er å teste om molekyler større enn andre tidligere testet også var i stand til å spesifikt merke auditive hårceller og gjennomsyre gjennom MET-kanalen.
